Inhibition of the CDK9-cyclin T1 protein-protein interaction as a new approach against triple-negative breast cancer

In coordination and supramolecular chemistry, there are many symmetrical imidazole and benzotriazole ligands, which have been widely used as the classical ligands. However, studies on unsymmetricalligands concerning imidazole and benzotriazole are rather insufficient. 1-[(2-ethyl-1H-imidazol-1-yl)methyl]-1HBenzotriazole(bmei) as a unsymmetrical ligand has been used in the construction of metal complexes due to strong coordination ability of the N-donor. In this work, through the reaction of 1-[(2-ethyl-1H-imidazol-1-yl)methyl]-1H-Benzotriazole(bmei) with metal salt Cu(Ac) and KSCN at room temperature, we obtained the title complex {[Cu(bmei)(SCN)(CHOH)]·CHOH}n, which is reported here. The complex exhibits onedimensional chain, each Cu(Ⅱ) ion is five-coordinated with two oxygen atoms from two CHOH molecules,two nitrogen donors from two ligands bmeitne nitrogen donor from SCN-. In addition, we research the vitro hypoglycemic activity of the complex. The hypoglycemicactivity in vitro tests show that the copper complex has a good hypoglycemic effect on alpha-amylase.

Cyclin-dependent kinase 9(CDK9) activity is correlated with worse outcomes of triplenegative breast cancer(TNBC) patients. The heterodimer between CDK9 with cyclin T1 is essential for maintaining the active state of the kinase and targeting this protein-protein interaction(PPI) may offer promising avenues for selective CDK9 inhibition. Herein, we designed and generated a library of metal complexes bearing the 7-chloro-2-phenylquinoline CN ligand and tested their activity against the CDK9cyclin T1 PPI. complex 1 bound to CDK9 via an enthalpically-driven binding mode, leading to disruption of the CDK9-cyclin T1 interaction in vitro and in cellulo. Importantly, complex 1 showed promising anti-metastatic activity against TNBC allografts in mice and was comparably active compared to cisplatin. To our knowledge, 1 is the first CDK9-cyclin T1 PPI inhibitor with anti-metastatic activity against TNBC. complex 1 could serve as a new platform for the future design of more efficacious kinase inhibitors against cancer, including TNBC.

The development of transition metal complex-based luminescent chemosensors has recently aroused increasing interest for protein biomarker labelling and detection,especially for the real-time diagnosis and treatment of *** is owing to their unique photophysical properties,particularly their long-lived and environmentally sensitive emission,which can be easily controlled via the structural modification of *** this overview,we highlight recent examples of protein biomarker detection using group 8–9 metal-based luminescent chemosensors,including the frequently employed ruthenium(II)and iridium(III)*** mechanisms and sensing modes are described and compared,and the outlook and future directions of this field are discussed as well.

Photocatalytic water oxidation based on semiconductors usually suffers from poor charge transfer from the bulk to the interface,which is necessary for oxygen ***,we construct a hybrid artificial photosynthesis system for photocatalytic water *** system consists of BiVO4as the light harvester,a transitional metal complex(M(dca)2,M=Co,Ni,dca:dicyanamide)as the water oxidation catalyst,and S2O82?as a sacrificial electron *** system exhibits enhanced oxygen evolution activity when M(dca)2is *** BiVO4/Co(dca)2and Bi‐VO4/Ni(dca)2systems exhibit excellent oxygen evolution rates of508.1and297.7μmol/(h·g)compared to the pure BiVO4which shows a photocatalytic oxygen evolution rate of252.2μmol/(h·g)during6h of photocatalytic ***(dca)2is found to be more effective than Ni(dca)2as a water oxidation *** enhanced photocatalytic performance is ascribed to the M(dca)2‐engineered BiVO4/electrolyte interface energetics,and to the M(dca)2‐catalyzed surface water *** two factors lead to a decrease in the energy barrier for hole transfer from the bulk to the surface of BiVO4,which promotes the water oxidation kinetics.

Determination of Stability Constants of Cadmium-Glycine complexes by a Unified Treatment for Potentiometric and Polarographic Data

The cadmium（Ⅱ）-glycine system was studied by the two experimental techniques, ion sensitive electrode （ISE） and differential pulse polarography （DPP）, and the experimental data obtained were used by a unified mathematical treatment to calculate the complex stability constants. The combination of the two techniques is of many advantages as ISE can be performed at low [LT]：[MT] ratios and significantly higher [MT], whereas DPP could be used well at large [LT]：[MT] ratios and much smaller [MT]. This makes it possible to study a metal-ligand system in a relatively broader range of experimental conditions that, in turn, provides more information about the metakligand system of interest. Applying the unified mathematical treatment to the cadmium-glycine system, two new complexes MHL and ML2（OH） as well as three complexes ML, ML2 and ML3, reported in literatures, could be modeled and all their stability constants have been refined.

A new kind of π-conjugated heterocyclic poly(Schiff base) was firstly prepared by the condensation reaction between tetrathiatetrahydropentalene-type diketone and bithiazole-diamine in good yields. The polymers were characterized by VPO, FTIR and 1H NMR spectroscopy. A large bathochromic shift was observed in UV-Vis spectra for these polymers due to the π-π* transition in the conjugated main chain. Brief examination indicated that the nitrogen- and sulfur-containing polymers exhibited an excellent chelating tendency to metal ions and the corresponding polymeric complexes would be expected to have potential in applications.

Polymer exchange membrane fuel cells （PEMFC） are objects of the current engineering technology and these are versatile generators for electrical energy. There are various kinds from them, but all of them are going on work at highest temperature. There isn＇t a PEMFC which can run at room temperature, like 20 ℃. In this study there is a aim for constructing such one for alternative fuels utilisation. PS and many electroconducting polymer formulations were proved by different researchers for PEM benefications, but here PS was synthesized without containing metalic contaminants and after converted to the PEM membrane.

The synthesis and characterization of N,N＇-bis（salicylidene） ethylenediamine, Salen said that were carried out. The behavior of this ligand has been studied in solution in solvent homogeneous and heterogeneous systems in sulphate medium. The use of pH-metric titration, GRAN and JOB＇s methods have permitted the determination of different parameters governing the behavior of Salen in solution such as the acidity constant, the stability constant, the nature of the interaction solvent-solute, the solvation number, the stoichiometry of extracted complex etc.. Then the extraction of the Salen was finally undertaken to words the copper （II） sulfate medium. The pH of complexation, extraction time, the number of protons exchanged and the number of ligands have been finally determined.

In this paper,O-vanilIin-2-furanthiocarboxyhydrazone(HL)and its copper(Ⅱ),nickel(Ⅱ)and cobalt(Ⅱ)complexes have been synthe-sized and characterized by elemental analysis,electronic and infrared *** results suggest an octahedral geometry for(HL)Cl·2HO and Co(HL)Cl,and a square geometry for Cu(HL)*** biological studies show thatthese compounds all exist antibacterial *** activity order is copper(Ⅱ)complex>nickel(Ⅱ)complex>HL>cobaIt(Ⅱ)complex.
