Determination of ifenprodil by LC–MS/MS and its application to a pharmacokinetic study in healthy Chinese volunteers

This paper reports the development and validation of an assay for ifenprodil based on liquid chromatography–tandem mass spectrometry(LC–MS/MS)and its application to a pharmacokinetic study involving single and multiple intravenous infusions to healthy Chinese *** sample preparation of plasma by liquid–liquid extraction with ethyl acetate,the analyte and internal standard,urapidil,were separated by reversed phase chromatography in a run time of 4 min and detected by positive ion electrospray ionization followed by multiple reaction monitoring of the precursor-to-product ion transitions at m/z 326.2-308.1 for ifenprodil and m/z 388.4-205.3 for *** assay was linear in the concentration range 0.2–50.0 ng/mL with recovery 476.4%.In the pharmacokinetic study of single intravenous infusions of 5,10 and 15 mg ifenprodil,peak plasma concentrations and areas under the plasma concentration–time curve were both linearly related to *** the pharmacokinetic study of multiple once daily intravenous infusions of 10 mg ifenprodil for 7 days,pharmacokinetic parameters were similar to those after the single dose showing that ifenprodil does not accumulate on repeated administration.

Changes of cerebral blood flow in rats with acute cerebral ischemia and the effect of nitric oxide donor S-nitroso-N-acetyl-penicillamine

BACKGROUND ： Previous studies show that nitric oxide donor can increase cerebral blood flow and improve the function of neurons in cerebral ischemia, but the change does not happen in all the models of cerebral ischemia. OBJECTIVE ： To observe the effects of nitric oxide donor S-nitroso-N-acetyl-penicillamine （SNAP） on the cerebral blood flow, cyclic guanosine monophosphate （cGMP） content in cerebral cortex, infarct volume and blood pressure in acute ischemic rat brain.DESIGN ： A randomized and control animal experiment SEI-FING ： Department of Neurosurgery, Aerospace Central Hospital, Peking University.MATERIALS： Twenty-eight male Wistar rats of SPF grade, weighing 250-300 g, aged 10-12 weeks were randomly divided into control group （n =14） and SNAP-treated group （n =14）. SNAP （5 mg/bottle） was provided by Beijing Chemical Reagent Company. Laser Doppler Flowmeter （FLO C1; Omegawave Inc., Tokyo, Japan） and immunoassay kit （Amersham Pharmacia Biotech, UK） were applied. METHODS： ① Model establishment： In the control group, models of cerebral ischemia were induced by ligating right common, internal and external carotid arteries; In the SNAP-treated group, models of cerebral ischemia were induced by ligating right common and external carotid arteries, followed by occluding middle cerebral artery and ligating internal carotid artery. ②Administration： In the SNAP-treated group, SNAP （100 μg/kg） was intravenously infused within 2 minutes, whereas in the control group, phosphate buffered saline （PBS, 1 mL） was intravenously infused （0.5 mL per minute）. Six rats were used to measure the volume of cerebral infarction, and the other 8 rats were used to determine other indexes in each group respectively. ③ Determination of indexes： Regional cerebral blood flow （rCBF） was continuously measured by laser-Doppler flowmetry in the ischemic penumbra and contralateral cortex under the continuous monitoring of blood pressure, cGMP concentrations in brain tissue were determined

Prolonged infusion of Low Dose Gemcitabine in Advanced Pancreatic Carcinoma

Background: Gemcitabine was established as a monotherapy or in combination for locally advanced or metastatic pancreatic carcinoma. Aim: This study aimed to evaluate the efficacy of the low-dose gemcitabine over 6-hour infusion in patients with advanced pancreatic adenocarcinoma. Methods: 26 patients with locally advanced or metastatic pancreatic carcinoma were recruited into the study from December 2013 to October 2014. Patients received the treatment in Clinical Oncology Department, Sohag University, and Medical Oncology Department, Assiut University. Patients received low-dose gemcitabine (250 mg/m2) over 6-hour infusion, weekly for seven weeks and then on days 1 and 8 every 3 weeks till unacceptable toxicity or progression of the disease. Results: Twenty-six patients were enrolled in this study. After starting 7 weeks of treatment, the disease control rate was 38.5% in the form of complete response in 3.8% of patients, partial response in 26.9%, and stationary response in 7.7%. A Clinical study on the treatment of Cerebral Infarct with Naloxone

OBJECTIVE: Naloxone and complex Danshen were used to treat cerebral infart in order to compare two groups′ Clinical effectiveness. BACKGROUND:The treatment of cervral infart with Naloxone is reported more often in abroad than in domestic. METHODS:144 cases were divided into two group randomly group treated with Naloxone: 74 cases (male 39 cases,female 33 cases,) average age 64.5yr. control group:70 cases (male 37 cases, female 33 cases), average age 65yr. 0.8mgNaloxone which was soluted in 250ml 0.9% Natrii chloride was used in the trcatment group by intravenous infusion, Qd, and the treatment lasted 14 days, On before and 4W and 8W both aftcr treatment, clotting time and blood flow dynamics were investigated. According to NDS and DLA standards, clinical effectiveness was compared and was studied by t test statistic method. RESULTS:Thc total response rate is 93.2% in Naloxonegroup, and 77.1% in control group (P＜0. 05), The differencebetween two groups is statistically significant.
