Detection of microbial antigenic components of circulating immune complexes in HIV patients:Involvement in CD4^+ T lymphocyte count depletion

Objective:To investigate the prevalence of microbial antigenic components of circulating immune complexes amongst grades of CD4 T lymphocyte counts in HIV sero positive and seronegative participants.Methods:Polyethelene glycol(PEG-600) and buffering methods of precipitation and dissociation of immune complexes was used to generate immune solution from sera of 100 HIV sero-positive and 100 HIV sero-negative participants.These were categorized into 3 grades based on CD4 count:> 500 cell/mm,200-499 cell/mm3 and microbial antigens and or antibodies. CD4 T cell counts were estimated using Patec Cyflow SL-3 Germany.Results:Antigenic component of immune complexes of various infectious agents was detected in 99 and 70 HIV seropositive and HIV sero-negative participants,respectively.In group A,there were 10 HIV positive participants,including 4(40.0%) had circulating immune complexes(CICs) due to Salmonella species only:1(10.0%) due to Salmonella-Plasmodium falciparum(P.falciparum),SalmonellaP. falciparum-HCV and P.falciparum antigens,respectively.In group B,45(45.4%) HIV seropositive participants with CICs had CD4 T lymphocyte count between 200-499 cells/mm^3.Out of these,20(44.4%) had CICs due to Salmonella species only:9(20%) due to Salmonella-P. falciparum.In group C,there were 44(44.4%) HIV sero-positive participants,including 3(6.8%) due to Salmonella species only:24(54.4%) due to Salmonella-P.falciparum:2(4.5%) due to P. falciparum only.Conclusions:In HIV sero-positive participants,presence of heterogeneity of Salmonella species-P.falciparum antigens was highly incriminated in CD4 count depletion but not homogeneity of malaria parasites antigens.Malaria parasites antigens only were incriminated in CD4^+ count depletion amongst HIV sero-negative participants.Before taking any decision on the management of HIV-1-positive individuals,their malaria and

Identification of microbial contaminants in sinus rinse squeeze bottles used by allergic rhinitis patients

Objective:To identify whether irrigation devices become contaminated when used by patients with allergic rhinitis (AR).Methods:Ten AR patients with no clinical or endoscopic evidence of active sinonasal infection were given a sinus rinse system and instructed on its proper use,cleaning,and storage.Two squeeze bottles (bottle A and bottle B) were given to each patient for twice-a-day rinsing.Bottle A was used in the morning and analyzed after four weeks.Bottle B was used in the evening and analyzed after 8 weeks of use.microbial contaminants were cultured from the nose pieces and the inner surface of the bottles obtained from patients.Results:Seventeen sinus rinse devices (17/20) from all individuals in this study grew bacteria commonly in the nozzles.Twenty-four bacterial isolates consisting of 14 different species were cultured and identified with most common organisms being bacilli and staphylococcus.In addition,no correlation was apparent between the length of bottle use and the degree of contamination (r =0.13,p =0.76).During the study period,no patient developed acute sinus infections.Conclusion:microbial contamination of the sinus rinse system occurs commonly,even in uninfected AR patients;however no evidence exists linking this to clinically relevant sinus infections.

Investigations of microbial origin of karst corrosion of soils depending on different temperatures

The acids accumulating in soils and controlling the solution of carbonates including the predominant CO2, mostly derive from three processes: i) root respiration of higher plants; ii) decomposition of soil organic matter by microorganisms (microbiota) and iii) other decomposition processes not associated with microbial activities. The solution effect under rendzina soils is primarily used for the dissolution of the enclosed limestone fragments and thus here the solution of bedrock is of limited scale. Below karst soils of high clay content the corrosion of bedrock is more intensive than under rendzinas. On the whole, the amount of carbonates dissolved and transported into the depths of the karst is smaller than below rendzinas. In each soil type studied the solution caused by microbial activities manifold exceeds the rate of solution resulting from temperature factor but there is a manifest dropping trend from rendzina to clays.

Improving tuberculosis infection control practice and microbial air quality in a general hospital after intervention

Objective:Hospital personnel,especially nurses are at risk for tuberculosis(TB) infection and the intervention for reducing the risk should be established.Methods:To compare the TB infection control practices and standard precaution in 154 registered nurses and auxiliary nurses working in risk wards.Additionally,microbial air quality in the studied wards was investigated before and after implementation of an intervention including twoday training program on TB infection control and standard precaution practices and managing the ward environments. Results:Post-intervention,the percentage of studied nurses who always practiced increased in every item of TB infection control practice(6 items) and standard precaution(9 items) when compared with the preintervention period.Both mean scores were significantly higher than those before intervention(5.0±1.0 vs 4. 4±1.1,and 7.6±1.3 vs 6.7±1.5,P500 cfu/m^3).Conclusion:Two-day training program and management of the ward environment could improve the scores of TB infection control practices and the standard precaution among studied nurses and reduce the microbial counts in air samples collected from the studied wards.

microbial population in rotten living body of Salix matsudana

The microbial population in rotten living body of Salix matsudana caused by Trametes suaveolens (L.) was researched. 11 bacteria species (1 species of Bacillus, 2 species of Clostridium and 8 species of non-brood-cell bacteria), 1 species of Actinomyces that belongs to Lavendulac, 8 species of fungi and 6 species of Trichoderma were isolated from rotten trunk. The hyphae of Trametes suaveolens mainly existed between rotten sections and discoioration sections. In over-rotten section and healthy section the fungi (Trametes suaveolens) were not isolated. The microbes that lived in the discoloration section were the most in kinds and number and they were the pioneer microbes of wood rotting. Only after they dwelled in wood and eIiminated its rot-resistance,could wood-rotting fungi invade wood and caused wood-rotting.

Role of preputial washing in reducing microbial load and improving bovine semen quality

Quality semen production remains the main focus and objective of semen processing laboratories throughout the world. Bacterial and other microbial contaminants affect the semen quality and hence the fertility, and also lead to reproductive disorders as well as lower conception rates and increased embryonic mortality, abortion and other complications in females. microbial contamination affects the semen adversely, by exerting direct spermicidal effect, formation of reactive oxygen species, toxin production, adherence with spermatozoa, deriving nutrients and oxygen from the medium and thus competing with spermatozoa for the factors of growth and normal functioning. Despite hygienic measures, several ubiquitous and opportunistic microbes find their ways into semen during collection, processing, and storage of semen, and survive even during freezing. Stringent sanitary precautions are therefore required at every step of collecting semen and its processing. Preputial cavity is considered as main source of semen contaminating microorganisms. Flushing the preputial cavity with normal saline or any suitable liquid combination with antimicrobial activity, prior to semen collection reduces the microbial load and thereby improves the semen quality.

VERY little is known about the roles of microorganisms in the metallogeny of weathering crust REE deposits, although there has been much literature on REE behavior in weathering conditions. The present note deals with the microorganisms in 4 weathering profiles of an REE deposit and their roles in migration and accumulation of REE in weathering conditions. 1 Cultivation and identification of microorganisms Under strict sterilization, samples were collected from various parts of 4 weathering profiles of granites in a weathering crust REE deposit in South China. The samples were then cultivated for microorganisms with ordinary agars as culture media and then the cultivated microorganisms were dyed and identifiedunder a microscope. Zinc carnosine-based modified bismuth quadruple therapy vs standard triple therapy for Helicobacter pylori eradication: A randomized controlled study

BACKGROUND Helicobacter pylori(H.pylori)infection is a worldwide problem with increasing burden on the health sector due to its increasing rate of resistance.The conventional triple therapy(TT)is becoming obsolete with a high failure rate of eradication,necessitating the need for better alternatives or regimens.AIM To investigate H.pylori eradication rate of TT vs modified bismuth quadruple therapy.METHODS Ninety-two patients with dyspepsia symptoms and positive ^(13)C-urea breath test were randomly assigned to two groups.The first group(control group)was treated for 14 d using standard TT protocol:Esomeprazole(40 mg twice daily),amoxicillin(1 g twice daily)and clarithromycin(500 mg twice daily).On the other hand,the second group was prescribed a 10-d course of modified bismuth quadruple therapy fortified with zinc carnosine:TT in addition to bismuth subcitrate(240 mg twice daily)and zinc carnosine(75 mg twice daily).A repeated 13C-urea breath test was done 4 wk after the completion of the eradication therapy.RESULTS Among the 92 subjects,67.4%were males and 32.6%were females.There were no differences in demographic characteristics(age,body mass index,smoking history,previous antibiotics use and ethnicity)between the modified bismuth quadruple therapy group and TT group.The eradication rate was higher[93.5%(43/46)]in the modified bismuth quadruple therapy group compared to 69.6%(32/46)in the standard TT group(P=0.003).Of the tested predictor variables,only nationality,smoking and therapy type were statistically significant.Besides dizziness,which was recorded in modified bismuth quadruple therapy group,there were no significant differences in side effects between the two groups.CONCLUSION Ten days of modified bismuth quadruple therapy fortified with zinc carnosine is superior to 14 d of conventional TT in eradicating H.pylori infection,with no additional significant adverse events.

A method for microbial decontamination of Acanthamoeba cultures using the peritoneal cavity of mice

Objective: To evaluate whether the inoculation of contaminated cultures in the peritoneal cavity of mice could implement decontamination of Acanthamoeba cultures.Methods: Suspensions of Acanthamoeba, Acanthamoeba polyphaga ATCC 30461, or Acanthamoeba spp. isolated from soil(Un B13 strain) were inoculated in the peritoneal cavity of Swiss mice(n = 24). After 1, 6, 12, or 24 h of exposure the peritoneal cavity was washed and assessed for the presence of bacteria, fungi, and Acanthamoeba.Results: After 1 h of intraperitoneal inoculation at least 97% of the bacteria and 96% of the fungi(P < 0.05) and 99% of the bacteria(P < 0.05) were successfully eliminated from the ATCC 30461 strain and from the soil isolate Un B13 strain, respectively. This method also allowed the recovery of most trophozoites and cysts from both Acanthamoeba cultures at the end of 24 h.Conclusions: Our data demonstrated that this technique has great potential for decontamination of Acanthamoeba cultures in a short period of time.
