Short-term impacts of air pollution on the platelet-lymphocyte ratio and neutrophil-lymphocyte ratio among urban adults in China

The short-term impacts of urban air pollution on the platelet-lymphocyte ratio (PLR) and neutrophil-lymphocyte ratio (NLR) remain obscure.In this study,we included 3487 urban adults from the Wuhan-Zhuhai cohort.Individual inhalation exposure to air pollutants was estimated by combining participants’daily breath volume and ambient concentrations of six air pollutants (includingfine particulate matter (PM_(2.5)),inhalable particulate matter(PM_(10)),nitrogen dioxide (NO_(2)),sulfur dioxide (SO_(2)),carbon monoxide (CO) and ozone (O_(3))).The cumulative impacts were assessed by applying lag structures of up to 7 days before the survey date.Associations of air pollutants with PLR and NLR were assessed using a linear mixed model and Bayesian kernel machine regression (BKMR) model.We found that PLR was negatively related to PM_(2.5)(lag02-lag06),PM_(10)(lag02-lag07),NO_(2)(lag02-lag07),and SO_(2)(lag03-lag05) and NLR was negatively related to PM_(10)(lag05 and lag07).In the BKMR model,a negative joint association between the six-air-pollutant mixture and PLR and NLR was observed,whereas PM_(10)and NO_(2)appeared to be more important than the other pollutants in the mixture.The negative impact of air pollutants was stronger in males,participants with lower body mass index (<24 kg/m^(2)),those cooking meals at home,drinkers,and nonexercisers.In conclusion,short-term exposure to air pollutants is significantly related to PLR and NLR in peripheral blood.PLR and NLR may provide new insight into the molecular mechanism underlying the adverse health impact of air pollutants.

Yixin Ningshen Tablet Alleviates Comorbidity of Myocardial Infarction and Depression by Enhancing Myocardial Energy Metabolism and Increasing Availability of Monoamine Neurotransmitter

Objective: To investigate the therapeutic effect of Yixin Ningshen Tablet(YXNS) on comorbidity of myocardial infarction(MI) and depression in rats and explore the underlying mechanism. Methods: The Sprague-Dawley rats were randomly divided into 5 groups with 7 rats in each group according to their weights,including control, model, fluoxetine(FLXT, 10 mg/kg), low-dose YXNS(LYXNS, 100 mg/kg), and high-dose YXNS(HYXNS, 300 mg/kg) groups. All rats were pretreated with corresponding drugs for 12 weeks. The rat model of MI and depression was constructed by ligation of left anterior descending coronary artery and chronic mild stress stimulation. The echocardiography, sucrose preference test, open field test, and forced swim test were performed. Myocardial infarction(MI) area and myocardial apoptosis was also detected. Serum levels of interleukin(IL)-6, IL-1β, tumor necrosis factor-α(TNF-α), 5-hydroxytryptamine(5-HT), adrenocorticotrophic hormone(ACTH), corticosterone(CORT), and norepinephrine(NE) were determined by enzyme linked immunosorbent assay. The proteins of adenosine 5’-monophosphate-activated protein kinase(AMPK), p-AMPK,peroxisome proliferator-activated receptor gamma coactivator-1α(PGC-1α), and nuclear respiratory factor 1(NRF1) in heart were detected by Western blot analysis. The expression levels of TNF-α, IL-6, indoleamine 2,3-dioxygenase(IDO1), kynurenine 3-monooxygenase(KMO), and kynureninase(KYNU) in hippocampus were detected by real-time quantitative polymerase chain reaction. Results: Compared with the model group,the cardiac function of rats treated with YXNS significantly improved(P<0.01). Meanwhile, YXNS effectively reduced MI size and cardiomyocytes apoptosis of rats(P<0.01 or P<0.05), promoted AMPK phosphorylation,and increased PGC-1α protein expression(P<0.01 or P<0.05). HYXNS significantly increased locomotor activity of rats, decreased the levels of TNF-α, IL-6 and IL-1β, and increased the serum levels of 5-HT, NE, ACTH,and CORT(all P<0.05). Previous studies have shown that vagus nerve stimulation can improve patients' locomotor function.The stimulation of the auricular vagus nerve,which is the only superficial branch of the vagus nerve,may have similar effects to vagus nerve stimulation.However,the precise mechanisms remain poorly understood.In this study,rat models of cerebral ischemia/reperfusion injury were established by modified Longa ligation.Twenty-four hours later,7-day auricular vagus nerve stimulation was performed.The results showed that auricular vagus nerve stimulation promoted the secretion of acetylcholine,inhibited the secretion of interleukin-1β,interleukin-6,and tumor necrosis factor-α,and reduced connexin 43 phosphorylation in the ischemic penumbra and motor cortex,promoting locomotor function recovery in rats with cerebral ischemia/reperfusion injury.These findings suggested that auricular vagus nerve stimulation promotes the recovery of locomotor function in rats with cerebral ischemia/reperfusion injury by altering the secretion of acetylcholine and inflammatory factors and the phosphorylation of connexin 43.This study was approved by the Animal Use and Management Committee of Shanghai University of Traditional Chinese Medicine on November 8,2019(approval No.PZSHUTCM191108014).

Piperine treating sciatica through regulating inflammation and MiR-520a/P65 pathway

Although the etiology of sciatica remains uncertain,there is increasing evidence that the disease process of sciatica is associated with the levels of inflammatory factors.Piperine,an alkaloid isolated from Piper nigrum,has previously been demonstrated to inhibit inflammation and analgesic effects.The purpose of this study is to verify the regulatory relationship between miR-520a and p65 and to explore how miR-520a/P65 affects the level of cytokines under the action of piperine,so as to play a therapeutic role in sciatica.Through ELISA experiment,we confirmed that four inflammatory factors(IL-1β,TNF-α,IL-10,TGF-β1)can be used as evaluation indexes of sciatica.The differentially expressed miRNA was screened as miR-520a,by microarray technology,and the downstream target of miR-520a was P65 by bioinformatics.Real-time fluorescence quantitative PCR confirmed that the expression of miR-520a was negatively correlated with pro-inflammatory cytokines,positively correlated with anti-inflammatory cytokines and negatively correlated with p65 expression at mRNA level.The expression of p65 was positively correlated with pro-inflammatory cytokines and negatively correlated with anti-inflammatory cytokines at the protein level verified by ELISA and Western blot.HE staining analysis showed that the nerve fibers were repaired by piprine,the vacuoles were significantly reduced,and the degree of nerve fiber damage was also improved.Immunohistochemical analysis showed that the expression of p65 decreased after administration of piperine.Dual-luciferase reporter gene assay confirmed that the luciferase signal decreased significantly after cotransfection of miR-520a mimics and p653'UTR recombinant plasmid.To sum up,in the rat model of non-compressed lumbar disc herniation,piperine plays a significant role in analgesia.MiR-520a can specifically and directly target P65,and piperine can promote the expression of miR-520a,then inhibit the expression of p65,down-regulate the pro-inflammatory factors IL-1βand TNF-α,and up-regulate the effec

Effect of temperature maintenance by forced-air warming blankets of different temperatures on changes in inflammatory factors in children undergoing congenital hip dislocation surgery

Background:Hypothermia is associated with many adverse clinical outcomes in pediatric patients,and thus,it is important to an effective and safe method for preventing peri-operative hypothermia and its associated adverse outcomes in pediatric patients.This study aimed to investigate the effect of forced-air warming blankets with different temperatures on changes in the transforming growth factor-β(TGF-β),tumor necrosis factor(TNF)-α,interleukin(IL)-1β,and IL-10 levels in children undergoing surgical treatment for developmental displacement of the hip(DDH).Methods:The study included 123 children undergoing surgery for DDH under general anesthesia.The patients were randomly assigned to three groups,using a random number table:the 32,38,and 43°C groups according to the temperature setting of the forced-air warming blankets.For each patient,body temperature was recorded immediately after anesthesia induction and intubation(T0),at initial incision(T1),at 1 h after incision(T2),at2 h after incision(T3),at the end of surgery(T4),immediately upon return to the ward after surgery(T5),and then at 12 h(T6),24 h(T7),36 h(T8),and 48 h(T9)after the surgery.The serum levels of TGF-β,TNF-α,IL-1β,and IL-10 were measured at To and T4 for all groups.Results:The number of patients with fever in the 38°C group was significantly less than those in the 32 and 43°C groups(x^2=6.630,P=0.036).At To,the body temperatures in the 38 and 43°C groups were significantly higher than that in the 32°C group(F=17.992,P<0.001).At T2,the body temperature was significantly higher in the 43°C group than those in the 32 and 38°C groups(F=12.776,P<0.001).Moreover,at T4,the serum levels of TGF-β(F=3286.548,P<0.001)and IL-10(F=4628.983,P<0.001)were significantly increased in the 38°C group,and the serum levels of TNF-α(F=911.415,P<0.001)and IL-10(F=322.191,P<0.001)were significantly decreased in the 38°C group,compared with the levels in the 32 and 43°C groups.Conclusion:Force-air warming blankets set at 38°C maintained stable body temperature with less ad

Effects of dexmedetomidine on the expression of inflammatory factors in children with congenital heart disease undergoing intraoperative cardiopulmonary bypass:A randomized controlled trial

Importance:Dexmedetomidine inhibits the inflammatory response associated with cardiopulmonary bypass(CPB)and protects neural function.However,the mechanism of dexmedetomidine’s anti-inflammatory pathway is unclear.Objective:To investigate the effect of dexmedetomidine on the cognitive level and expression of inflammatory factors in children with congenital heart disease undergoing intraoperative CPB.Methods:Ninety children with congenital heart disease were recruited and randomly divided into 3 groups of 30 children in each.In Group 1,a 1.0μg·kg-1·h-1 intravenous bolus of dexmedetomidine was administered 10 minutes after induction of anesthesia,followed by a 0.2μg·kg-1·h-1 infusion until the surgical incision.In Group 2,a 0.5μg/kg intravenous bolus of dexmedetomidine was administered 10 minutes after induction of anesthesia,followed by a 0.1μg·kg-1·h-1 infusion until the surgical incision.The control group was given physiological saline using the same method as in Groups 1 and 2.The serum levels of nuclear factor-kappa B(NF-κB),S-100βprotein,neuron-specific enolase(NSE),tumor necrosis factor-α(TNF-α),and interleukin-6(IL-6)were measured before the surgery(T1),at the end of CPB(T2),2 hours after CPB(T3),6 hours after CPB(T4),and 24 hours after CPB(T5).The Wechsler Intelligence Scale for children(WISC)was measured before the operation and at 3,6,and 12 months after the operation to evaluate the neurodevelopmental state of the children.Results:The levels of the NF-κB,S-100βprotein,NSE,TNF-α,IL-6 were significantly higher at T2,T3,or T4 than before the surgery(T1)in the control group or the dexmedetomidine groups.However,the increases of NF-κB,TNF-α,IL-6,S-100βand NSE levels were significantly smaller in the dexmedetomidine groups than those in the control group(Pinflammatory reaction and the increase S-100βprotein and NSE levels indicated t

Effect of Acupuncture on Ovary Morphology and Function in DHEA-Induced Polycystic Ovary Syndrome Model Rats

Objective To investigate the effects of acupuncture on ovary morphology and function in dehydroepiandrosterone(DHEA)-induced polycystic ovary syndrome(PCOS)model rats.Methods A total of 40 adult female Wistar rats were randomly allocated to 4 groups by a random number table,including control,model,metformin and acupuncture groups,10 rats in each group.PCOS rat model was developed by injecting with DHEA(6 mg/100 g body weight)in 0.2 mL of oil subcutaneously.Electrical stimulation(2 Hz,3 mA)was applied to Guanyuan(CV 4),Zigong(EX-CA1)and Qihai(CV 6)acupoints for 30 min daily in the acupuncture group,and metformin(200 mg/kg)was given to rats in the metformin group,both once per day for 21 consecutive days,and rats in the normal group was fed with normal saline and fed regularly.After 21 days of administration,the rat blood samples were collected for detecting the reproductive hormonal levels[luteinizing hormone(LH),follicle stimulating hormone(FSH),estradiol(E2),progesterone(P),testosterone(T)]and inflammatory factors(visfatin,IL-6)analysis.Ovary tissue was used for histopathological analysis.Results Compared with the model group,rats in the acupuncture and metformin groups were significantly lower in weight gain,FSH,LH and T levels,and E2 and P levels significantly increased(alll P0.05).Ovarian diameter in the acupuncture and metformin groups were smaller than the model group(P0.05).Conclusion Acupuncture might improve ovary morphology and its function in DHEA-induced PCOS model rat

Effect of early restrictive fluid resuscitation on inflammatory and immune factors in patients with severe pelvic fracture

Purpose:To study the effect of early restrictive fluid resuscitation(EFR)on inflammatory and immune factors in patients with severe pelvic fracture(SPF).Methods:A total of 174 SPF patients in the Department of Orthopaedics,the First Affiliated Hospital of Chengdu Medical College from July 2015 to June 2018 were involved in this study and divided into EFR group(n=87)and control group(n=87)using the random number table method.Conventional fluid resuscitation(CFR)was performed in control group,and EFR was performed in EFR group.The incidences of acute respiratory distress syndrome(ARDS)and multiple organ dysfunction syndrome(MODS)during rescue,successful rescue rate,blood transfusion volume,fluid input,and resuscitation time were compared between the two groups.The parameters including prothrombin time(PT),hematocrit(HCT),platelet(PLT)and blood lactate(BL)at the 4th hour after fluid resuscitation were recorded.The levels of inflammatory factors(TNF-α,IL-6,CRP)and immune factors(CD3^+,CD4^+,CD8^+,CD4^+/CD8^+)were compared between the two groups before treatment and 7 days after treatment.The revised acute physiologic and chronic health evaluation system and the sequential organ failure assessment scores were adopted for evaluation before treatment and 7 days after treatment.Results:The incidences of ARDS and MODS during rescue in EFR group were significantly lower than those in control group(p=0.015 and 0.010 respectively),and the successful rescue rate in EFR group was significantly higher than that in control group(p=0.011).The blood transfusion volume,fluid input,resuscitation time in EFR group were significantly lower than those in control group(p=0.016,0.002 and 0.001 respectively).At the 4th hour after fluid resuscitation,PT and BL in EFR group were significantly lower than those in control group(p=0.021 and 0.003 respectively),while HCT and PLT in EFR group were significantly higher than those in control group(p=0.016 and 0.021 respectively).On day 7 after treatment,TNF-α,IL-6,CRP and CD8^+in EFR group were si

Advantages of nanocarriers for basic research in the field of traumatic brain injury

A major challenge for the efficient treatment of traumatic brain injury is the need for therapeutic molecules to cross the blood-brain barrier to enter and accumulate in brain tissue.To overcome this problem,researchers have begun to focus on nanocarriers and other brain-targeting drug delivery systems.In this review,we summarize the epidemiology,basic pathophysiology,current clinical treatment,the establishment of models,and the evaluation indicators that are commonly used for traumatic brain injury.We also report the current status of traumatic brain injury when treated with nanocarriers such as liposomes and vesicles.Nanocarriers can overcome a variety of key biological barriers,improve drug bioavailability,increase intracellular penetration and retention time,achieve drug enrichment,control drug release,and achieve brain-targeting drug delivery.However,the application of nanocarriers remains in the basic research stage and has yet to be fully translated to the clinic.

Single Wall Carbon Nanotube Induced Inflammation in Cruor-Fibrinolysis System

Objective To study single wall carbon nanotubes （SWCNT） and its role in inducing inflammatory cytokines in the cruor-fibrinolysis system of rat. Methods Twenty one Wistar rats were divided into four groups： 1） control; 2） low-dose SWCNT （0.15 mg/kg BW）; 3） medium-dose SWCNT （0.75 mg/kg BW）; 4） high-dose SWCNT （1.5 mg/kg BW）. Intratracheal instillation of SWCNT suspensions was administered to rats once per day for 21 days. In order to assess the exposure effect of SWCNT to the rats, activity of inflammatory cytokine was measured and markers of cruor-fibrinolysis system were studied via ELSIA. Also, change in clotting time was recorded and histopathology was studied. Results IL-6 and IL-8 concentrations of rats exposed to SWCNT were significantly higher than those in controls （P〈0.05）. The activity of inflammatory cytokines and histopathological change indicated that oxidative damage occurred. Change in clotting time in rats exposed to SWCNT decreased compared with controls. Meanwhile, t-PA （tissue-tupe plassminogen activator） and AT-Ⅲ （antithrombin-Ⅲ） levels in rats exposed to particulates increased or decreased significantly compared with controls （P〈0.05）. A similar trend was observed for D-dimer （D2D） levels, indicating that SWCNT can impact the cruor-fibrinolysis system ofrat. Conclusion The results from our study suggest that an increased procoagulant activity and reduced fibrinolytic activity in rats exposed to SWCNT can cause pulmonary oxidative stress and inflammation, due to the release of pro-thrombotic and inflammatory cytokines into the blood circulation of rat.
