Effect of Gubi prescription on caveolin-1 expression and phosphoinositide 3 kinase/protein kinase B and Fas signal pathways in rats with knee osteoarthritis

OBJECTIVE: To investigate the effects of Gubi prescription on the expression of caveolin-1, and the phosphoinositide 3 kinase/protein kinase B(PI3 K/Akt) and Fas signal pathways in rats with knee osteoarthritis(KOA).METHODS: Forty KOA model rats were established using a modification of Hulth's method. Rats were divided into five groups by the random number method: model, positive drug(Vicolli group), and high-, medium-, and low-dose Gubi prescription groups(n = 8/group). In the sham surgery group(n = 8), only anterior and posterior cruciate ligaments of rats were exposed during surgery. A normal group(n = 8) consisted of rats with no treatment. Rats were intragastrically administered corresponding drugs once every day for eight consecutive weeks. Then, rat synovial membranes were extracted and histomorphological changes were recorded. m RNA expression was measured by *** superoxide dismutase(SOD), malondialdehyde(MDA), nitric oxide(NO), and interleukin 1β(IL-1β) levels were measured. Western blotting determined the effects of Gubi prescription on protein expressions of caveolin-1, Bax, Bcl-2, Fas, and caspase-3 in chondrocytes from KOA rats. The knee cartilage of rats was excised and cultured under aseptic conditions. After coincubation of chondrocytes with Gubi prescription-containing serum, IL-1β, and si RNA, Western blotting was used to determine the protein expressions of caveolin-1, Bax, Bcl-2, Fas, and ***: The morphological score of the articular synovium in the model group was significantly higher than in the normal group(P 1). The morphological score in the high-and mediumdose Gubi prescription groups was lower than in the model group(P < 0.05). Chondrocytes from the decoction-containing serum group had a lower expression of Bax(P < 0.05), and higher expressions of Bcl-2(P < 0.05) and caspase-3(P < 0.05) compared with the model group. OBJECTIVE:To evaluate the protective effect of Shouzhangshen(Rhizoma Gymnadeniae Crassinervidis)extract against acute high altitude hypoxia-induced brain injury in ***:Sixty C57BL/6J mice were selected and assigned to six groups(n=10):normal control group,low-pressure hypoxia group,positive control group(dexamethasone 500 mg/kg),and three groups treated with Shouzhangshen extract(250,500,and 750 mg/kg,respectively).The Morris water maze test was performed to evaluate alterations in spatial learning and memory *** staining was performed to detect Nissl bodies and neuron ***-inducible factor(HIF)-1α,interleukin(IL)-1β,tumor necrosis factor(TNF)-α,vascular endothelial growth factor(VEGF),and malondialdehyde(MDA)expression in brain tissue and serum,as well as superoxide dismutase(SOD)and glutathione(GSH)activity in brain tissues were measured by enzyme-linked immunosorbent assays,quantitative real-time-polymerase chain reaction and western ***:The Morris water maze test results showed that Shouzhangshen extract can significantly reduce the latency and swimming distance to escape onto a visible platform,increase neuron density and hierarchy and the number of pyramidal neurons,and decrease the expression of HIF-1α,IL-1β,TNF-α,and VEGF mRNAs and proteins in both brain tissue and serum(P0.05).CONCLUSION:Our findings suggest that Shouzhangshen extract may have a significant effect on acute high altitude hypoxia-induced brain injury in mice.

Effect of Rongchang capsule (茸菖胶囊) on seizure behavior,cognitive impairment,and hippocampal DNA damage and inflammatory factors in epileptic rats

OBJECTIVE: To observe the behavioral changes and changes in DNA fragments and related inflammatory factors in the hippocampus of epileptic rats pretreated with Rongchang capsule(茸菖胶囊).METHODS: Eighty Sprague-Dawley rats were randomly divided into the normal group(NG), model group(MG), sodium valproate group(VG), and Rongchang capsule group(RG)(n = 20 in each group). Pentylentetrazol was administered to the MG, VG, and RG to induce epilepsy. The VG and RG were pretreated with 1/2 the therapeutic dose of sodium valproate and Rongchang capsule, respectively. Changes in convulsion behavior and water maze learning were observed. Single cell gel electrophoresis was used to detect changes in the DNA in the hippocampus. The tail length(TL) and Olive tail moment(OTM) of cells were analyzed by GASP software. The expression of interleukin-1β(IL-1β),high mobility group box 1(HMGB1), transforming growth factor-β(TGF-β), and CCL4 in the hippocampus was determined by Western ***: Rongchang capsule had a weaker effect on convulsive latency than sodium valproate, but significantly reduced seizure susceptibility. The spatial learning ability of the RG was better than that of the VG(P ≤ 0.01). The TL and OTM were significantly higher in the MG than the NG(P 1). The RG had a better TL and OTM than the VG(P 1).Combined with the microscopy results, DNA damage was most pronounced in the MG. Drug intervention decreased the DNA damage in the VG and RG. The expressions of IL-1β, CCL4, and HMGB1 in the hippocampus were significantly greater in the MG than the NG(P 1), and were significantly reduced in the RG and VG compared with the MG(P 1);however, there was no intergroup difference in the expression of TGF-β. The average values for the expression of inflammatory factors in the hippocampus were higher in the RG than in the VG;thus, Rongchang capsule may have a weaker effect on reducing the expression of inflammatory factors in the hippocampus than sodium valp

Effect of moxibustion plus Duhuo Jisheng decoction on hs-CRP,IL-1β and TNF-α levels in middle-aged and elderly patients with knee osteoarthritis

Objective： To observe the clinical effect of moxibustion combined with Duhuo Jisheng decoction for middle-aged and elderly patients with knee osteoarthritis（KOA） and its impact on serum high sensitive C-reactive protein（hs-CRP）,interleukin-1β（IL-1β） and tumor necrosis factor-α（TNF-α） ***： A total of 90 eligible KOA patients were randomized into an observation group（n=45） and a control group（n=45）.Cases in the observation group received moxibustion plus oral administration Duhuo Jisheng decoction, while cases in the control group received Duhuo Jisheng decoction. Then the hs-CRP, IL-1β and TNF-α levels were examined and the clinical effect was ***： Before treatment, the hs-CRP, IL-1β and TNF-α levels showed no between-group statistical differences（all P〉0.05）. After 8 weeks of treatment, the hs-CRP, IL-1β and TNF-α levels all dropped significantly in both groups（P〈0.01 or P〈0.05）, and the treatment group showed more substantial changes than the control group（all P〈0.05）. The total effective rate was 97.8% in the treatment group, versus 86.7% in the control group, showing a statistical difference（P〈0.05）.Conclusion： Moxibustion plus Duhuo Jisheng decoction has a good clinical effect for middle-aged and elderly KOA patients,and the effect may relate to the decreased hs-CRP, IL-1β and TNF-α levels.

Background S100B is involved in brain *** objective of this study was to determine plasma and cerebral spinal fluid(CSF) levels of S100B in patients with spontaneous intracerebral hemorrhage (ICH),and to correlate S100B levels with Glasgow Coma Scale(GCS) scores,ICH volumes, presence of intraventricular hemorrhage(IVH),and survival rate,and to correlate CSF S100B levels with plasma S100B levels as well as CSF interleukin-lbeta(IL-1β) *** Ten patients with suspicion of subarachnoid hemorrhage and thirty-eight patients with spontaneous basal ganglia hemorrhage were *** and CSF samples were *** concentrations of IL-1 6 in CSF and S100B in plasma and CSF were analyzed by enzyme-linked immunosorbent *** Plasma or CSF S100B levels in the ICH group were significantly higher than those in the control group(178.7±74.2 versus 63.2±23.0 pg/mL;P0.05).Conclusions High levels of S100B are present in the cerebrospinal fluid and peripheral blood of patients with ICH and may contribute to the inflammatory processes of *** levels of CSF and plasma S100B after spontaneous onset of ICH seem to correlate with clinical outcome in these *** in peripheral S100B properly reflect brain injury,and plasma S100B levels can be served as useful clinical markers for evaluating the prognosis of ICH.
