Objective Behavioral flexibility plays critical roles in adaptation to unstable environment for ***,little is known regarding the cellular and molecular mechanism underlying behavioral *** By combining an inducible,reversible chemical-genetic technique with in vitro electrophysiology and behavioral methods,we manipulated the activity of ot-Ca/calmodulin-dependent protein kinaseⅡ(αCaMKII),on the time scale of minutes in specific brain subregions of freely behaving mice to investigate the effect of aCaMKII overexpression on synaptic plasticity at thalamic input synapses onto the lateral amygdale(T-LA) pathway,amygdale-related behavioral flexibility and emotional behaviors inαCaMKII-F89G transgenic ***(1) Low frequency stimulation(1 Hz,15 min and 3 Hz,5 min) failed to induce the robust NMDA-dependent long term depression(LTD) at T-LA pathway inαCaMKII-F89G transgenic slices as did in wild-type ***,0.5μm NM-PP1,a specific inhibitor ofαCaMKII-F89G could restore deficit of LTD in the transgenic ***,no significant difference in long term potentiation(LTP) between CaMKII-F89G transgenic and wild-type slices.(2) Fear extinction,conditioned taste aversion extinction were utilized to test the behavioral flexibility in CaMKII-F89G transgenic ***αCaMKII-F89G transgenic mice exhibited impaired performances,which were reversed by oral treatment of NM-PP1(0.5μM).(3) Transgenic animals had normal locomotor activity,exploratory ability, motor coordination and ability in open field and rotarod *** addition,there was no significant difference in level of anxiety and depression between two groups in elevated plus maze,forced swimming and tail suspension *** Forebrain overexpression ofαCaMKII impairs behavioral flexibility and NMDAR-dependent LTD of amygdala,suggesting that the deficit of amydala LTD may underlie the performance deficits in behavioral flexibility.

Priming stimulation modifies synaptic plasticity in the perforant path of hippocampal slice in rat

Objective The potential of all central nervous system synapses to exhibit long term potentiation （LTP） or long term depression （LTD） is subject to modulation by prior synaptic activity, a higher-order form of plasticity that has been termed metaplasticity. This study is designed to examine the plasticity and metaplasticity in the lateral perforant path of rat. Methods Field potential was measured with different priming and conditioning stimulation protocols. Results Ten-hertz priming, which does not affect basal synaptic transmission, caused a dramatic reduction in subsequent LTP at lateral perforant path synapses in vitro, and the reduced LTP lasted for at least 2 h. The LTD was unaffected. The reduction of LTP in the lateral perforant path was also readily induced by applying priming antidromically at the mossy fibers. Conclusion Priming with 10 Hz, which is within a frequency range observed during physiological activity, can cause potent, long-lasting inhibition of LTP, but not LTD. This form of metaplasticity adds a layer of complexity to the activity-dependent modification of synapses within the dentate gyrus.
