Numerical simulation on the adaptation of forms in trabecular bone to mechanical disuse and basic multi-cellular unit activation threshold at menopause

The objective of this paper is to identify the effects of mechanical disuse and basic multi-cellular unit （BMU） activation threshold on the form of trabecular bone during menopause. A bone adaptation model with mechanical- biological factors at BMU level was integrated with finite element analysis to simulate the changes of trabecular bone structure during menopause. Mechanical disuse and changes in the BMU activation threshold were applied to the model for the period from 4 years before to 4 years after menopause. The changes in bone volume fraction, trabecular thickness and fractal dimension of the trabecular structures were used to quantify the changes of trabecular bone in three different cases associated with mechanical disuse and BMU activation threshold. It was found that the changes in the simulated bone volume fraction were highly correlated and consistent with clinical data, and that the trabecular thickness reduced signi-ficantly during menopause and was highly linearly correlated with the bone volume fraction, and that the change trend of fractal dimension of the simulated trabecular structure was in correspondence with clinical observations. The numerical simulation in this paper may help to better understand the relationship between the bone morphology and the mecha-nical, as well as biological environment; and can provide a quantitative computational model and methodology for the numerical simulation of the bone structural morphological changes caused by the mechanical environment, and/or the biological environment.

Women gradually lose bone from the age of〜35 years,but around menopause,the rate of bone loss escalates due to increasing bone resorption and decreasing bone formation levels,rendering these individuals more prone to developing *** increased osteoclast activity has been linked to a reduced estrogen level and other hormonal ***,it is unclear whether intrinsic changes in osteoclast precursors around menopause can also explain the increased osteoclast ***,we set up a protocol in which CD14f blood monocytes were isolated from 49 female donors(40-66 years old).Cells were differentiated into osteoclasts,and data on differentiation and resorption activity were *** multiple linear regression analyses combining in vitro and in vivo data,we found the following:(1)age and menopausal status correlate with aggressive osteoclastic bone resorption in vitro;(2)the type I procollagen N-terminal propeptide level in vivo inversely correlates with osteoclast resorption activity in vitro;(3)the protein level of mature cathepsin K in osteoclasts in vitro increases with age and menopause;and(4)the promoter of the gene encoding the dendritic cell-specific transmembrane protein is less methylated with *** conclude that monocytes are"reprogrammed"in vivo,allowing them to"remember"age,the menopausal status,and the bone formation status in vitro,resulting in more aggressive *** discovery suggests that this may be mediated through DNA *** suggest that this may have clinical implications and could contribute to understanding individual differences in age-and menopause-induced bone loss.

Objective:To investigate the mechanistic basis for the attenuation of bone degeneration by edible bird's nest(EBN)in ovariectomized ***:Forty-two female Sprage-Dawley rats were randomized into 7 groups(6 in each group).The ovariectomized(OVX)and OVX+6%,3%,and 1.5%EBN and OVX+estrogen groups were given standard rat chow alone,standard rat chow+6%,3%,and 1.5%EBN,or standard rat chow+estrogen therapy(0.2mg/kg per day),*** sham-operation group was surgically opened without removing the *** control group did not have any surgical *** 12 weeks of intervention,blood samples were taken for serum estrogen,osteocalcin,and osteoprotegerin,as well as the measurement of magnesium,calcium abd zinc *** femurs were removed from the surrounding muscles to measure bone mass density using the X-ray edge detection technique,then collected for histology and estrogen receptor(ER)***:Ovariectomy altered serum estrogen levels resulting in increased food intake and weight gain,while estrogen and EBN supplementation attenuated these *** also reduced bone ER expression and density,and the production of osteopcalcin and osteorotegerin,which are important pro-osteoplastic hormones that promote bone mineraliztion and ***,estrogen and EBN increased serum estrogen levels leading to increased bone ER expression,pro-osteoplastic hormone production and bone density(all P<0.05).Conclusion:EBN could be used as a safe alternative to hormone replacement therapys for managing menopausal complications like bone degeneration.

Effect of Jianpi Bushen formula on aromatase-inhibitor-associated bone loss after menopause

OBJECTIVE: To investigate the effect of Jianpi Bushen(JPBS) formula on aromatase inhibitor(AI)-associated bone loss after ***: Six-month-old female rats were randomly divided into 6 groups: a sham group, an ovariectomized(OVX) group, an OVX treated with exemestane and 3 OVX groups each treated with a different dose of JPBS formula. Bone mineral density(BMD) at the lumbar vertebrae, histology, bone markers and serum levels of estrogen were assessed. Furthermore, a cohort study was conducted in 130 postmenopausal women with breast cancer that had undergone treatment with AIs. The subjects were given JPBS + caltrate D or caltrate D only, administered orally. BMD at the lumbar vertebrae and femoral neck and bone markers were evaluated in both control and herbal treatment groups at baseline and 12 ***: Experimental results indicated that a high dose of JPBS significantly increased the trabecular bone area percentage(*** %) and broadened the trabecular thickness(***). The JPBS formula enriched the carboxyterrninal propeptide of type ipmcollagen and increased serum estrogen level significantly. The clinical investigation revealed that bone loss was decreased in the group treated with JPBS vs control(BMD T score at lumbar vertebrae, 3.9% increased vs 14.58% decreased, respectively, P = 0.004 and BMD T score on femoral neck, 1.8% decreased vs 22.45% decreased, respectively, P = 0.008). Besides, JPBS formula elevated Nmiddle osteocalcin and decreased type Ⅰ collagen cross-linked C-terminal ***: JPBS formula prevented aromatase-inhibitor-associated bone loss after menopause by inhibiting bone resorption and promoting bone formation.

menopause is characterized by various physical,mental and emotional *** 731®is a standardized extract from Rheum rhaponticum root and has been clinically studied for its role in reducing menopausal *** current systematic review and meta-analysis aimed to evaluate the efficacy of ERr 731®supplementation in alleviating the severity of menopausal *** this review,we searched across three online databases up to March 2023,evaluated the quality of the included studies by the Physiotherapy Evidence Database scale,and assessed the risk of bias by the Cochrane Risk of Bias *** then performed a metaanalysis using RevMan software to estimate the pooled mean difference(MD).The study protocol was registered in the Prospective Register of Systematic Reviews(CRD42023416808).After screening and evaluation,we included four high-quality studies(a total of 390 participants;the ERr 731®group:193 participants;the control group:197 participants)in the *** results showed that ERr 731®supplementation significantly reduced the menopause Rating Scale score(MD:–15.12;Pmenopause *** bias and high heterogeneity in the results warrant further clinical studies.

Soy food consumption in relation to breast cancer modified by menopause status

It has been found that lifestyle and diet are associated with the development of *** mortality of breast cancer in Wuhan city is increasing,according to the statistics of recent *** case control study was aimed to provide data for alimentary therapeutics for breast *** included a case group(n 5196)and an age frequency-matched control group(n 5202).A validated food frequency questionnaire was used to obtain information on usual food *** results indicate that intake of soy foods more than 5 times per week was associated with a decreased risk of breast cancer,especially in premenopausal *** adjusted OR was 0.294[95%CI was(0.158–0.546),P 50.000].Our results indicate that alimentary therapeutics for breast cancer can be adjusted by status of menopause.

Royal jelly for genitourinary syndrome of menopause:A randomized controlled trial

Objective:Genitourinary syndrome of menopause is a progressive condition characterized by a decrease in estrogen,which causes bothersome genital *** purpose of the study was to determine the effect of royal jelly on the genitourinary syndrome of *** and methods:A randomized controlled trial was carried out from November 2018 to June 2019 in Bandar Abbas,*** trial was registered in the Iranian Registry of Clinical Trials(1RCT20181107041585N1)with the main objective of determining if royal jelly could reduce the genitourinary syndrome of menopausal *** women were randomly assigned to receive either daily 1g of oral royal jelly or placebo for 8 *** urogenital subscale of the Menopausal Rating Scale was used to determine genitourinary *** samples t-test was used for inter-group comparisons and paired samples t-test for pre-and post-treatment ***:There were no differences in the severity of sexual problems,bladder complications,or vaginal dryness between groups before *** the intervention group's bladder complications improved slightly after eight weeks of royal jelly treatment compared to the control group(p?0.04),there were no significant changes in vaginal dryness,sexual problems,or total urogenital *** within-group changes(before and after treatment)also showed no differences in urogenital ***:A daily dose of 1g royal jelly taken orally for 8 weeks did not alleviate menopausal genitourinary *** serious side effects were *** make more reliable decisions about the use and safety of royal jelly in the future,different doses of royal jelly and longer trials are required.

menopause in German and Chinese Women——An Analysis of Symptoms,TCM-diagnosis and Hormone Status

Objective： To investigate menopause-related symptoms, traditional Chinese Medicine （TCM）-diagnosis and hormone status of two comparable urban samples of menopausal women, one in Essen （Germany） and the other in Shanghai （China）. Methods： Patients suffering from menopausesyndrome were recruited from the TCM-outpatient clinic of the University of Duisburg-Essen in Germany （35 subjects） and from the Shuguang Hospital Affiliated to Shanghai University of Traditional Chinese Medicine in China （35 subjects）. The Kupperman-lndex for tracing menopausal symptoms was applied. The complete TCM-diagnosis was carried out by the same investigator in China as well as in Germany. Testosterone and estrogen blood samples were collected once from every woman. Results： There were significant differences in specific symptoms of the Kupperman-lndex, such as a higher prevalence of formication and depression in German menopausal women; whereas Chinese menopausal women suffered significantly more from vertigo, headache and paraesthesia symptoms. Concerning TCM- diagnosis, Shen （肾）-yang deficiency was diagnosed in 51.43% of the German women in contrast to 5.71 % of the Chinese women; 17.14 % of the German women were diagnosed as having Shen-yin deficiency compared to 74.29 % of the Chinese women. The German women showed significantly lower mean hormone levels for testosterone compared to the Chinese women （P≤0.0005）. Conclusions： German and Chinese menopausal women do not show different prevalence but have different patterns of menopausal symptoms. Furthermore, from a TCM point of view, German women suffer more from Shen-yang deficiency whereas Chinese women suffer more from Shen-yin deficiency syndrome. These results are supported by significantly lower levels of testosterone in German women compared to Chinese women, which, in TCM, is a characteristic of yang deficiency.
