Massive pro-inflammatory cytokines production has been correlated with the pathogenesis of severe dengue *** active compound of mangosteen fruit pericarps,α-mangostin,has been commonly used as traditional medicine and dietary *** examined the effect ofα-mangostin against dengue virus(DENV)infection in human peripheral blood mononuclear cells(PBMC)by the measurement of virus titer and TNF-αand IFN-γcytokines concentration post *** concentration ofα-mangostin inhibited virus replication and reduced inflammatory cytokines expression at 24-and 48-h post *** results support the potential use ofα-mangostin as anti-antiviral and anti-inflammatory therapies in the treatment of dengue.

Enhanced Skin Permeation of Mangostin

mangostin, Garcinia mangostana L. is used as a traditional medicine in Southeast Asia for inflammatory and septic ailments, and α-mangostin is one of a major active component from the fruit rind extract of mangosteen. The aim of this study was to develop a topical formulation containing α-mangostin to relieve joint pain, reduce swelling and combat inflammation from osteoarthritis. To determine the concentration of mangostin, an isocratic HPLC method was developed. The assay consisted of a reversed-phase HPLC column and an ultraviolet detector. The mobile phase consisting of methanol and water （90:10 %v/v） was delivered at a flow rate of 1.0 ml/min, and UV detector was set at 320 nm. The assay was shown to be linear over the range 0.24-240 μg/mL （R2 ≥ 0.999）. Solubility of mangostin in different mediums, including aqueous solutions, organic solvents and organic-inorganic solvent mixtures, was analyzed by using the HPLC method. Based on the solubility study, release study was conducted using synthetic membranes to find out the release profile of mangostin from the formulations. Finally, the permeation of mangostin from various formulations was determined with a horizontal diffusion cell by applying excess amount of mangostin in the donor compartment. The in vitro skin permeation study showed the permeation of mangostin can be improved by adding chemical skin penetration enhancers significantly.
