Exercise promotes angiogenesis by enhancing endothelial cell fatty acid utilization via liver-derived extracellular vesicle miR-122-5p

The proteome and metabolome are directly interconnected since protein levels affect the metabolic reprogramming and metabolites’ concentration also influence the expression of related-proteins and activity of enzymes in cells. Therefore, the integration of proteomics and metabolomics may be a powerful tool to understand the process of biological alteration. However, such combined approach has to date been applied in the filed of uncovering the action mechanisms of drug to a limited extent. In this study, we performed an integrated strategy combining two omics to explore the anti-cancer mechanisms of norcantharidin（NCTD） in MHCC97 H and BEL-7402 cells. Sixty-seven responsive（up/down-regulated） proteins and thirty-four significantly altered metabolites were identified in cells exposed to NCTD for 48 h based on proteomics and metabolomics, respectively. Additionally, the integrated analysis of the two omics data with bioinformatics tools showed that several altered biological pathways were highlighted. Of note, the metabolic shift was mediated by NCTD treatment, resulting in increased oxidative phosphorylation and decreased glycolysis coincident with increased cell death as well as reduced colony formation. Moreover, NCTD-induced metabolic shift was also supported by complementary data obtained by western blotting, flow cytometry and seahorse analyses, as evidenced by the attenuation of the extracellular acidification rate, increase of oxygen-consumption rate, down-regulation of glycolysis-related protein levels, and up-regulation of reactive oxygen species. Collectively, our studies provided comprehensive profiling of the proteomic and metabolomic consequences of NCTD-treated cells and revealed a novel mechanism of NCTD in the regulation of cancer cellular metabolism.
