maize chlorotic mottle virus(MCMV) is a quarantine microorganism in ***-infection of MCMV with other viruses in the family of Potyviridae can induce maize lethal disease and impair yield of maize ***,the mechanism of MCMV pathogenesis is largely *** de-epoxidase(VDE) plays important roles in plant defense against biotic and abiotic *** this research,the interaction between MCMV P31 and maize violaxanthin de-epoxidase(ZmVDE) was identified by Y2H and *** knocking down the transcriptional level of ZmVDE led to decreased MCMV accumulation level and CP relative *** the transcriptional level of ZmVDE and its enzymatic activity were down-regulated significantly when symptom appeared,which demonstrated that relative expression level of ZmVDE,as well as the enzymatic activity was attenuated by MCMV *** indicated that the MCMV infection was correlated with *** together,this research demonstrated that MCMV could interfere with the relative expression level and enzymatic activity of ZmVDE,which was correlated with MCMV accumulation.

maize（Zea mays L.） has been widely cultivated in the world,but unfortunately virus diseases of maize severely threat maize *** chlorotic mottle virus（MCMV） is one of the most important virus pathogensin corn which can induce symptoms such as mosaic,stunting and leaf *** worse,MCMV couldcause corn lethal necrosis（CLN） when co-infect with a *** the only established member of the genus Machlomovirus in the family Tombusviridae,MCMV can be transmitted through infected maize seeds,beetles and mechanical inoculation leading to high risk of the transmission of ***,it is crucial to develop a rapid and reliable method to detect *** this study,we established a rapid and effective method to detect MCMV using reverse transcription loop-mediated isothermal amplification（RT-LAMP）.The results showed that the reaction mixtures kept at 63℃ for 60 min were optimal and the sensitivity of the RT-LAMP was 10 to 100 fold than conventional PCR method for detecting *** cross-reactivity was detected with other most common and important viral pathogens infecting maize in *** addition,RT-LAMP products could be stained with SYBR Green Ⅰ in-tube,which facilitated detection of MVMC through observing fluorescentrather than ethidium staining following gel *** method was verified by testing field samples collected from Yunnan province and showed high reliability and *** together,we established a rapid,reliable and visible method for MCMV detection in maize.

maize chlorotic mottle virus(MCMV) is the only member of the genus Machlomovirus in the family Tombusviridae which leads to serious yield losses in maize(Zea mays L.),especially can cause maize lethal necrosis disease by synergistic infection with *** oxygen species(ROS) act as signaling molecules and can be induced during plant-pathogen *** contribute to the synthesis of the phytohormones such as jasmonic acid,auxin,and salicylic acid and play an important role in ROS *** is a class of highly conserved enzymes catalyzing the conversion of hydrogen peroxide(HO) to water and oxygen and plays a key role in the removal of excessive amounts of *** it is important to investigate the relationship between MCMV infection and ROS.A unique protein of MCMV,p31,expressed as a readthrough extension of p7 a,is required for efficient systemic *** identified an interaction between MCMV p31 and maize catalase1(ZmCAT1),a vital enzyme localized in peroxisomes,using the yeast two-hybrid;and the interaction was confirmed via in vivo co-immunoprecipitation assay in Nicotiana benthamiana and bimolecular fluorescence complementation assay in maize *** addition,we obtained evidence that the expression level of ZmCAT1 was upregulated during MCMV *** results provide new insights into the correlation between ROS and virus infection.

maize(Zea mays),as a staple food and an important industrial raw material,has been widely cultivated for centuries especially by smallholder *** lethal necrosis disease(MLND)is a serious disease infecting maize,which caused devastating damage in the African region *** is induced by co-infection of maize chlorotic mottle virus and one of several cereal-infecting viruses in the Potyviridae family,with the symptoms ranging from chlorotic mottle to plant death at different infection *** pest management for MLND needs strengthening detection,focusing on prevention and effective *** detection system of MLND has been successfully established by serological methods,nucleic acid-based methods,next-generation sequencing,*** practices,such as using certified seeds,sanitary measures,crop rotation,tolerant or resistant varieties etc.,have been considered as the effective,economical and eco-friendly way to prevent and control MLND.
