Dietary lycopene supplementation improves meat quality,antioxidant capacity and skeletal muscle fiber type transformation in finishing pigs

This study aimed to investigate effects of dietary lycopene supplementation on meat quality,antioxidant ability and muscle fiber type transformation in finishing pigs.In a 70-day experiment,18 DurocLandraceYorkshire barrows were randomly allocated to 3 dietary treatments including a basal diet supplemented with 0,100 and 200 mg/kg lycopene,respectively.Each dietary treatment had 6 replicates with one pig each.Results showed that dietary 200 mg/kg lycopene supplementation increased muscle redness a^(*)value,intramuscular fat and crude protein contents,and decreased muscle lightness L^(*)and yellowness b^(*)values(Plycopene to the diet of finishing pigs improved color,nutritional value and juiciness of pork after slaughter.Results also showed that dietary lycopene supplementation enhanced antioxidant capacity of finishing pigs(Plycopene significantly increased slow myosin heavy chain(MyHC)protein level and slow-twitch fiber percentage,and decreased fast MyHC protein level and fast-twitch fiber percentage(Plycopene to the diet of finishing pigs promoted muscle fiber type conversion from fast-twitch to slow-twitch.Together,we provide the first evidence that dietary 200 mg/kg lycopene supplementation improves meat quality,enhances antioxidant capacity and promotes muscle fiber type transformation from fast-twitch to slow-twitch in finishing pigs.

lycopene can reduce prostate-specific antigen velocity in a phase Ⅱ clinical study in Chinese population

Background Epidemiological studies have shown that lycopene has anti-prostate cancer effect.In vitro tests also confirmed that it can promote apoptosis of prostate cancer cells.We investigated the effect of whole-tomato supplement lycopene on the prostate-specific antigen velocity in selected prostate cancer patients.Methods Twenty selected prostate cancer patients were given whole-tomato supplement lycopene 10 mg per day for about 6 months.Blood samples of patients were collected weekly to measure serum prostate-specific antigen （PSA） values.PSA velocity slope,which reflects the change of PSA,and the degree of change were also calculated.By comparing the values of average PSA velocity slope （rise or fall of PSA） before and after the administration of lycopene,the effect of lycopene can be evaluated.Blood chemistry analysis was regular followed as safety control.Results Three patients in the research group withdrew within 3 weeks because of inability to conform.The rest 17 patients continued for an average period of 6 months.Two patients withdrew because of cancer progression （PSA rise） who later received active treatment.The average fall in PSA was equivalent to 2.56% over （i.***.an average slope/d of -0.000 28） the first 3 months.In the last 3 months,average fall in PSA was equivalent to 31.58% （i.***.an average slope/d of-0.003 51）.The Wilcoxon rank-sum test showed a statistically significant decrease of PSA velocity slope overall （P=0.000 9）.Analysis of the PSA doubling time （pre-vs.post-treatment） showed a median increase over 3 months but this was not statistically significant （P=0.21）.No toxic side effect was observed during the whole process.The results indicate that the average PSA change is ＂decline＂ in patients,and the degree of the decline is accelerated.Conclusion Administration of lycopene was able to reduce PSA velocity in this study group.

lycopene Accumulation Affects the Biosynthesis of Some Carotenoid-related Volatiles Independent of Ethylene in Tomato

For elucidating the regulatory mechanism of ethylene on carotenoid-related volatiles （open chain） compounds and the relationship between lycopene and carotenoid-related volatiles, transgenic tomato fruits in which ACC synthase was suppressed were used. The transgenic tomato fruit showed a significant reduction of lycopene and aroma volatiles with low ethylene production. 6-methyl-5-hepten-2-one, 6-methyl-5-hepten-2-ol and geranylacetone, which were suspected to be lycopene degradation products, were lower than those in wild type tomato fruits. In order to identify whether lycopene accumulation effects the biosynthesis of some carotenoid-related volatiles independent of ethylene in tomato or not, the capability of both wild type and transgenic tomato fruits discs to convert lycopene into carotenoid-related volatiles was evaluated. The data showed that external lycopene could convert into 6-methyl-5-hepten-2-one and 6-methyl-5-hepten-2-ol in vivo, indicating that the strong inhibition of ethylene production had no effect on enzymes in the biosynthesis pathway of some carotenoid-related volatUes. Therefore, in ACS-suppression transgenic tomato fruits, the low levels of 6-methyl- 5-hepten-2-one, 6-methyl-5-hepten-2-ol was due to decreased lycopene accumulation, not ethylene production. Ethylene only affected the accumulation of lycopene, and then indirectly influenceed the level of lycopene-related volatiles.

lycopene is a natural compound that alleviates oxidative stress and inflammation,exerting therapeutic effects in a number of cancers.The aim of this study is to investigate the efficacy of lycopene in inhibiting prostate cancer.Cell viability assays indicated the dose-and time-dependent toxicity of lycopene in prostate cancer cells.Annexin V/propidium iodide double-staining assays revealed the strong apoptotic effects of lycopene.The levels of inflammatory factors,including interleukin-1 (ILl),IL6,ILS,and tumor necrosis factor-α(TNF-α),in lycopene-treated cells were also reduced by lycopene treatment.With the increasing dose of lycopene,the survival of mice bearing prostate cancer xenografts was significantly improved (P lycopene is a promising chemotherapy drug,which inhibits prostate cancer progression by suppressing the inflammatory response.

Excessive amounts of reactive oxygen species （ROS） cause a state of oxidative stress, which result in sperm membrane lipid peroxidation, DNA damage and apoptosis, leading to decreased sperm viability and motility. Elevated levels of ROS are a major cause of idiopathic male factor infertility, which is an increasingly common problem today. lycopene, the most potent singlet oxygen quencher of all carotenoids, is a possible treatment option for male infertility because of its antioxidant properties. By reacting with and neutralizing free radicals, lycopene could reduce the incidence of oxidative stress and thus, lessen the damage that would otherwise be inflicted on spermatozoa. It is postulated that lycopene may have other beneficial effects via nonoxidative mechanisms in the testis, such as gap junction communication, modulation of gene expression, regulation of the cell cycle and immunoenhancement. Various lycopene supplementation studies conducted on both humans and animals have shown promising results in alleviating male infertility--lipid peroxidation and DNA damage were decreased, while sperm count and viability, and general immunity were increased. Improvement of these parameters indicates a reduction in oxidative stress, and thus the spermatozoa is less vulnerable to oxidative damage, which increases the chances of a normal sperm fertilizing the egg. Human trials have reported improvement in sperm parameters and pregnancy rates with supplementation of 4-8 mg of lycopene daily for 3-12 months. However, further detailed and extensive research is still required to determine the dosage and the usefulness of lycopene as a treatment for male infertility.

The Protective Role of Procyanidins and lycopene Against Mercuric Chloride Renal Damage in Rats

Objective This study aims to investigate the protection of procyanidins and lycopene from the renal damage induced by mercuric chloride.Methods Rats were treated with either procyanidins or lycopene 2h before HgCl 2 subcutaneously injection,once daily treatment for 2 successive days.Results In comparison with HgCl 2 group,markers of renal function such as blood urea nitrogen in serum and urinary protein were decreased to （18.45±11.63） mmol/L and （15.93±9.36） mmol/L,（4.54±0.78） g/（g Cr） and （4.40±1.12） g/（g Cr）.N‐acetyl‐beta‐D‐glucosaminidase,lactate dehydrogenase,alkaline phosphatase in urine were depressed to （125.49±11.68） U/（g Cr）,（103.73±21.79） U/（g Cr）,（101.99±12.28） U/（g Cr）,and （113.19±23.74） U/（g Cr）,（71.14±21.80） U/（g Cr）,（73.64±21.51） U/（g Cr） in procyanidins and lycopene groups.Indicators of oxidative stress,for example,Glutathion was reduced to （45.58±9.89） μmol/（g pro） and （45.33±5.90） μmol/（g pro）,and antioxidant enzymes such as superoxide dismutase,glutathione‐peroxidase were enhanced to （43.07±10.97） U/（mg pro） and （39.94±6.04） U/（mg pro）,（83.85±18.48） U/（mg pro）,and （85.62±12.68） U/（mg pro）.Malondialdehyde was lowered to （0.95±0.12）（μmol/g pro） and （1.03±0.12） μmol/（g pro） in procyanidins and lycopene groups.ROS generation was decreased by 27.63% and 16.40% and apoptosis was also decreased in procyanidins and lycopene groups respectively.Pathological changes were much better as well.Conclusion Procyanidins and lycopene play some protective role against mercury kidney damage.

Effect of lycopene on androgen receptor and prostate-specific antigen velocity

Background There is increasing interest in the role of dietary factors in both the development and behaviour of prostate cancer.This study was carried out to evaluate the impact of the dietary factor lycopene on DNA synthesis,activity and expression of the androgen receptor gene element in prostate LnCaP cells and to report our pilot phase Ⅱ study investigating its effect on prostate-specific antigen velocity over one year.Methods LnCaP cells were grown with or without different concentrations of lycopene or tetrahydrofuran （THF solvent）added to the culture medium for 48 hours.DNA synthesis was measured by the incorporation of bromodeoxyuridine （Brdu） into DNA during a 4-hour pulse, followed by immunostaining and visualization of stained cells using fluorescence microscopy.A transient transfection of a plasmid DNA recombinant containing an androgen receptor element-luciferase （ARE-Luc） report gene into LnCaP cells was developed and the impact of different concentrations of lycopene on the androgen receptor element was reflected by quantitative analysis of the luciferase enzyme function.Expression of the androgen gene was also studied by Western blotting.The phase Ⅱ pilot study patients （n=41） previously diagnosed with prostate cancer were enrolled and given lycopene supplement, 10 mg per day, and response was measured by observing changes in the plasma prostate-specific antigen （PSA） levels.Results The addition of 0.5 μmol/L, 5 μmol/L, 10 μmol/L and 15 μmol/L of lycopene was shown to inhibit cell growth by 2.66%, 4.29%, 3.73% and 13.66%, respectively, compared with the THF solvent control samples （P=0.015）.As compared with the RPMI1640 medium group, cell proliferation in the presence of 5 μmol/L, 10 μmol/L, and 15 μmol/L lycopene was inhibited by 8.12%, 6.33% and 12.00%, respectively （P=0.024）.We showed for the first time that lycopene inhibited the activity of the androgen receptor gene element in a dose-related manner.Inhibition was seen in the transcription of the luciferase construct and confirmed by androgen

Possible mechanisms of lycopene amelioration of learning and memory impairment in rats with vascular dementia

Oxidative stress is involved in the pathogenesis of vascular dementia. Studies have shown that lycopene can significantly inhibit oxidative stress;therefore, we hypothesized that lycopene can reduce the level of oxidative stress in vascular dementia. A vascular dementia model was established by permanent bilateral ligation of common carotid arteries. The dosage groups were treated with lycopene(50, 100 and 200 mg/kg) every other day for 2 months. Rats without bilateral carotid artery ligation were prepared as a sham group. To test the ability of learning and memory, the Morris water maze was used to detect the average escape latency and the change of search strategy. Hematoxylin-eosin staining was used to observe changes of hippocampal neurons. The levels of oxidative stress factors, superoxide dismutase and malondialdehyde, were measured in the hippocampus by biochemical detection. The levels of reactive oxygen species in the hippocampus were observed by dihydroethidium staining. The distribution and expression of oxidative stress related protein, neuron-restrictive silencer factor, in hippocampal neurons were detected by immunofluorescence histochemistry and western blot assays. After 2 months of drug administration,(1) in the model group, the average escape latency was longer than that of the sham group, and the proportion of straight and tend tactics was lower than that of the sham group, and the hippocampal neurons were irregularly arranged and the cytoplasm was hyperchromatic.(2) The levels of reactive oxygen species and malondialdehyde in the hippocampus of the model group rats were increased, and the activity of superoxide dismutase was decreased.(3) lycopene(50, 100 and 200 mg/kg) intervention improved the above changes, and the lycopene 100 mg/kg group showed the most significant improvement effect.(4) Neuron-restrictive silencer factor expression in the hippocampus was lower in the sham group and the lycopene 100 mg/kg group than in the model group.(5) The above data indicate that lycopene 100 mg/kg co

lycopene modulates cellular proliferation, glycolysis and hepatic ultrastructure during hepatocellular carcinoma

AIM To investigate the effect of lycopene extracted from tomatoes(LycT) on ultrastructure, glycolytic enzymes, cell proliferation markers and hypoxia during N-Nitrosodiethylamine(NDEA)-induced hepatocarcinogenesis. METHODS Female BALB/c mice were randomly divided into four groups: The Control, NDEA(200 mg NDEA/kg b.w. given i.p.), LycT(5 mg/kg b.w. given orally on alternate days) and Lyc T + NDEA group. The m RNA and protein expression of various cell proliferation markers(PCNA, Cyclin D1, and p21) were assessed by reverse transcription-polymerase chain reaction and enzyme linked immunosorbent assay, respectively. The ultrastructure of hepatic tissue was analyzed using scanning and transmission electron microscopy. The enzymatic activity of glycolytic enzymes was estimated using standardized protocols, while glucose-6-phosphate dehydrogenase activity level was estimated using a kit obtained from Reckon Diagnostic P. Ltd.(India). RESULTS Uncontrolled proliferation in the liver of NDEA(P ≤ 0.001) mice was evident from the high expression of cell-proliferation associated genes(PCNA, Cyclin D1, and p21) when compared to control and Lyc T mice. In addition, enhanced activities of hexokinase, phosphoglucoisomerase, aldolase, glucose-6-phosphatedehydrogenase and hypoxia-inducible factor-1α were observed in NDEA mice as compared to control(P ≤ 0.001) and Lyc T(P ≤ 0.001) mice. The alterations in hepatic ultrastructure observed in the NDEA group correlated with the changes in the above parameters. Lyc T pre-treatment in NDEA-challenged mice ameliorated the investigated pathways disrupted by NDEA treatment. Moreover, hepatic electron micrographs from the Lyc T + NDEA group showed increased macrophages, apoptotic bodies and well-differentiated hepatocellular carcinoma(HCC) in comparison to undifferentiated HCC as observed in the NDEA treated group. CONCLUSION This study demonstrates that dietary supplementation with Lyc T has a multidimensional role in preventing HCC development.

lycopene stabilizes lipoprotein levels during D-galactosamine/lipopolysaccharide induced hepatitis in experimental rats

Objective:To investigate the effect of lycopene on lipoprotein metabolism during D-galactosamine/lipopolysacchoride(D-Gal/LPS)induced hepatitis in experimentul rats.Methods:The efficacy of lycopene was validated during D-Gal/LPS induced hepatitis by analyzing the activity of lipid metabolizing enzymes such as lipoprotein lipase(LPL),lecithincholesterol acyl transferase(LCAT)and hepatic triglyceride lipase(HTCL).Lipo protein analyses were done by the estimation of very low density lipoprotein cholesterol(VLDL),low density lipoprotein cholesterol(LDL)and high density lipoprotein cholesterol(HDL).Remits:The toxic insult of D-galactosamine/lipopolysaccharide(D-Gal/LPS)in experimental group of animals reduces the normal values of lipid metabolizing enzymes due to liver injury.The significant drop in the levels of HDL and concomitant increase in the values of VLDL and LDL were observed.The pretreatment of lycopene restore these altered values to near normal level in experimental group of animals.Conclusions:In the light of results,it can be concluded that administration lycopene stabilizes the lipoprotein levels by regulating the lipid metabolizing enzymes through its antioxidant defense and helps to maintain the normal lipid metabolism during toxic injury in liver.
