Mapping the distributions of blood-sucking mites and mite-borne agents in China:a modeling study

Background:Emerging mite-borne pathogens and associated disease burdens in recent decades are raising serious public health concerns,yet their distributions and ecology remain under-investigated.We aim to describe the geographical distributions of blood-sucking mites and mite-borne agents and to assess their ecological niches in China.Methods:We mapped 549 species of blood-sucking mites belonging to 100 genera at the county level and eight mite-associated agents detected from 36 species of blood-sucking mites in China during 1978–2020.Impacts of climatic and environmental factors on the ecology of 21 predominant vector mites and a leading pathogen,Orientia tsutsugamushi,were assessed using boosted regression tree(BRT)models,and model-predicted risks were mapped.We also estimated the model-predicted number,area and population size of afected counties for each of the 21 mite species in China.Results:Laelaps echidninus is the leading mite species that potentially afects 744 million people,followed by La.jettmari(517 million)and Eulaelaps stabularis(452 million).Leptotrombidium scutellare is the mite species harboring the highest variety of mite-borne agents including four Rickettsia species and two viruses,followed by Eu.stabularis(2 agents),L.palpale(2)and La.echidninus(2).The top two agents that parasitize the largest number of mite species are O.tsutsugamushi(28 species)and hantavirus(8).Mammalian richness,annual mean temperature and precipitation of the driest quarter jointly determine the ecology of the mites,forming four clusters of major mite species with distinct geographic distributions.High-risk areas of O.tsutsugamushi are mainly distributed in southern and eastern coastal provinces where 71.5 million people live.Conclusions:Ecological niches of major mite species and mite-borne pathogens are much more extensive than what have been observed,necessitating expansion of current fled surveillance.

Miniature inverted-repeat transposable elements （mites） are widespread in both prokaryotic and eukaryotic genomes, where their copy numbers can attain several thousands. Little is known, however, about the genetic factor（s） affecting their transpositions. Here, we show that disruption of a gene encoding ubiquitin-like protein markedly enhances the transposition activity of a mite roPing in intact rice plants without any exogenous stresses. We found that the transposition activity of roPing is far higher in the lines harboring a non-functional allele at the Rurml （Rice ubiquitin-related modifier-I） locus than in the wild-type line. Although the alteration of cytosine methylation pattern triggers the activation of transposable elements under exogenous stress conditions, the methylation degrees in the whole genome, the roPing-body region, and the roPing-flanking regions of the non-functional Rurml line were unchanged. This study provides experimental evidence for one of the models of genome shock theory that genetic accidents within cells enhance the transposition activities of transposable elements.

Differential levels of mite infestation of wheat and barley in Czech grain stores

While mites are able to utilize numerous food sources, the suitability of the food strongly influences population growth. The different suitabilities of various stored agricultural products will thus affect the level of infestation. In this study, we compared field mite infestation rates in two stored cereals： wheat and barley. We analyzed mite abundance, frequency and species composition in samples of grain obtained from 79 selected Czech grain stores. Stored barley seemed to be more vulnerable to mite attack than wheat, as we consistently found more infested samples, more species and higher mean and median mite abundance per sample in barley as compared to wheat. The mean mite abundance per sam- ple were 55 and 506 individuals for wheat and barley, respectively. In barley, 10% of sampies exceeded allergen risk threshold （i.e., 1 000 individuals per kg of grain）. Altogether, 25 species were identified from approximately 35 000 individuals. The most frequently identified species were the same in wheat and barley, that is, Tydeus interruptus Sig Thor, Acarus siro L., Tarsonemus granarius Lindquist, Lepidoglyphus destructor （Schrank） and Tyrophagus putrescentiae （Schrank）. Based on principal components analysis, we found a closer association of T. interruptus, T. putrescentiae, L. destructor and Cheyletus eruditus （Schrank） with barley samples, corresponding to the high frequency and abundance values of these mites. The probable reasons for the higher infestation, especially mite abundance in barley, are discussed in relation to the higher proportion of crushed parts, which may release favorable nutrient sources and amplify the abundance values.

Plant mites: Useful Tools for Plant Genetics and Genomics

MTTEs (Miniature inverted-repeat transposabie elements) are reminiscence ot non-autonomous DNA (class Ⅱ) elements, which are distinguished from other transposable elements by their small size, short terminal inverted repeats (TIRs), high copy numbers, genie preference, and DNA sequence identity among family members. Although mites were first discovered in plants and still actively reshaping genomes, they have been isolated from a wide range of eukaryotic organisms. mites can be divided into Tourist-like, Stowaway-like, and pogo-like groups, according to similarities of their TIRs and TSDs (target site duplications). In despite of several models to explain the origin and amplification of mites, their mechanisms of transposition and accumulation in eukaryotic genomes remain poorly understood owing to insufficient experimental data. The unique properties of mites have been exploited as useful genetic tools for plant genome analysis. Utilization of mites as effective and informative genomic markers and potential application of mites in plants systematic, phylogenetic, and genetic studies are discussed.

Quantitative impact of mating duration on reproduction and offspring sex ratio of Phytoseiulus persimilis(Acari: Phytoseiidae)

Phytoseiulus persimilis is an important biological control agent, commercially produced worldwide. To increase understandings of its reproduction, herein we provided quantitative descriptions of the inflation and deflation of its spermathecae as a function of time and copulation duration, and the quantitative impact of copulation termination on volume of spermatheca vesicle, fecundity, and offspring sex ratio. After mating started, at least one spermatheca started to inflate immediately, but no egg was produced until spermatheca volume reached 1 021 μm3. Beyond this size, cumulative fecundity was linearly correlated with vesicle volume. Producing one egg required 36 μm3 volume increase of the vesicle. Each spermatheca vesicle reached its peak size at the end of mating duration(ca. 2.38 h after mating started), and started to shrink immediately. In 24 h, ca. 71% individuals had one completely shriveled spermatheca, while in 72 h all individuals had at least one shriveled spermatheca. The estimated maximum cumulative fecundity per female after a single mating is(69.4±7.7) eggs. No significant impact of mating termination on offspring sex ratio was observed after mating duration reached 60 min, while higher proportion of male offspring was observed when mating duration is 15 or 30 min. This study is the first step to investigate possible male impact on offspring sex ratio in P. persimilis. Our results suggested the number of sperms received during mating be a restriction factor of P. persimilis offspring sex ratio regulation.

Decreasing detection frequency of mite(MCLas-A)in the population of‘Candidatus Liberibacter asiaticus'recently collected in southern China

An active miniature inverted-repeat transposable element(mite),MCL as-A,was previously identified from'Candidatus Liberibacter asiaticus'known to be associated with citrus Huanglongbing(HL B,yellow shoot disease).To explore the recent transposition status of MCLas-A,389'Ca.L.asiaticus'strains collected from nine regions in China were amplifed using a specific primer set and three representative'Ca.L.asiaticus'strains were analyzed by next-generation sequencing(NGS)approach.PCR and genomic analysis showed that the entire MCLas A was only present in 1.80%(7/389)and the jumping-out type of the mite was predominant(8123%)in samples tested,suggesting high frequency transposition occurred in'Ca.L.asiaticus'strains recently collected from China.Biological roles of transposition of the active mite remain to be determined.

A mite insertion into the 3′-UTR regulates the transcription of TaHSP16.9 in common wheat

Miniature inverted-repeat transposable elements(mites) are a type of DNA transposon frequently inserted into promoters, untranslated regions(UTR), introns, or coding sequences of genes. We found a 276-bp tourist-like mite insertion in the 3′-UTR of a 16.9 k Da small heat shock protein gene(TaH SP16.9-3A) on chromosome 3A of common wheat. Haplotype analysis revealed two haplotypes, s HSP-W(wild type without mite insertion) and s HSP-M(mutant with mite insertion), present in wheat germplasm. Both semiquantitative PCR and quantitative real-time PCR analyses showed increased transcription levels of TaH SP16.9-3A in s HSP-M compared with those of s HSP-W after heat treatment at 42 °C. It appeared that the mite insertion into the 3′-UTR enhances the transcription of TaH SP16.9-3A.

Effect of a commercial air ionizer on dust mites Dermatophagoides pteronyssinus and Dermatophagoides farinae(Acari:Pyroglyphidae) in the laboratory

Objective:To investigate the short and long tenn efficacy of a commercial air ionizer in killing Dermatophagoides pteronyssinus(D.pteronyssinus)and Dermalophagoides farinae(D.farinae)mites.Methods:The effect of a commercial ionizer on D.pteronyssinus and D.farinae was evaluated in the laboratory,using a specially designed test.Mortality was assessed after 6,16and 24 hours for direct exposure and after 24,36,48,60 and 72 hours for exposure in simulated mattress.New batches of mites were used for each exposure time.Results:LT_(50)for direct exposure of ionizer was 10 hours for D.pteronyssinus and 18 hours for D.farinae.The LT_(50)for exposure in simulated mattress was 132 hours or 5.5 days for D.pteronyssinus and 72 hours or 3days for D.farinae.LT_(95)for direct exposure of ionizer was 36 hours for D.pteronyssinus and D.farinae.Meanwhile,the LT_(95)for exposure in simulated mattress was 956 hours or 39.8 days for D.pteronyssinus and 403 hours or 16.8 days for D.farinae.Conclusions:This study demonstrates the increasing mite mortalities with increasing exposure time of a commercial ionizer and suggests that negative ions produced by an ionizer kill dust mites and can be used to reduce natural mile populations on exposed surfaces such as floors,clothes,curtains,etc.However,there is reduced efficacy on mites inside stuffed materials as in mattresses and furniture.

Acarine ectoparasites of Panti Forest Reserve in Johore,Malaysia

Objective:To identify the presence of acarine ectoparasites and determine whether there is any potential public health risk in Panti Forest Reserve,Johore,Malaysia.Methods:Trapping of animals and avifauna was conducted simultaneously along 5 expedition trails using 150 wire traps,10 harp traps and 30 mist nets for 6 consecutive nights.A total of 140 animals consisting of 7 species of birds,19 species of bats,6 species of rodents and 1 species of tree-shrew as well as 8 myriapods were examined.Results:Infestation rates of ticks,mesostigmatid mites and chiggers on animals examined were 24.3%,28.6%and 27.9%,respectively.Infestation on bats was low(1.5%) and none occurred on birds.Majority of ticks extracted were at immature stages(78.9%). Genera of ticks on animals were Amblyomma,Dermacentor,Haemaphysalis and Ixodes.Ixodes granulatus was the only species of licks identified from the animals.Examination of ticks under vegetation revealed 54%adults leading to identification of 3 species of ticks.A total of 7 species of mesostigmatid mites were found.6 species were on rodent,Maxomys mrifer and another one species,Laelaps nuttalli was found only on Leopoldamys sabanus.Laelaps sanguisugus was the only mesostigmatid found infesting tree-shrews.Seven genera of chiggers were identified.From this,5 genera were on rodents,4 genera on tree-shrews and 1 genus on a bat.Conclusions:A total of 16 genera,2 sub-genus and 14 species of acarine ectoparasites were found in this area. Findings of the survey demonstrate the presence of three spesies of acarine ectoparasites which have potential health risk i.e.Ixodes granulalus,Laelaps nuttalli and Leptotrombidium deliense.

Sublingual immunotherapy for pediatric allergic rhinitis: The clinical evidence

Allergic rhinitis affect 10%-20% of pediatric population and it is caused by the Ig E-sensitization to environmental allergens, most importantly grass pollens and house dust mites. Allergic rhinitis can influence patient's daily activity severely and may precede the development of asthma, especially if it is not diagnosed and treated correctly. In addition to subcutaneous immunotherapy, sublingual immunotherapy(SLIT) represents the only treatment being potentially able to cure allergic respiratory diseases, by modulating the immune system activity. This review clearly summarizes and analyzes the available randomized, double-blinded, placebo-controlled trials, which aimed at evaluating the effectiveness and the safety of grass pollen and house dust mite SLIT for the specific treatment of pediatric allergic rhinitis. Our analysis demonstrates the good evidence supporting the efficacy of SLIT for allergic rhinitis to grass pollens in children, whereas trials regarding pediatric allergic rhinitis to house dust mites present lower quality, although several studies supported its usefulness.
