A metabolomics approach for monitoring thermal processing temperature of bovine milk using ultra-performance liquid chromatography tandem Q-Exactive mass spectrometry and multivariate data analysis

Due to the lack of effective assessment method,overheated milk commodities are often marketed as pasteurized milk on the market,which was sold in high price by fraud.Thus,this article aims to establish an approach based on metabolomics to monitor thermal processing temperature of bovine milk.metabolomics data of bovine milk samples heated at temperatures ranging from 60℃to 150℃were achieved by ultra-performance liquid chromatography-high-resolution mass spectrometry(UPLC-HRMS)platform,followed by multivariate data analysis.A regular variation pattern of chemical composition as temperature rises was pictured,furthermore.N^(∈)carboxymethyl lysine(CML),N^(∈)-carboxyethyl lysine(CEL),pentosidine.pvrraline and lysinoalanine(LAL)were identified as 5 of the most contributed compounds to discriminate pasteurized and ultra-high-temperature(UHT)milk.By the comprehensive study on their content changes,we concluded that the optimal temperature range was 90-100℃for the generation of CML and CEL in this experiment,moreover.110~120℃for LAL.S0~100℃for pentosidine and 130-140℃for pvrraline.Finally,a predicted rule to discriminate pasteurized and UHT milk was preliminarily established based on the ratios of CML/CEL.CEL/pentosidine and CML/pentosidine.which could be applied in food labelling authentication of commercial bovine milk after further validation.

Sophisticated analytical instruments are used for quality evaluation and scientific validation to compliance the Sophisticated analytical instruments are used for quality evaluation and scientific validation to compliance the market demands.Recently,increase in demand of Curcuma longa has led to a high swing in small and medium food industry.The present review is devoted to an in-depth information on metabolite database of C.longa from different analytical techniques such as high-performance thin-layer chromatography(HPTLC),liquid chromatography-mass spectrometry,gas chromatography-mass spectrometry and nuclear magnetic resonance.This work highlights different extraction methods,phytochemistry along with structure activity relationship,nutritional profiling,market viability and pharmacology of C.longa.The bioactives of C.longa depends on various factors such as plants growth,temperature,humidity,soil type,collection,drying and extraction process.These factors can contribute in quality evaluation of the food supplements if need to be included.The introduction of omics approaches can helpful to explore the quality and identification of potential lead of C.longa.The results could play an essential role in the area of food-phyto based profiling.Further,metabolomics techniques may provide a analytical platfom by combination of different"omics"data souces for future esearch to food sand herbal based products.

The objective of this study was to determine the differences of aroma and taste in three black sesame originsbefore and after processing via flavor and widely metabolomics.By analyzing the sensory characteristics and metabolites of raw and treated black sesame from China,Vietnam,and Myanmar,treated Chinese sesame have the most significant change in hardness after thermal processing,low viscosity and was easy to chew.The electronic nose could distinguish between raw and treated sesame due to the aroma distribution.The reason of treated sesame from China was“fragrant”is due to the highest content(2545.50μg/kg)of total pyrazines including 2,5-dimethylpyrazine,2-ethyl-5-methylpyrazine,2,3,5-trimethylpyrazine,3-ethyl-2,5-dimethylpyrazine.933 metabolites were detected via a wide targeted metabolomics in the taste of raw and treated sesame.Based on the analysis of metabolites related to bitterness,145 substances were selected.The main bitter contributors may be amino acids,dipeptides and organic acids.

Background:Cooked rhubarb(CR)is obtained by steaming raw rhubarb(Rheum palmatum L.,Rheum officinale Baill.,or Rheum tanguticum Maxim.ex Balf.)with millet wine.It is used as a traditional Chinese medicine for treating cerebral stroke,where patients often face severe constipation.This study explored the pharmacological effects and mechanisms of CR against ischemic stroke(IS)in rats.We used integrated analysis of gut microbiota,metabolomics,and network pharmacology.Methods:The compounds in CR were identified using LC-MS/MS.The impact of CR on rat middle cerebral artery occlusion/reperfusion(MCAO/R)-induced cerebral infarct size and brain tissue pathology was assessed through 2,3,5-triphenyl tetrazolium chloride and HE staining.Changes in hemorheology were measured by evaluating blood viscosity,and serum levels of pro-inflammatory cytokine were also determined.Gut microbiota composition was analyzed through 16S rRNA gene sequencing.Serum metabolites were examined using untargeted metabolomics and Spearman correlation analysis.The pseudo-germ-free test was used to determine whether tumour necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)improvement in IS are dependent on gut microbiota.Results:In vivo studies showed CR enhances neurobehavioral function in MCAO/R rats and reduces cerebral infarction area.CR also ameliorates brain and intestinal barrier damage caused by stroke.It also decreased inflammation and oxidative stress in IS rats.Furthermore,CR promotes the growth of Akkermansia and Verrucomicrobia,aiding in intestinal barrier repair.Notably,CR primarily influences the primary bile acid biosynthesis pathway.The pseudo-germ-free experiment and network pharmacology confirmed TNF-αand IL-1βas potential targets.CR relies on gut microbiota for its anti-inflammatory effects to improve IS.Conclusion:Cooked rhubarb offers neuroprotective benefits by enhancing beneficial bacteria abundance and regulating bile acid metabolism.It emerges as a potential therapeutic agent for IS.

The environmental behavior of and risks associated with nanoplastics(NPs)have attracted considerable attention.However,compared to pristine NPs,environmental factors such as ultraviolet(UV)irradiation that lead to changes in the toxicity of NPs have rarely been studied.We evaluated the changes in morphology and physicochemical properties of polystyrene(PS)NPs before and after UV irradiation,and compared their hepatotoxicity in mice.The results showed that UV irradiation caused particle size reduction and increased the carbonyl index(CI)and negative charge on the particle surface.UV-aged PS NPs(aPS NPs)could induce the generation of hydroxyl radicals(·OH),but also further promoted the generation of·OH in the Fenton reaction system.Hepatic pathological damage was more severe in mice exposed to aPS NPs,accompanied by a large number of vacuoles and hepatocyte balloon-like changes and more marked perturbations in blood glucose and serum lipoprotein,alanine aminotransferase and aspartate aminotransferase levels.In addition,exposure to PS NPs and aPS NPs,especially aPS NPs,triggered oxidative stress and significantly damaged the antioxidant capacity of mice liver.Compared with PS NPs,exposure to aPS NPs increased the number of altered metabolites in hepatic and corresponding metabolic pathways,especially glutathione metabolism.Our research suggests that UV irradiation can disrupt the redox balance in organisms by promoting the production of·OH,enhancing PS NPs-induced liver damage and metabolic disorders.This study will help us understand the health risks of NPs and to avoid underestimation of the risks of NPs in nature.

Bioremediation became a promising technology to resolve arsenic(As)contamination in aquatic environment.Since monoculture such as microalgae or bacteria was sensitive to environmental disturbance and vulnerable to contamination,green microalgae Chlorella vulgaris and arsenite(As(Ⅲ))-oxidizing bacteria Pseudomonas sp.SMS11 were co-cultured to construct algal-bacterial consortia in the current study.The effects of algae-bacteria(A:B)ratio and exposure As(Ⅲ)concentration on algal growth,As speciation and metabolomic profile were investigated.Algal growth arrested when treated with 100 mg/L As(Ⅲ)without the co-cultured bacteria.By contrast,co-cultured with strain SMS11 significantly enhanced As tolerance in C.vulgaris especially with A:B ratio of 1:10.All the As(Ⅲ)in culture media of the consortia were oxidized into As(Ⅴ)on day 7.Methylation of As was observed on day 14.Over 1% and 0.5% of total As were converted into dimethylarsinic acid(DMA)after 21days cultivation when the initial concentrations of As(Ⅲ)were 1 and 10 mg/L,respectively.Metabolomic analysis was further performed to reveal the response of consortia metabolites to external As(Ⅲ).The enriched metabolomic pathways were associated with carbohydrate,amino acid and energy metabolisms.Tricarboxylic acid cycle and glyoxylate and dicarboxylate metabolism were upregulated under As stress due to their biological functions on alleviating oxidative stress and protecting cells.Both carbohydrate and amino acid metabolisms provided precursors and potential substrates for energy production and cell protection under abiotic stress.Alterations of the pathways relevant to carbohydrate or amino acid metabolism were triggered by energy requirement.

Gut microbiota and circulating metabolite dysbiosis predate important pathological changes in glucose metabolic disorders;however,comprehensive studies on impaired glucose tolerance(IGT),a diabetes mellitus(DM)precursor,are lacking.Here,we perform metagenomic sequencing and metabolomics on 47 pairs of individuals with IGT and newly diagnosed DM and 46 controls with normal glucose tolerance(NGT);patients with IGT are followed up after 4 years for progression to DM.Analysis of baseline data reveals significant differences in gut microbiota and serum metabolites among the IGT,DM,and NGT groups.In addition,13 types of gut microbiota and 17 types of circulating metabolites showed significant differences at baseline before IGT progressed to DM,including higher levels of Eggerthella unclassified,Coprobacillus unclassified,Clostridium ramosum,L-valine,L-norleucine,and L-isoleucine,and lower levels of Eubacterium eligens,Bacteroides faecis,Lachnospiraceae bacterium 3_1_46FAA,Alistipes senegalensis,Megaspaera elsdenii,Clostridium perfringens,α-linolenic acid,10E,12Z-octadecadienoic acid,and dodecanoic acid.A random forest model based on differential intestinal microbiota and circulating metabolites can predict the progression from IGT to DM(AUC=0.87).These results suggest that microbiome and metabolome dysbiosis occur in individuals with IGT and have important predictive values and potential for intervention in preventing IGT from progressing to DM.

Herbivory is common in mammals,yet our understanding of detoxification processes used by mammals to biotrans-form plant secondary compounds(PSCs)is limited.Specialist herbivores are thought to have evolved detoxifica-tion mechanisms that rely more heavily on energetically cheap Phase I biotransformation reactions to process high levels of PSCs in their diets.We explored this hypothesis by comparing the urinary metabolite patterns of two spe-cialist herbivores(genus Neotoma).Neotoma stephensi is an obligate specialist on one-seeded juniper(Juniperus monosperma).Neotoma lepida is a generalist forager across its range,yet populations in the Great Basin specialize on Utah juniper(J.osteosperma).While both juniper species have high levels of terpenes,the terpene profiles and quantities differ between the two.Individuals from both woodrat species were fed diets of each juniper in a cross-over design.Urine,collected over a 24-h period,was extracted and analyzed in an untargeted metabolomics approach using both GC-MS and HPLC-MS/MS.The obligate specialist N.stephensi excreted a unique pattern of Phase I metabolites when fed its native juniper,while N.lepida excreted a unique pattern of Phase II metabolites when fed its native juniper.Both woodrat species utilized the Phase II metabolic pathway of glucuronidation more heavily when consuming the more chemically diverse J.osteosperma,and N.stephensi utilized less glucuronida-tion than N.lepida when consuming J.monosperma.These results are consistent with the hypothesis that obligate specialists may have evolved unique and efficient biotransformation mechanisms for dealing with PSCs in their diet.

Background Our previous study has reported that supplementation of oligosaccharide-based polymer enhances gut health and disease resistance of pigs infected with enterotoxigenic E.coli(ETEC)F18 in a manner similar to carbadox.The objective of this study was to investigate the impacts of oligosaccharide-based polymer or antibiotic on the host metabolic profiles and colon microbiota of weaned pigs experimentally infected with ETEC F18.Results Multivariate analysis highlighted the differences in the metabolic profiles of serum and colon digesta which were predominantly found between pigs supplemented with oligosaccharide-based polymer and antibiotic.The relative abundance of metabolic markers of immune responses and nutrient metabolisms,such as amino acids and carbohydrates,were significantly differentiated between the oligosaccharide-based polymer and antibiotic groups(q2.0).In addition,pigs in antibiotic had a reduced(P<0.05)relative abundance of Lachnospiraceae and Lactobacillaceae,whereas had greater(P<0.05)Clostridiaceae and Streptococcaceae in the colon digesta on d 11 post-inoculation(PI)compared with d 5 PI.Conclusions The impact of oligosaccharide-based polymer on the metabolic and microbial profiles of pigs is not fully understood,and further exploration is needed.However,current research suggest that various mechanisms are involved in the enhanced disease resistance and performance in ETEC-challenged pigs by supplementing this polymer.
