Identification of protein targets for the antidepressant effects of Kai-Xin-San in Chinese medicine using isobaric tags for relative and absolute quantitation

Kai-Xin-San consists of Ginseng Radix, Polygalae Radix, Acori Tatarinowii Rhizoma, and Poria at a ratio of 3:3:2:2. Kai-Xin-San has been widely used for the treatment of emotional disorders in China. However, no studies have identified the key proteins implicated in response to Kai-Xin-San treatment. In this study, rat models of chronic mild stress were established using different stress methods over 28 days. After 14 days of stress stimulation, rats received daily intragastric administrations of 600 mg/kg Kai-Xin-San. The sucrose preference test was used to determine depression-like behavior in rats, while isobaric tags were used for relative and absolute quantitation-based proteomics to identify altered proteins following Kai-Xin-San treatment. Kai-Xin-San treatment for 2 weeks noticeably improved depression-like behaviors in rats with chronic mild stress. We identified 33 differentially expressed proteins: 7 were upregulated and 26 were downregulated. Functional analysis showed that these differentially expressed proteins participate in synaptic plasticity, neurodevelopment, and neurogenesis. Our results indicate that Kai-Xin-San has an important role in regulating the key node proteins in the synaptic signaling network, and are helpful to better understand the mechanism of the antidepressive effects of Kai-Xin-San and to provide objective theoretical support for its clinical application. The study was approved by the Ethics Committee for Animal Research from the Chinese PLA General Hospital(approval No. X5-2016-07) on March 5, 2016.

Sensory and motor nerve fibers of peripheral nerves have different anatomies and regeneration functions after injury. To gain a clear understanding of the biological processes behind these differences, we used a labeling technique termed isobaric tags for relative and absolute quantitation to investigate the protein profiles of spinal nerve tissues from Sprague-Dawley rats. In response to Wallerian degeneration, a total of 626 proteins were screened in sensory nerves, of which 368 were upregulated and 258 were downregulated. In addition, 637 proteins were screened in motor nerves, of which 372 were upregulated and 265 were downregulated. All identified proteins were analyzed using the Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analysis of bioinformatics, and the presence of several key proteins closely related to Wallerian degeneration were tested and verified using quantitative real-time polymerase chain reaction analyses. The differentially expressed proteins only identified in the sensory nerves were mainly relevant to various biological processes that included cell-cell adhesion, carbohydrate metabolic processes and cell adhesion, whereas differentially expressed proteins only identified in the motor nerves were mainly relevant to biological processes associated with the glycolytic process, cell redox homeostasis, and protein folding. In the aspect of the cellular component, the differentially expressed proteins in the sensory and motor nerves were commonly related to extracellular exosomes, the myelin sheath, and focal adhesion. According to the Kyoto Encyclopedia of Genes and Genomes, the differentially expressed proteins identified are primarily related to various types of metabolic pathways. In conclusion, the present study screened differentially expressed proteins to reveal more about the differences and similarities between sensory and motor nerves during Wallerian degeneration. Comparison of isobaric and Non-isobaric Operations on a Two-Phase Dynamic Model for PEFC Cathode Gas Diffusion Layer

Two 1-D dynamical and isothermal models of cathode gas diffusion layer(GDL) with isobaric and non-isobaric operations for polymer electrolyte fuel cells(PEFCs) were developed and implemented in COMSOL Multiphysics v3.5.The artificial diffusion coefficient was introduced as well to make the numerical computation be stable.In the non-isobaric model,the pressure of gas mixture was obtained by summing up the governing equations of gaseous components,instead of Navier-Stoks equation.Comparison of the two models were carried out with the steady-states and dynamical simulations under given conditions.The corresponding analysis based on the simulated results was also given simultaneously.This paper is contributed to finding the differences between the isobaric and non-isobaric operation in the two-phase model of cathode GDL.

The isoscaling and the isobaric yield ratio difference(IBD) probes, both of which are constructed by yield ratio of fragment, provide cancelation of parameters. The information entropy theory is introduced to explain the physical meaning of the isoscaling and IBD probes. The similarity between the isoscaling and IBD results is found, i.e.,the information uncertainty determined by the IBD method equals to β- α determined by the isoscaling(α(β) is the parameter fitted from the isotopic(isotonic) yield ratio).

Probing Temperature from Intermediate Mass Fragment by isobaric Yield Ratio in Heavy-Ion Collisions

A probe to extract the temperature(T_(iyr)) from intermediate-mass fragment has been proposed using the isobaric yield ratio(IYR) difference based on the modified Fisher model.By analyzing the isobaric cross section in the measured 140 A MeV^(40,48)Ca+~9Be reactions,the Tiyr can well fit the IYR difference for the large mass fragments.Because of the fragments are produced in different colliding regions,it is also suggested to select the fragments according to different mechanisms when using TIYR.

Effect of inlet rotating swirl on endwall film cooling for two representative hole arrangements

The swirl generator is widely used in lean-burn combustor to guarantee flame stability and reduce NOxemissions. Thus, the non-uniformities induced by the swirler would affect and damage film-cooling effectiveness on the turbine components, even blow-off coolant coverage, to some certain extent. The arrangement of film-cooling holes was normally designed to be perpendicular to the axial direction and in standard straight row. In this work we experimentally studied the effects of inlet swirl and mass flow ratio on traditional film cooling holes arrangement and a new arrangement pattern whose holes are located along the isobars. Results indicated that the swirl perturbation would damage film coverage. The film-cooling effect of endwall on which the holes are along the isobars, is invariably more promising than that of endwall on which hole arrangement is perpendicular to axial with the same mass flow of coolant, whether the inlet conditions is uniform or not.

iTRAQ-based proteomics analysis of aqueous humor in patients with dry age-related macular degeneration

AIM: To preliminarily test proteomics in aqueous humor in patients with dry age-related macular degeneration(AMD) by using the proteomic technology.METHODS: Aqueous humor samples were collected from patients with or without dry AMD, who underwent cataract surgery. The aqueous samples were analyzed with isobaric tags for relative and absolute quantification(i TRAQ) combined with liquid chromatography tandem mass spectrometry(LC-MS/MS) technology. The differential expressed proteins were analyzed with gene ontology(GO) enrichment, Kyoto Encyclopedia of Genes and Genomes(KEGG) and protein-protein interaction(PPI) network analysis. The data were partly validated by ELISA and Western blot. False discovery rate(FDR) was used for statistical analysis. RESULTS: A total of 244 proteins were detected, in which 38 proteins were up-regulated and 51 were down-regulated significantly in patients with dry AMD compared with that in control groups(FDR value <1.0%). Several proteins, e.g., protein S100-A8(S10 A8), dystroglycan(DAG1), Ig alpha-1 chain C region(IGHA1), carbonic anhydrase 3(CAH3) and alpha-1-acid glycoprotein(A1 AG1) were increased more than 5 times of that in control group. The bioinformatics analysis showed that dry AMD is closely associated with inflammation or immune reaction, oxidative stress, blood coagulation and remodeling of extracellular matrix.CONCLUSION: i TRAQ-based proteomic analysis of aqueous humor demonstrate the differential expressions of proteins between dry AMD and control groups, providing the clues to understand the mechanisms and possible treatments of dry AMD.

Residue Coulomb Interaction Among Isobars and Its Influence in Symmetry Energy of Neutron-Rich Fragment

The residue Coulomb interaction(RCI), which affects the result of symmetry-energy coefficient of neutronrich nucleus in isobaric yield ratio(IYR) method, is difficult to be determined. Four RCI approximations are investigated:(i) The M1–RCI adopting the ac/T(the ratio of Coulomb energy coefficient to temperature) determined from the IYR of mirror-nucleus fragments;(ii) The M2–RCI by fitting the difference between IYRs;(iii) The M3–RCI adopting the standard Coulomb energy at a temperature T = 2 Me V; and(iv) Neglecting the RCI among isobars. The M1–, M2– and M3–RCI are no larger than 0.4. In particular, the M2–RCI is very close to zero. The effects of RCI in asym/T of fragment are also studied. The M1– and M4–asym/T are found to be the lower and upper limitations of asym/T, respectively. The M2–asym/T overlaps the M4–asym/T, which indicates that the M2–RCI is negligible in the IYR method, and the RCI among the three isobars can be neglected. The relative consistent low values of M3–asym/T(7.5 ± 2.5) are found in very neutron-rich isobars.

The isobaric vapor-liquid equilibrium data of butanone(1)-N, N-dimethylformamide (DMF)(2) at 100.92kPa, 93.32kPa, and 79.99kPa and of toluene(1)-DMF(2) at 100.92kPa were measured using a modified Rose-Williams still. The above data met the thermodynamic consistency test and were correlated with the Wilson,NRTL, and UNIQUAC equations. These data can be used in the analysis and design of the process that involves separating DMF from butanone and toluene in the leather synthesis industry.

Study Pressure Fields Affecting Cyclone Rainfall: Case Study of Iran

The aim of this study is the synoptic analysis of the atmospheric conditions concurrent with the heavy precipitations of Lorestan, Fars and Hormozgan provinces and determination of the temporal and spatial distribution of them. Requested atmospheric data for this research was taken from databases related to the National Organization of Atmosphere and Oceanology of United States. More maps of the site were taken. Then the earth surface, 500,700 and 850 Hecto Pascal maps, orbital and meridional winds, earth temperature of earth surface and top of the atmosphere were prepared in Grads software and some of maps were drawn in GIS software. The following results were obtained: On the earth surface map, the main controller role is with the Siberian high-pressure, the Azores’s dynamic high-pressure and integrated mode of the Sudan and the Mediterranean low-pressure systems. The main controller role in high levels is with the high altitude center of Azores in the back of the system, the high altitude center of Arabia in the front of the system and deep traffic of North Africa. Another important factor is the moisture mixture of Mediterranean Sea, Red Sea, Persian Gulf, Oman Sea and Indian Ocean. Air rise factor with enhancing meridional winds that causes subsidence of cold air, rising warm air and strengthening the polar front is one of the heavy rainfall conditions too. Eastern winds cause entering of moisture from the Indian Ocean and western winds cause strengthening of cyclones and heavy rainfall in the region.
