Craniofacial therapy:advanced local therapies from nano-engineered titanium implants to treat craniofacial conditions

Nano-engineering-based tissue regeneration and local therapeutic delivery strategies show significant potential to reduce the health and economic burden associated with craniofacial defects,including traumas and tumours.Critical to the success of such nano-engineered non-resorbable craniofacial implants include load-bearing functioning and survival in complex local trauma conditions.Further,race to invade between multiple cells and pathogens is an important criterion that dictates the fate of the implant.In this pioneering review,we compare the therapeutic efficacy of nano-engineered titanium-based craniofacial implants towards maximised local therapy addressing bone formation/resorption,soft-tissue integration,bacterial infection and cancers/tumours.We present the various strategies to engineer titanium-based craniofacial implants in the macro-,micro-and nano-scales,using topographical,chemical,electrochemical,biological and therapeutic modifications.A particular focus is electrochemically anodised titanium implants with controlled nanotopographies that enable tailored and enhanced bioactivity and local therapeutic release.Next,we review the clinical translation challenges associated with such implants.This review will inform the readers of the latest developments and challenges related to therapeutic nano-engineered craniofacial implants.

The corrosive environment in long-distance natural gas pipeline was simulated by the online high shear stress flow test platform.The interaction between flow fields and local corrosion in different local corrosion stages was studied by machining different depths rectangular defect pit(RDP)on X80 pipe steel specimens.The electrochemical signals of each specimen under high shear stress flow were measured online using an integrated three-electrode and electrochemical system.Raman spectroscopy confirmed that the corrosion scale of X80 pipeline steel in CO_(2)-saturated National Association of Corrosion Engineers solution was composed of FeCO_(3).The scanning electron microscope images displayed variations in microstructure of the corrosion scale at different RDP depths and different areas.The flow field fluctuations induced by RDP were analyzed by computational fluid dynamics simulations and the development of local corrosion pits was discussed in terms of integrity of corrosion scale,convective mass transfer,and diffusion mass transfer.

In this paper,the influence of laser wavelength instability,polarization fading and phase fluctuation on local heterodyne detection wavelength scanning Brillouin optical time domain reflectometer(WS-BOTDR)is theoretically analyzed,and a local heterodyne detection WS-BOTDR system is built for experimental verification.The experimental results show that with the increase of sensing distance,the adverse effect of laser wavelength instability,polarization fading and phase fluctuation on local heterodyne detection WS-BOTDR is gradually aggravated,which will lead to the broadening and distortion of the wavelength power spectrum(WPS),resulting in large errors in demodulated Brillouin central wavelength(BCW)and temperature.The average temperature measurement errors at the positions of 1 km,5 km,9 km non-heating section and 9.45 km heating section are 1.76℃,3.42℃,3.89℃and 4.3℃,respectively.

Pyroptosis represents a host-protective mechanism that promotes clearance of pathogens by initiating the recruitment of immune cells to the site of infection.In a study recently published in Nature,Deng et al.extensively outlined a hitherto unknown role of the group A Streptococcus(GAS)-derived cysteine protease streptococcal pyrogenic exotoxin B(SpeB)in gasodermin A(GSDMA)cleavage as the primary step in induction of pyroptosis.As clearly shown in GSDMA-deficient mice infected with SpeBproducing GAS,this cleavage event is key to preventing the systemic spread and fatal course of GAS following local skin infection[1].

local community detection aims to find a cluster of nodes by exploring a small region of the network.local community detection methods are faster than traditional global community detection methods because their runtime does not depend on the size of the entire network. However, most existing methods do not take the higher-order connectivity patterns crucial to the network into consideration. In this paper, we develop a new local Community Detection method based on network Motif(LCD-Motif) which incorporates the higher-order network information. LCD-Motif adopts the local expansion of a seed set to identify the local community with minimal motif conductance, representing a generalization of the conductance metric for network motifs. In contrast to PageRanklike diffusion methods, LCD-Motif finds the community by seeking a sparse vector in the span of the local spectra,such that the seeds are in its support vector. We evaluate our approach using real-world datasets across various domains and synthetic networks. The experimental results show that LCD-Motif can achieve a higher performance than state-of-the-art methods.

Neurons of the mature central nervous system(CNS,mainly the brain and spinal cord)are unable to regenerate spontaneously after a lesion,in contrast to neurons of the peripheral nervous system(PNS).While the extraneuronal environment was long thought to be limiting,evidence was given less than 15 years ago that neurons themselves are critical players of their own regeneration(Park et al.,2008).Indeed,CNS neurons show a decline of axon growth capacity as they mature and after an injury.Today,the role of axonal translation is actively explored in the paradigm of embryonic neuronal growth and in peripheral nerve injury and regeneration,but less is known about the role of local protein synthesis in regrowth of adult CNS axons.Here we discuss how the current understanding of axonal translation in the CNS may contribute to the development of novel strategies to enhance axon regeneration in the injured CNS.

Optical nanoantennas can convert propagating light to local fields.The local-field responses can be engineered to exhibit nontrivial features in spatial,spectral and temporal domains,where local-field interferences play a key role.Here,we design nearly fully controllable local-field interferences in the nanogap of a nanoantenna,and experimentally demonstrate that in the nanogap,the spectral dispersion of the local-field response can exhibit tuneable Fano lineshapes with nearly vanishing Fano dips.A single quantum dot is precisely positioned in the nanogap to probe the spectral dispersions of the local-field responses.By controlling the excitation polarization,the asymmetry parameter q of the probed Fano lineshapes can be tuned from negative to positive values,and correspondingly,the Fano dips can be tuned across a broad spectral range.Notably,at the Fano dips,the local-field intensity is strongly suppressed by up to ~50-fold,implying that the hot spot in the nanogap can be turned into a cold spot.The results may inspire diverse designs of local-field responses with novel spatial distributions,spectral dispersions and temporal dynamics,and expand the available toolbox for nanoscopy,spectroscopy,nano-optical quantum control and nanolithography.

The thalamostriatal pathway is implicated in Parkinson's disease(PD); however, PD-related changes in the relationship between oscillatory activity in the centromedian-parafascicular complex(CM/Pf, or the Pf in rodents) and the dorsal striatum(DS) remain unclear.Therefore, we simultaneously recorded local field potentials(LFPs) in both the Pf and DS of hemiparkinsonian and control rats during epochs of rest or treadmill walking. The dopamine-lesioned rats showed increased LFP power in the beta band(12 Hz–35 Hz) in the Pf and DS during both epochs, but decreased LFP power in the delta(0.5 Hz–3 Hz) band in the Pf during rest epochs and in the DS during both epochs, compared to control rats. In addition,exaggerated low gamma(35 Hz–70 Hz) oscillations after dopamine loss were restricted to the Pf regardless of the behavioral state. Furthermore, enhanced synchronization of LFP oscillations was found between the Pf and DS after the dopamine lesion. Significant increases occurred in the mean coherence in both theta(3 Hz–7 Hz) and beta bands,and a significant increase was also noted in the phase coherence in the beta band between the Pf and DS during rest epochs. During the treadmill walking epochs, significant increases were found in both the alpha(7 Hz–12 Hz)and beta bands for two coherence measures. Collectively,dramatic changes in the relative LFP power and coherence in the thalamostriatal pathway may underlie the dysfunction of the basal ganglia-thalamocortical network circuits in PD, contributing to some of the motor and non-motor symptoms of the disease.
