Recent applications and strategies in nanotechnology for lung diseases

lung diseases,including COVID-19 and lung cancers,is a huge threat to human health.However,for the treatment and diagnosis of various lung diseases,such as pneumonia,asthma,cancer,and pulmonary tuberculosis,are becoming increasingly challenging.Currently,several types of treatments and/or diagnostic methods are used to treat lung diseases;however,the occurrence of adverse reactions to chemotherapy,drug-resistant bacteria,side effects that can be significantly toxic,and poor drug delivery necessitates the development of more promising treatments.Nanotechnology,as an emerging technology,has been extensively studied in medicine.Several studies have shown that nano-delivery systems can significantly enhance the targeting of drug delivery.When compared to traditional delivery methods,several nanoparticle delivery strategies are used to improve the detection methods and drug treatment efficacy.Transporting nanoparticles to the lungs,loading appropriate therapeutic drugs,and the incorporation of intelligent functions to overcome various lung barriers have broad prospects as they can aid in locating target tissues and can enhance the therapeutic effect while minimizing systemic side effects.In addition,as a new and highly contagious respiratory infection disease,COVID-19 is spreading worldwide.However,there is no specific drug for COVID-19.Clinical trials are being conducted in several countries to develop antiviral drugs or vaccines.In recent years,nanotechnology has provided a feasible platform for improving the diagnosis and treatment of diseases,nanotechnology-based strategies may have broad prospects in the diagnosis and treatment of COVID-19.This article reviews the latest developments in nanotechnology drug delivery strategies in the lungs in recent years and studies the clinical application value of nanomedicine in the drug delivery strategy pertaining to the lung.

lung diseases and their related complications represent a critical source of morbidity and mortality globally and have become a research focus in recent years.There are plenty of hazards that threaten the health of lung by exposure to external environmental stimuli,such as dust,cigarette smoke,PM2.5,air pollution and pathogen infection.These risks lead to the impairment of lung function and subsequent lung diseases including pneumonia,chronic obstructive pulmonary disease(COPD),asthma and idiopathic pulmonary fibrosis(IPF).Compared with antibiotics and corticosteroids therapies,traditional Chinese medicine prescriptions are more effective with fewer side effects.A considerable variety of bioactive ingredients have been extracted and identified from Chinese herbal medicines and are used for the treatment of different lung diseases,including resveratrol.Increasing studies have reported promising therapeutic effects of resveratrol against lung diseases by inhibiting oxidative stress,inflammation,aging,fibrosis and cancer both in vitro and in vivo.In this review,the recent progress in the studies of lungprotective effects and underlying mechanisms of resveratrol and also highlight the potency of resveratrol and traditional Chinese prescriptions containing resveratrol as promising therapeutic options were summarized for the treatment of lung and respiratory diseases.

Objective:To summarize the rules of acupoint selection of acupoint application to prevent and treat lung diseases under the background the post-epidemic era using data-mining technology.Method:The CNKI,Wanfang database,and VIP database were searched for clinical study articles on lung diseases treated by acupoint application published in the past 5 years.Data-eligible papers were extracted to establish a database of acupoint application for lung disease using Microsoft Excel 2019,with the goal of analyzing the frequency of acupoints,acupoint-meridian association,acupoint-location association,specific acupoint frequency,and cluster analysis.Association rules,consisting of acupoints with an application frequency of≥10,were devised by the Apriori algorithm to explore the correlation between acupoint groups and to analyze the rules of the compatibility of acupoint prescriptions.Results:A total of 229 eligible papers met our inclusion criteria.Forty-seven acupoints were applied,for a total frequency of acupoints of 1,035 times.Among these,acupoints for lung diseases were primarily distributed in the back-and-waist and chest-and-abdomen areas.From the analysis of the association rules,we obtained four groups of acupoint association rules based on acupoint clusters with a frequency≥10 and found that Feishu(BL 13),Tiantu(CV 22),Dazhui(GV 14),Dingchuan(EX-B1),and Danzhong(CV 17)constitute the core acupoints of prescriptions for clinical acupoint application to prevent and treat lung diseases.Conclusion:It is clearly shown that the core acupoints are relatively concentrated and that the selected acupoints were mainly locally selected,which could be a matching reference for the long-term prevention and treatment of lung diseases,including COVID-19.

lungs are structurally and functionally complex organs consisting of diverse cell types from the proximal to distal axis.They have direct contact with the external environment and are constantly at risk of various injuries.Capable to proliferate and differentiate,pulmonary endogenous progenitor stem cells contribute to the maintenance of lung structure and function both under homeostasis and following injuries.Discovering candidate pulmonary endogenous progenitor stem cell types and underlying regenerative mechanisms provide insights into therapeutic strategy development for lung diseases.In this review,we reveal their compositions,roles in lung disease pathogenesis and injury repair,and the underlying mechanisms.We further underline the advanced progress in research approach and potential therapy for lung regeneration.We also demonstrate the feasibility and prospects of pulmonary endogenous stem cell transplantation for lung disease treatment.

Several subtypes of avian influenza A have been shown to cross the species barrier and infect humans, leadingto human cases of avian influenza） Till June 2, 2009, globally there were 433 confirmed human cases of avian influenza caused by H5N1 virus, with a death rate of 60.5%.3 This is far higher than the reported 11% death rate of severe acute respiratory syndrome （SARS）.4 The epidemiologic features of human case of influenza A subtype H5N1 virus infection consist of high incidence rate in cold weather, high susceptibility in population of younger age associated with rapid onset of the disease and devastating illness in humans.12＇5＇6 H5N1 virus is mostly transmitted to humans directly through contact with infected birds or their secretions and the patients present with an influenza type illness with fever, cough, sore throat, malaise, and gastrointestinal symptoms; which can result in a rapidly progressive primary viral pneumonia and respiratory failure.2 Patients above the age of 5 years are likely to have an adverse course of disease.5

There is a growing concern in mining community about the contribution of nano-particulates to miner's health.Despite the health influence of respirable dusts and associated lung diseases have been recognized for decades in the mining industry,the nano-scale particulates accompanying with complicated physiochemical properties and their enor-mous contribution in quantity have been drawing attentions only in recent a few years because of the advancement of nano-science discipline.In this review,we examine the current regulations of dusts exposure and the dominant mass-based monitoring methods to point out the ignorance of nano-particulates in mining industry.The recognized mining-related nano-particulates sources are summarized to identify the mechanically generated finer particulates including particles and aerosols.In addition,the mechanism of adverse health impact on miner with exposure to nano-scale particulates is discussed in a detail to emphasize their substantial detriment as a potential respiratory hazard.Characterization of the complex physiochemical properties of nano-particulates are then summarized and discussed because these properties could be different from regular respirable dusts due to their dramatically increased surface area and particulate counts.The intent of this review is to demonstrate the potential of adverse health effect of nano-particulate on the mine personals throughout the mining cycle and to identify the research gaps of the mine nano-particulate characterization and quantification.We suggest that further understanding of the mining induced nano-particulate properties and their pathogenesis are critical for the future engineering control measure to mitigate the potential health threat for future miners.

BACKGROUND Cryptococcus presenting as an opportunistic pathogen mainly affects immunocompromised patients,but the disseminated form of infection is rare among immunocompetent populations.The partial radiographic characteristics of pulmonary cryptococcosis mimic lung carcinoma,leading to unnecessary open chest exploratory surgery,and the lack of a gold-standard noninvasive diagnostic increases the risk of misdiagnosis.Positron emission tomography/computed tomography(PET/CT),a sensitive method for distinguishing malignant tumors,coupled with cryptococcal latex agglutination test showing a high positive rate may overcome these issues.CASE A 36-year-old man presented for general examination,without health complaints.Routine CT showed multiple pulmonary nodules and a mass with high maximum standardized uptake value.Initially,we suspected primary malignancy with hematogenous metastasis.Although his routine fungal analysis had been negative,subsequent CT-guided percutaneous core needle biopsy and histopathology examination indicated a diagnosis of pulmonary cryptococcosis.Fluconazole(200 mg/d)antifungal drug treatment was initiated,and 1 mo later the pulmonary mass had reduced in size markedly(on chest CT scan)without any complications.CONCLUSION Serologic and PET/CT examinations may not rule out cryptococcosis,and percutaneous lung puncture is critical under all circumstances.

Rationale and Objectives: Cystic lung disease may be accurately diagnosed by imaging interpretation of specialist radiologists, without other information. We hypothesized that with minimal training non-specialists could perform similarly to specialist physicians in the diagnosis of cystic lung disease. Methods: 72 cystic lung disease cases and 25 cystic lung disease mimics were obtained from three sources: 1) a prospective acquired diffuse lung disease registry, 2) a retrospective search of medical records and 3) teaching files. Cases were anonymized, randomized and interpreted by 7 diffuse lung disease specialists and 15 non-specialist radiologists and pulmonologists. Clinical information other than age and sex was not provided. Prior to interpretation, non-specialists viewed a short PDF training document explaining cystic lung disease interpretation. Results: Correct first choice diagnosis of 85%-88% may be achieved by high-performing specialist readers and 71%-80% by non-specialists and lower-performing specialists, with mean accuracies in the diagnosis of LAM (91%, p Conclusion: With specific but limited training, non-specialist physicians can diagnose cystic lung diseases from CT appearance alone with similar accuracy to specialists, correctly identifying approximately 75% of cases.

Innate lymphoid cells(ILCs),a newly identified member of the lymphoid population,play a critical role in the transition from innate to adaptive immunity in host defense.ILCs are important in mucosal barrier immunity,tissue homeostasis,and immune regulation throughout the body.Significant alterations in ILC responses in lung diseases have been observed and reported.Emerging evidence has shown that ILCs are importantly involved in the pathogenesis and development of a variety of lung diseases,i.e.,helminth infections,allergic airway inflammation,and airway hyper-responsiveness.However,as a tissue-resident cell population,the role of ILCs in the lung remains poorly characterized.In this review,we discuss the role of ILCs in lung diseases,the mechanisms underlying the ILCmediated regulation of immunity,and the therapeutic potential of modulating ILC responses.
