Metabolomics and gene expression levels reveal the positive effects of teaseed oil on lifespan and aging process in Caenorhabditis elegans

As an irreplaceable dietary constituent,lipids play a vital important role in health,but their effects on aging process and longevity are still not well known yet.In this paper,the metabolic profiling and gene expression levels of Caenorhabditis elegans were investigated to explore the effects of different edible oils on senescence and lifespan.The results showed that teaseed oil(TO)could prolong the life expectancy and slow down the aging process of C.elegans.Compared to the control group,the intake of lard oil(LO)and TO increased the expression levels of genes related to inhibition of protein aggregation(akt-1,daf-16,hsf-1,hsp-16.2)and lipid metabolism(daf-7,daf-1,mdt-15,lipl-4,fat-5,fat-6,fat-7),with a more significant alteration in TO group.Metabolomics revealed that palm oil can upregulated plenty of fatty acids(palmitic acid,stearic acid,tetracosanoic acid),together with some amino acids(tryptophan,L-aspartate,L-valine)and carbohydrate(D-glucose),while the trend was opposite in TO group.Besides,moderate-to-strong correlations were found between differential metabolites and changed genes.In general,this paper claimed that TO could prolong lifespan and slow down aging process via regulating the lipids,amino acids and carbohydrates metabolism.

A number of research has shown that the plant polyphenol resveratrol,one of the most prominent small molecules,has beneficial protective effects in multiple organisms,including worms,flies,and killifish.To understand the effects of resveratrol on lifespan,we evaluated its effects in the silkworm Bombyx mori.In this study,we found that lifespan was significantly prolonged in both female and male silkworms treated with resveratrol.Silkworm larval weight was significantly increased from day 3 of the 5th larval instar(L5D3)to day 7 of the 5th larval instar(L5D7).However,the weight of the pupa,cocoon,and total cocoon was not significantly different in female silkworms with resveratrol treatment than that in controls.Meanwhile,resveratrol significantly improved the thermotolerance of the silkworms,which enhanced their survival rate.Moreover,antioxidant activity was increased by resveratrol in both female and male silkworms.Furthermore,an antioxidant-related signalling pathway,SIRT7-FoxO-GST,was activated in silkworms with resveratrol treatment.Collectively,these results help us to understand the molecular pathways underlying resveratrol induced pro-longevity effects and indicate that silkworm is a promising animal model for evaluating the effects of lifespan-extending drugs.

In this paper, we consider the lifespan of solution to the MHD boundary layer system as an analytic perturbation of general shear flow. By using the cancellation mechanism in the system observed in [12], the lifespan of solution is shown to have a lower bound in the order of ε^(-2-) if the strength of the perturbation is of the order of ε. Since there is no restriction on the strength of the shear flow and the lifespan estimate is larger than the one obtained for the classical Prandtl system in this setting, it reveals the stabilizing effect of the magnetic field on the electrically conducting fluid near the boundary.

Diets these days contain more fats. High fat diet (HFD) is a model of unhealthy eating in experimental animals. It is known to induce inflammatory responses and oxidative stress. Cannabidiol (CBD), the non-psychoactive component of Cannbis sativa has been legalized for medicinal use in many countries of the world. Omega 3 fatty acid, commonly found in fish oil is medicinal and necessary for brain development. The antioxidant and neuroprotective functions of CBD and omega 3 have made them relevant in many researches. In this study, 45 mice were used and divided into three groups of 15 animals each: Group 1 (normal feed and water ad libidum);Group 2 (HFD ad libidum);Group 3 (HFD + CBD + Omega 3) for 16 weeks. Thereafter, five animals from each group were selected and their frontal lobes were harvested for histological analyses using H and E staining. The remaining mice were allowed to feed on normal diet and observed till death. At the end, HFD significantly reduced life span (57.4 ± 0.3) when compared to control (78.9 ± 1.6) and HFD + CBD + omega 3 group (74.5 ± 0.8) at p < 0.05. HFD also caused significant brain ischemic damage, neuronophagia and significant perivascular oedema. CBD + Omega 3 induced significant astrocytosis compared to control and HFD group. The immune stimulation by the CBD + Omega 3 could be responsible for tissue survival and longevity by protection from inflammatory and oxidative injuries. Ischaemic tissue death could have been prevented by amelioration of artheroma formation due to HFD. Further studies will be required to ascertain other possible mechanisms behind these findings.

Effect of β-guanidinopropionic acid on the lifespan of drosophila and the underlying mechanism

C.elegans have six longevity models,including isp-1,rsks-1,daf-2,eat-2,glp-1,let-363.One interesting question caught attention of us was whether the six longevity models share same pathway to regulate longevity.Recently,Louis R.Lapierre found that HLH-30 regulates autophagy and regulates lifespan in six longevity models.In C.elegans,different mutants with diverse life spans are being used to investigate genes influence life span and various of signal pathways in the aging process.Paraquat（PQ）,a kind of Reactive Oxygen Species（ROS） inducer,recent studies show that low dose PQ is beneficial for lifespan.In this study,we found a significant raise in ROS in six longevity models.Though,ROS are potentially toxic in the body,however,they are also important signaling molecules for regulating aging.Here we demonstrated that ROS activate ERK,which was associated with autophagy process.The phosphorylation of ERK promotes the SKN-1/Nrf2 accumulation of nucleation.Together our findings illustrate the mechanism that the production of ROS.

Aim β-guanadinopropionic acid （β-6PA）, a creatine analog has been demonstrated to lead to thechronic activation of AMPK with increasing mitochondrial biogenesis. However, little is known about the effect of β-GPA on lifespan extension. In current study, we explore whether β-GPA produces the lifespan extension in adult Drosophila and underlying mechanism. Methods The flies were fed in a normal chow diet containing different concentrations （300 raM,900 mM,2700 mM）of β-GPA. Western blot was also used to investigate protein expres- sions. RNA interference was employed to examine effects of lifespan in Drosophila by 13-GPA. Results 900 mM and 2700 mM β-GPA significant extended the lifespan and activated AMPK both in male and female Drosophila with elevated level of autophagy. We found that Atg8 was significantly upregulated and P62 was significantly down- regulated by 900 mM 13-GPA, on the other hand, when the expression of Atg5 protein, an essential protein for au- tophagy, was reduced by RNAi, the effect of β-GPA on lifespan extension was abolished. Inhibition of AMPK by either AMPK-RNAi or compound （2, significantly attenuated the expression of autophagy-related proteins and lifes- pan extension in Drosophila. We found that the activation of AMPK induced by β-GPA inhibited mTORC1 activity, implicating that mTORC1 may not prevent the activation of ULK1/Atgl by AMPK. Conclusion β-GPA can ex- tend the lifespan of Drosophila and this effect is mediated by AMPK-Atgl dependent autophagy.
