A Cyclometalated iridium（ⅲ） complex Serves as a Phosphorescent Probe for Specific Mitochondrial Imaging in Living Cells

A new cyclometalated iridium（ⅲ） complex [Ir（2-pq）2（HPIP）]C1 （lrl, 2-pq=3-phenylisoquinoline, HPIP= 2-（2-hydroxyphenyl）imidazo[4,5-f] 1,10-phenanthroline） was synthesized and applied to image mitochondria in living cells. Irl displayed excellent ability to selectively accumulate in mitochondria of live cells with no requirement of replacement of the culture medium. Due to Irl exhibiting better photostability than the commercially available mitochondrial trackers, this complex could be applicable for the imaging and tracking of the mitochondrial mor- phological changes over long periods of time. In addition, in vitro cytotoxicity investigation revealed that it! showed negligible cytotoxicity at the concentrations employed. Based on these, Irl is suitable for organelle-selective imaging in living cells.

iridium（ⅲ） complexes are one of the most important electrophosphorescent dyes with tunable emissions in the range of visible and near infrared lights,high photoluminescence yields and short lifetimes for high-efficiency organic light-emitting diodes（OLED） with 100% exciton harvesting.This review summarizes the recent development of electroluminescent Ir^（3＋） complexes functionalized with host-featured carrier-transporting groups,with emphasis on correlations between functionalization,optoelectronic properties and device performance.According to the introducing approaches,the complexes were sorted with conjugated and aliphatic linkages,as well as the types of functional groups.The modification effect on physical properties and the state-of-the-art device performances were discussed.

Synthesis and Luminescent Properties of Cationic Ir(Ⅲ) complexes of 2-Phenylquinoline Derivatives

Two cationic iridium（ⅲ） complexes, [（pqcm）2Ir（pybz）]（PF6）（Ir1） and [（pqcm）2Ir（apybz）]（PF6）（Ir2）（pqcmH = 2-phenyl-quinoline-4-carboxylic acid methyl ester, pybz = 2-pyridyl-benzimidazole, apybz = 1-allyl-2-pyridyl-benzimidazole）, were readily synthesized from the reaction of IrⅢ-μ-chloro-bridged dimer [Ir（pqcm）2（Cl）]2 and corresponding ancillary ligands, and characterized by NMR and mass spectroscopies. The structure of It2 was also confirmed by single-crystal X-ray diffraction. The photophysical properties of the two complexes were also investigated. Irl shows deep red emission peaked at around 652 nm with the phosphorescence quantum yield of ca. 0.29 and the emission lifetime of 233 ns, while Ir2 shows red emission peaked at around 615 nm with the phosphorescence quantum yield of ca. 0.13 and the emission lifetime of 430 ns. The active hydrogen on pybz ligand is believed to have a great influence on the photophysical properties of Ir1.

A series of novel metal iridium complexes containing phenanthroline derivatives were designed and synthesized by changing the substituent groups on the phenanthroline derivatives [1]. However, its biocompatibility is often unsatisfactory, and we need to find a new way to deal with it. In many drug-loading systems, liposome is a low-toxic, biodegradable common drug-loading material. Due to the hydrophobic nature of phospholipids, liposomes can self-assemble to form vesicles with phospholipid bilayers. And the vesicles can encapsulate hydrophilic or hydrophobic molecules. In addition, when the liposome is modified by polyethylene glycol phospholipid（PEG） or its derivative, the material will greatly improve stability and safety, and can prolong its half-life to achieve the effect of long-circulating preparation. Therefore, the iridium complex was prepared using lecithin and PEG to prepare a long-circulating liposome. In this way, not only the toxicity of the complex to the tumor cells is greatly improved, but also the characteristics of the long-circulating preparation can be utilized, and it is better applied to the vivo [2]. The result shows, Ir-1-Ir-3 complex did not significantly inhibit the growth of BEL-7402 cells（IC50:＞＞ 200）, but Ir-1-lipo, Ir-2-lipo and Ir-3-lipo showed Significant inhibition, IC50 values were 11.0 ± 0.5, 5.8 ± 0.2 and 28.9 ± 3.5, respectively.

Ir（Ⅲ） complexes with high quantum yields for triplet excited-state formation, long triplet lifetimes, and bright luminescence are potential theranostic photodynamic therapy（PDT） reagents. More importantly, the triplet excited-state characteristics of these complexes are readily tuned via alternation of the ligands However, most of the currently studied Ir（Ⅲ） complex photosensitizers mainly absorb in the UV and blue spectral region, which is not suitable for potential clinical applications. To red-shift the ground-state absorption and meanwhile to retain the desired triplet excited-state characteristics, we synthesized severa series of cationic or neutral Ir（Ⅲ） complexes that have strong absorption in the visible to the red spectra region and are emissive in the red/NIR region. Their photophysical properties and in vitro/in vivo theranostic PDT effects were systematically evaluated. Alternation of the different types of ligands dramatically impacted the natures and energies of the lowest singlet and triplet excited states of these complexes. Many of the complexes can efficiently generate singlet oxygen in solutions and other reactive oxygen species in vitro, and exhibited strong PDT effects upon visible or red light activation. One of the complexes showed the largest phototherapeutic index（＞1600） for the reported Ir（Ⅲ） complexes toward the SKMEL28 cells upon white light activation. These complexes also produce strong intracellular luminescence which highlights their potential as theranostic PDT agents.

The high incidence of cancer and high mortality force humans’ extreme desire for effective treatment of cancer.Both the iridium(Ⅲ) polypyridine compounds and cinnamaldehyde in nature have outstanding antitumor effects. To this end,we combined cinnamaldehyde with iridium(Ⅲ) polypyridine and explored the effect of unsaturated bonds on tumor activity.The complexes [Ir(ppy)(PHIP)](PF)(Ir-1),[Ir(ppy)(SHIP)](PF)(Ir-2) and [Ir(ppy)(PYHIP)](PF)(Ir-3) were characterized by ESI-MS,H NMR,C NMR.The toxicity test results indicated that complexes Ir-1,Ir-2 and Ir-3 can effectively inhibit the cell growth of SGC-7901 cells,and the measured IC values are 5.2 ± 0.4,5.8 ± 0.7 and 4.4 ± 0.1 μM,respectively.AO/EB staining and flow apoptosis confirmed that SGC-7901 cells were caused apoptosis after being treated with the complexes.Along with the increase of endogenous ROS and Ca levels,mitochondrial membrane potential collapse and massive release of cytochrome c,it is fully demonstrated that these complexes induce apoptosis through ROS-mediated mitochondrial pathway.Combined with the above results,it provides a favorable foundation for the future development of more effective anti-tumor drugs.
