Vδ2Dδ3 rearrangements of T cell receptor (TCR) gene from cerebralspinal fluid (CSF) cells was detected for diagnosis and monitoring of central nervous system leukemia (CNSL) in children with acute lymphoblastic leukemia (ALL) using polymerase chain reaction (PCR) technique. 20 patients were studied and in 12 of them the results of PCR and dot hybridization with clonospecific probes were positive, showing the presence of minimal blast cells in CSF. Our study suggested that the PCR method is an effective tool for clinical diagnosis of CNSL and is much more sensitive than routine CSF examination.

Objective： To investigate the effect of Radix Isatidis polysaccharides （RIP） on the immunological function and expression of immune related cytokines in mice. Methods： Sixty mice were randomly divided into six groups, the normal group, cyclophosphamide （Cy） model group, Levamisole （positive control） group, RIP low, medium and high dose groups （0.08 g/kg, 0.16 g/kg, 0.32 g/kg, respectively）, ten in each group. By detecting the value of abdominal macrophage phagocytic index, serum hemolysin （HC50）, proliferation of lymphocytes and expression of related cytokines, interleukin （IL-2） and interferon γ, （INF-γ）, the effect of RIP on immunological function and its mechanism were studied. Results： RIP could improve the level of hemolysin in immunological function depressed mice. The results showed that the value of macrophage phagocytic index in the high dose RIP group increased from 1.11 ±0.13 to 1.42±0.26. The level of IL-2 and INF- γ, could be decreased by Cy to 38.12±6.88 ng/L and 139.23±29.87 ng/L, respectively, while RIP in high dose could increase the secretion of IL-2 and INF- γ, to 53.54 ± 14.43 ng/L and 189.91 ± 32.63 ng/L, respectively. Conclusion： RIP could enhance non-specific immunological function, humoral immunity and cellular immunity in mice.

In order to study the effect of excessive iodine on immune function of lymphocytes and the role of selenium supplementation with excessive iodine intake, the changes of T lymphocyte number, ratio of subsets, activity of natural killer （NK） cells and lymphocytes proliferation response were investigated. 150 female BALB/C mice were randomly divided into 5 groups in terms of their body weight （n=30 in each group）, and 10 of each group were taken as one batch for test. Mice in the 5 groups were orally administrated with iodine 0 （group Ⅰ ）, 1500 （group Ⅱ）, 3000 （group Ⅲ）, 6000 μg/L （group ）, iodine 6000 μg/L plus selenium 0.3 mg/L （group Ⅴ） respectively for 30 days. lymphocyte proliferation response, CD4^＋/CD8^＋, Th1/Th2 and the activity of NK cells were measured. CD4^＋/CD8^＋ was significantly lower, while lymphocyte proliferation response stronger, and Th1/Th2 and the activity of NK cells significantly higher in group Ⅳ than in group Ⅰ （P〈0.01）. There was no significant difference in all indexes between group Ⅴ and group Ⅰ （P〉0.05）. It was suggested that excessive iodine as exogenous chemical materials can induce disorders of T lymphocyte immune function in mice. 0.3 mg/L selenium supplementation can protect mice against toxicity induced by 6000 μg/L iodine.

Recently, the human cochlea has been shown to contain numerous resident macrophages under steady-state. The macrophages accumulate in the stria vascularis, among the auditory nerves, and are also spotted in the human organ of Corti. These macrophages may process antigens reaching the cochlea by invasion of pathogens and insertion of CI electrode. Thus, macrophages execute an innate, and possibly an adaptive immunity. Here, we describe the molecular markers CD4 and CD8 of T cells, macrophage markers MHCⅡ and CD11b, as well as the microglial markers TEME119 and P2Y12, in the human cochlea. Immunohistochemistry and the advantageous super-resolution structured illumination microscopy(SR-SIM) were used in the study. CD4^+ and CD8^+ cells were found in the human cochleae. They were seen in the modiolus in a substantial number adjacent to the vessels, in the peripheral region of the Rosenthal's canal, and occasionally in the spiral ligament. While there are a surprisingly large number of macrophages in the stria vascularis as well as between the auditory neurons,CD4^+ and CD8^+ cells are hardly seen in these areas, and neither are seen in the organ of Corti. In the modiolus,macrophages, CD4^+ and CD8^+ cells appeared often in clusters. Interaction between these different cells was easily observed with SR-SIM, showing closely placed cell bodies, and the processes from macrophages reaching out and touching the lymphocytes. Otherwise the CD4^+ and CD8^+ cells in human cochlear tissue are discretely scattered. The possible roles of these immune cells are speculated.

Pancreatic cancer is the third leading cause of cancer mortality in both men and women in the United States,with poor response to current standard of care,short progression-free and overall survival.Immunotherapies that target cytotoxic T lymphocyte antigen-4,programmed cell death protein-1,and programmed death-ligand 1 checkpoints have shown remarkable activities in several cancers such as melanoma,renal cell carcinoma,and nonsmall cell lung cancer due to high numbers of somatic mutations,combined with cytotoxic T-cell responses.However,single checkpoint blockade was ineffective in pancreatic cancer,highlighting the challenges including the poor antigenicity,a dense desmoplastic stroma,and a largely immunosuppressive microenvironment.In this review,we will summarize available clinical results and ongoing efforts of combining immune checkpoint therapies with other treatment modalities such as chemotherapy,radiotherapy,and targeted therapy.These combination therapies hold promise in unleashing the potential of immunotherapy in pancreatic cancer to achieve better and more durable clinical responses by enhancing cytotoxic T-cell responses.

The highly complex immuno-hematological system of the recipient has to rebalance itself when the liver is replaced with a graft that has its own system.This gives us an opportunity for observation.Here we consider the graft-to-recipient direction with passenger lymphocyte syndrome(PLS)as well as the recipient-to-graft direction with Factor VIII(FVIII)inhibitors,paroxysmal nocturnal hemoglobinuria(PNH)and graft endothelial replacement with liver transplantation.PLS extends beyond the ABO blood groups to any situation where the donor has been sensitized to a recipient antigen.PLS directed against ABO or minor blood group antigens is usually self limiting whereas Rhesus(Rh)PLS persists with life threatening immune hemolysis.Human platelet antigen(HPA)1A PLS results in life threatening immune thrombocytopenia.Treatments of severe PLS may include reduction in immunosuppression,anti-B-cell therapy,plasmapheresis and splenectomy.Liver transplantation into recipients with FVIII inhibitors has been difficult.Donors with acquired hemophilia may transmit the capacity to make FVIII inhibitors by PLS and should be avoided.Patients with PNH have been transplanted successfully but a considerable cost in the continued use of high dose eculizumab.We speculate that combined bone marrow and liver transplantation would be a better option for recipients with FVIII inhibitors or PNH.Replacement of liver graft endothelium with recipient cells is common and may explain relative transplant tolerance that is believed to occur with liver transplantation.

Amino acids in individual human lymphocytes were determined by capillary zone electrophoresis with electrochemical detection after oncapillary derivatization. In order to inject cells easily, a cell injector was designed. Four amino acids (serine, alanine, taurine, and glycine) in single human lymphocytes have been identified. Quantitation has been accomplished through the use of calibration curves.

We studied apoptosis induced by duck swollenhead hemorrhagic disease virus (DSHDV) in vivo,focusing on the lymphoid organs that constitute the main targets for infection:bursa of Fabricius (BF), thymus,and spleen.72 ducks were inoculated intramuscularly with a virulent strain of DSHDV.The morphology of lymphoid organs of these infected ducks was observed by light microscopy and transmission electron microscopy.lymphocyte apoptosis was observed in DSHDV-infected BF,thymus, and spleen.The apoptotic peak appeared at 72h postinoculation.Cell death by classic necrosis was also observed in dead ducks after 72h postinoculation.,and it was further confirmed by in situ terminal deoxynucleotidyl transferase dUTP nick-end labeling (TUNEL).We conclude in DSHDV-infected ducks that apoptosis of lymphocytes resulted in the depletion of iymphocytes,may play an important role in the pathogenesis of duck viral swollenhead hemorrhagic disease.
