Inhibitory effect of iron on in vitro proliferation of smooth muscle cells

Background iron is a biocorrodible metal that might be used in bioabsorbable stents.This study investigated the effects at the cellular and protein levels of soluble divalent iron （ferrous gluconate） and soluble trivalent iron （ferric chloride） on the proliferation of human aortic smooth muscle cell （HASMC） in vitro.Methods The water-soluble tetrazolium （WST-1） test was used to evaluate the effect of iron on proliferation of HASMC and Western blotting was used to measure the levels of signaling proteins involved in proliferative and apoptosis pathways.Results HASMC proliferation was inhibited in a concentration dependent manner after treatment with soluble divalent and trivalent iron at concentrations of 100-500 μmol/L.Western blotting analysis showed that the proliferating cell nuclear antigen （PCNA） expression following treatment with soluble divalent iron and trivalent iron at 100,300 and 500 μmol/L was reduced compared to the control.The PCNA expression decreased with increasing iron concentration and to a greater extent with the trivalent iron than with the divalent iron treatment group.The p53 expression was markedly increased in a concentration dependent manner in both iron treatment groups.Conclusion The soluble divalent iron and,to a greater degree trivalent iron,inhibited HASMC proliferation in a dosedependent manner,which may be attributed to reduction of PCNA expression and increase of p53 expression.

Effects of iron on Growth and Intracellular Chemical Contents of Microcystis aeruginosa

Objective To investigate the effect of iron on the growth, physiology and photosynthesis of cyanobacteria. Methods A gradient of iron concentrations was employed to investigate the growth, photo-pigments （chlorophyll A and phycocyanin）, and cell chemical contents （C, N, P） of Microcystis aeruginosa in response to different iron additions. Results The specific growth rate during the exponential growth phase, as well as the cell chlorophyll A and the phycocyanin content, was limited by iron below 12.3μmol Fe.L^-1. The growth was inhibited when the iron concentration was at 24.6 μmol Fe.L^-1. The cell chlorophyll A and the phycocyanin content were saturated when the iron concentration was above 12.3μmol Fe.L^-1 and declined slightly at 24.6 μmol Fe.L^-1. At a low iron concentration （about 6.15 μmol Fe-L^-1 and less）, the cell nitrogen and carbohydrate content were iron limited, and the variation of the cell phosphorus content was similar to that of the nitrogen and carbohydrate, with a transition point of 12.3 μmol Fe.L^-1. Conclusion The variation ofcynobacteria growth is synchronous with that of the photo-pigments or the cell chemical content, and there exist relationships among photosynthesis, growth and internal chemical content, which could be useful for the growth estimation from the cell characteristics.

Prevalence of iron deficiency in patients aged 75 years or older with heart failure

Background The latest studies presented at the American Heart Association (AHA) meeting on heart failure and the update of the European Cardiology Society’s (ECS) recommendations on heart failure in 2016 recommend intravenous iron supplementation in patients with heart failure, reduced ejection fraction and iron deficiency for improves walking performance and quality of life, and reduces morbidity. In the present study, we investigated the prevalence of iron deficiency in heart failure patients aged 75 years or older, as there is currently no data on these patients. Methods We performed an observational study on hospitalized patients in Geriatric Cardiology Department. Among the 462 patients hospitalized during eight months, 176 were eligible for inclusion;22 patients was significant interference with an inflammatory syndrome (high ferritin with high C-reactive protein), and for 13 patients iron-related data were not available. For each patient included, a complete iron assessment and type of heart failure was available. Results A total of 141 patients were included, the mean age was 88 years (range: 75–101), and there were 52 (36.9%) of patients with reduced ejection fraction (EF), 37 (26.2%) with mid-range EF, and 52 (36.9%) with preserved EF. Irrespective of heart failure type, 73.8% had iron deficiency (95% CI: 65.7%–80.8%);this was found in 57.7%(95% CI: 43.2%–71.3%) of those with reduced EF, 78.4%(95% CI: 61.8%–90.2%) of those with mid-range EF, and 86.5%(95% CI: 74.2%–94.4%) of those with preserved EF (P = 0.003). Conclusion The prevalence of iron deficiency was very high in very elderly patients with heart failure, especially those with HF with mid-range EF or HF with preserved EF.

iron and inorganic carbon in Liaodong Gulf sediments of Bohai Sea in China

iron in seawater is an essential trace metal for phytoplankton that plays an important role in the marine carbon cycle. But most studies focused on oceanic iron fertilization in high nutrient low chlorophyll （HNLC） seawaters. A study of inorganic carbon （IC） forms and its influencing factors was presented in Liaodong Gulf sediments, and especially the influence of iron was discussed in detail. Inorganic carbon in Liaodong Gulf sediments was divided into five forms： NaCl, NH3·H2O, NaOH, NH2OH·HCl and HCl. The concentration of NaCl and NaOH forms were similar and they only occupied the minority of total inorganic carbon （TIC）. However, NH3 ·H2O, NH2OH · HCl and HCl forms were the principal forms of TIC and accounted for more than 80% of TIC. Especially, the percentage of NH3·H2O form was much higher than that in the Changjiang River Estuary and Jiaozhou Bay sediments. All forms of inorganic carbon were influenced by organic carbon, pore water, iron, pH, redox potential （Eh） and sulfur potential（Es） in sediments, moreover, the influences had different characteristics for different IC forms. However, the redox reactions of iron affected mainly active IC forms. iron had little effect on NH2OH· HCl and HCl forms of IC which were influenced mainly by pH. iron had a stronger influence on NaCl, NaOH and NH3· H2O forms of IC ; the influence of Fe^2＋ was higher than Fe^3＋ and its effect on NH3 ·H2O form was stronger than on NaCl and NaOH forms.

Mechanical and corrosion properties of partially degradable bone screws made of pure iron and stainless steel 316L by friction welding

This paper reports a series of in vitro, ex vivo and in vivo mechanical and corrosion studies of pin and screw prototype made of friction welded pure iron and 316 L type stainless steel aiming to evaluate the applicability of the partially removable bone screws. Results showed that the pin possesses bending, tensile and torsional strengths of1706±147, 666±7 and 0.34±0.03 MPa, respectively. The pin degraded at an average weight loss rate of 17.15×10-5 g cm-2 day-1 and released Fe ions at an average concentration of 2.38 ppm. Plastic deformation induced by torsion increased the corrosion rate of the pin from 0.0014 to 0.0137 mm year-1.The maximum pull-out load of the screw prototypes was3800 N with a calculated failure strength by shear load equal to22.2 k N which is higher than the strength of the cortical bone.Detailed analysis of the rat's blood cells during 60 days of the pin implantation indicated a normal response with low neutrophils/lymphocytes ratio of 0.3-0.5. iron ion concentration in the rat's blood slightly increased from 55 to 61 ppm without affecting the tissue recovering and healing phase. Histological evaluation confirmed the presence of macrophage cells as a normal response to the released iron particles around the iron section of the pin.

Reconstruction of three-dimensional grain structure in polycrystalline iron via an interactive segmentation method

Using a total of 297 segmented sections,we reconstructed the three-dimensional(3D) structure of pure iron and obtained the largest dataset of 16254 3D complete grains reported to date.The mean values of equivalent sphere radius and face number of pure iron were observed to be consistent with those of Monte Carlo simulated grains,phase-field simulated grains,Ti-alloy grains,and Ni-based super alloy grains.In this work,by finding a balance between automatic methods and manual refinement,we developed an interactive segmentation method to segment serial sections accurately in the reconstruction of the 3D microstructure;this approach can save time as well as substantially eliminate errors.The segmentation process comprises four operations:image preprocessing,breakpoint detection based on mathematical morphology analysis,optimized automatic connection of the breakpoints,and manual refinement by artificial evaluation.

Clinical Factors Associated with Hepatocellular iron Deposition in End-stage Liver Disease

Background and Aims: Hepatocellular iron accumulation in patients with chronic liver disease has been linked to adverse outcomes. The objective of this study was to identify clinical factors associated with hemosiderosis. Methods: A total of 103 consecutive liver transplant recipients were identified, in whom liver biopsy had been performed prior to transplantation. Laboratory and clinical data at biopsy and transplant were abstracted from the medical records and hepatocyte iron was graded in the biopsy and explant. The association of change in iron score from biopsy to transplant, with the time interval between these two events, was examined using linear mixed model analysis for repeated measures. Results: Most subjects had advanced fibrosis (F3-F4) at liver biopsy, which was performed on average about 2.5 years before transplant. Over 80% of patients had no or 1+ hepatocyte iron at biopsy;iron increased between biopsy and transplant in about 40%. The only demographic or clinical feature that correlated with increased iron was the presence of a transjugular intrahepatic portosystemic shunt. Increased iron at transplant was associated with higher serum iron and transferrin saturation at biopsy, and with lower hemoglobin level, greater mean corpuscular volume, mean corpuscular hemoglobin and mean corpuscular hemoglobin concentration, higher ferritin and model for end-stage liver disease score at transplant.Conclusions: The development of hemosiderosis in end-stage liver disease is associated with lower hemoglobin levels and alterations in red blood cell indices that are suggestive of hemolysis. These observations suggest that extravascular hemolysis may play a role in the development of secondary iron overload.

Oxidation mechanism and non-isothermal kinetic studies on carbonate iron ore by thermogravimetric analysis

Due to the uncertain and heterogeneous compositions of carbonate iron ore after magnetization roasting,an oxidation pretreatment was applied to increase the uniformity of the composition of the ore.The oxidation of carbonate iron ore was characterized using a thermogravimetric method over a temperature range of 723-948 K.It is shown that the oxidation process of carbonate iron ore is complicated and consists of multiple reactions that include the decomposition and oxidation of carbonates such as FeCO3,MnCO3,MgCO3 and CaMg(CO3)2.The main product that results from oxidation is Fe2O3.Because of the low content of other carbonates (MnCO3,MgCO3 and CaMg(CO3)2)in raw carbonate iron ore and the small change in the apparent activation energies during oxidation,the oxidation of carbonate iron ore can be treated as a single-step FeCO3 oxidation reaction.Based on the Ozawa-Flynn-Wall method,which is one of the model-free methods, the apparent activation energy for the oxidation of carbonate iron ore is 284.50 kJ mol^-1.The most probable reaction model for the oxidation of carbonate iron ore is the Sestak-Berggren model as determined by the theoretical and actual master plot and eigenvalues of the Malek method.The reaction orders n and m are 1.317 and 0.0295,respectively.The preexponential factor is 2.628 ×10^17 min^-1.
