The knowledge on the formation,structure,and reactivity of low-coordinate transition-metal species lays the basis for modern organometallic chemistry.While the conventional wisdom of organometallic chemistry in this aspect has been focusing on the noble metal complexes,the renaissance of base metal catalysis in the recent years urges a deeper understanding on low-coordinate 3d metal chemistry.This presentation will introduce our studies on iron and cobalt complexes with N-heterocyclic carbene（NHC）ligation.NHCs are found excellent ancillary ligands to stabilize iron（IV,III,II,I,0）/cobalt（V,IV,III,II,I,0）complexes with their coordination numbers ranging from 4to 2（Fig.1）.[1-5]The reactivity pattern of the low-coordinate species of oxidative addition,group-transfer,sigma-bond metathesis,and metal-carbon bond homoleptic scission will be discussed..

We herein report a novel iron（Ⅲ） activated, anticancer, iridium prodrug. This complex contains a m-imino catechol group which can selectively bond to, and be oxidized by, free iron（Ⅲ） inside the cell. Subsequent oxidative rearrangement releases iron（Ⅱ） and hydrolysis of the amine bond, in acidic conditions, produces an aminobipyridyl iridium complex and o-hydroxy-p-benzoquinone. This chain of reactions has a threefold effect. First iron（Ⅱ） can catalyze the decomposition of hydrogen peroxide into hydroxyl radicals. Second, benzoquinone compounds can interfere with the respiratory chain. Third, the conversion of the prodrug to the amino-iridium complex reveals a remarkable increase in luminescence. These properties, as well as the high iron（Ⅲ） content and acidity of cancer cells, make Ir-diol a highly selective and efficient theranostic agent. To the best of our knowledge, Ir-diol is the first metal-based compound that has been developed for chemotherapy using iron（Ⅲ） to enhance both selectivity and potency.

Cell wall is a fundamental plant constituent,rigid but plastic,which provides structural support for plant development and diverse cell shape formation.Cell wall is composed of the polysaccharides cellulose,hemicellulose,pectin andlignin,meanwhile residues a large number of glycoproteins.Several glycosylphosphatidylinositol(GPI)-anchored glycoproteins have been described as important regulators for cell wall formation,while the underlying mechanism is poorly understood.Here,we identify a glycosylphosphatidylinositol(GPI)-anchored glycoprotein SKU5,as well as its homolog SKU5-similar 1(SKS1),which are highly co-expressed with cell wall regulatory genes at transcription level.Further studies show thatsku5 sks1 double mutant decreases crystallized cellulose level while elevates callose deposition along the cell wall at the epidermis-cortex junction area,consequently exhibiting pronounced cell wall protrusion.As multi-copper oxidases,SKU5 and SKS1 might function as cell wall ferroxidase to regulate iron homeostasis,leading to the unexpected callose deposition;meanwhile,GPI anchor motifs are dispensable for the location and functionality of SKU5/SKS1.This study unveils a novel regulatory module of cell wall pattern formation,by which SKU5 and SKS1 coordinate CESA-dependent cellulose synthesis as well as iron-dependent callose deposition,which helps more insights into the mechanism of plant cell shape formation.

＜正＞The effects of myricetin on 6-hydroxydopamine induced neurodegeneration in the substantia nigra-striatum system were investigated.By high-performance liquid chromatography electrochemical detection,we showed that the dopamine content in the striatum decreased after 6-hydroxydopamine treatment,which could be restored by myricetin. The immunohistochemistry and semi-quantitative RT-PCR studies showed that myricetin could prevent the 6- hydroxydopamine-induced decrease of tyrosine hydroxylase positive neurons and the tyrosine hydroxylase mRNA expression in the substantia nigra.Perls’ iron staining study further demonstrated that myricetin prevented the 6- hydroxydopamine-induced increase of iron-staining cells in the substantia nigra.These results suggested that the protective effects of myricetin on the toxicity of 6-hydroxydopamine could be attributed to the myricetin-suppressed iron toxicity.

Background: Multiple sclerosis（MS） is a central nervous system（CNS） inflammatory disorder, which characterized by demyelination and axonal damage. It dustups neuronal communication in CNS leading to uncoordinated movement and muscle weakness for the patients. One feature of MS is iron accumulation in affected brain regions, and we propose to facilitate iron exportation as disease treatment. Methods: An experimental autoimmune encephalomyelitis（EAE） mouse was generated to mimic MS. 10 mg/kg Ceruloplasmin（Cp） was injected by i.p. and the behaviors of the treated mice were recorded. The mice were sacrificed10 days after Cp treatment, and the spinal cord and brain slices of each group were stained by Luxol Fast Blue（LFB） for myelination. Protein expression of Cp in the spinal cord was detected by western blot. The immune cells were analyzed by flow cytometry. Metal concentrations were detected by AAS. Results: We found a dose-dependent protection/rescue of EAE mice by Cp treatment, evidenced by reduced neuroscore, improved myelination, suppressed neuroinflammation, and reduced iron.Conclusions: Cp can be further investigated to treat MS.

＜正＞Objective Curcumin is a well-defined flavonoid.It has a neuroprotective effect due to its free radical scavenging, iron-chelating and anti-inflammatory actions in different tissues.But the mechanism of its protective effects on dopaminergic neurons is still not clear.Methods HPLC-ECD was used to determine the content of DA and its metabolites:3,4- dihydroxyphenylacetic acid（DOPAC）,homovanillic acid（HVA） in striatum（Str）.Immunohistochemistry assay was used to detect the numbers of dopaminergic neurons.Perl’s iron-staining to detect the iron levels in substantia nigra（SN）.Results （1） After 6-OHDA lesions,the DA and its metabolite DOPAC contents in the Str decreased compared with that of control, while the HVA level was not changed.In the curcumin treatment group,the DA and its metabolites contents increased compared with that of 6-OHDA treatment group.（2） The number of tyrosine hydroxylase（TH） positive neurons in the SN decreased after 6-OHDA lesions compared with that of control group.Curcumin significantly inhibited 6-OHDA-induced decrease of the numbers of TH positive neurons.（3） The number of iron-staining cells increased in the SN decreased after 6-OHDA lesions compared with that of control group.Curcumin significantly inhibited 6-OHDA-induced increase of the numbers of iron-staining cells.Conclusion Curcumin could significantly reduce the toxic effects of 6-OHDA on dopaminergic neurons in rat SN via its iron-chelating property.

Objective The purpose of this study was to elucidate whether Rg 1 affected the levels of iron and the expression of iron transporter DMT1 in MES23.5 dopaminergic cells after MPP treatment and investigate the possible signal pathway. Methods Western blots was used to examine the expression of DMT1 and NF-κB.Ferrous iron influx into MES23.5 cells was determined by the quenching of calcein fluorescence.The production of ROS was assessed by the oxidation of 2’,7’- dichlorodihydrofluorescein diacetate(H2DCFDA).And changes in the mitochondrial membrane potential were measured by rhodaminel23 using flow cytometry.Results Findings showed introduction of Rg1 before MPP inhibited ferrous iron influx and iron induced cell damage by preventing the upregulation of DMT1-IRE.Further study showed this translocation of NF-κB could be inhibited by Rg1 pretreatment.Furthermore an antioxidant,N-acetyl-L-cysteine(NAC),pretreatment could also abolish this translocation,suggesting MPP-induced translocation of NF-κB was caused by the increased production of ROS.Conclusion These results suggested the neuroprotective effect of Rg1 was attributed to the potent antioxidant,preventing nuclear translocation of cell death promoting NF-κB,attenuating the upregulation of DMT1-IRE and decreasing the iron influx and iron-induced oxidative stress.
