Advancing toxicity Predictions:A Review on in Vitro to in vivo Extrapolation in Next-Generation Risk Assessment

As a key step in next-generation risk assessment(NGRA),in vitro to in vivo extrapolation(IVIVE)aims to mobilize a mechanism-based understanding of toxicology to translate bioactive chemical concentrations obtained from in vitro assays to corresponding exposures likely to induce bioactivity in *** conversion can be achieved via physiologically-based toxicokinetic(PBTK)models and machine learning(ML)*** last 5 years have witnessed a period of rapid development in IVIVE,with the number of IVIVE-related publications increasing *** Review aims to(1)provide a comprehensive overview of the origin of IVIVE and initiatives undertaken by multiple national agencies to promote its development;(2)compile and sort out IVIVE-related publications and perform a clustering analysis of their high-frequency keywords to capture key research hotspots;(3)comprehensively review PBTK and ML model-based IVIVE studies published in the last 5 years to understand the research directions and methodology developments;and(4)propose future perspectives for IVIVE from two aspects:expanding the scope of application and integrating new *** former includes focusing on metabolite toxicity,conducting IVIVE studies on susceptible populations,advancing ML-based quantitative IVIVE models,and extending research to ecological *** latter includes combining systems biology,multiomics,and adverse outcome networks with IVIVE,aiming at a more microscopic,mechanistic,and comprehensive toxicity *** Review highlights the important value of IVIVE in NGRA,with the goal of providing confidence for its routine use in chemical prioritization,hazard assessment,and regulatory decision making.

Nefang is a polyherbal product composed of Mangifera indica（bark and leaf）,Psidium guajava,Carica papaya,Cymbopogon citratus,Citrus sinensis and Ocimum gratissimum（leaves）.It is often used in Cameroon traditional medicine as an *** studies have demonstrated its in vitro antiplasmodial activity,synergistic interactions between its constituents[1],antioxidant properties[2]and in vivo antiplasmodial efficacy（unpublished）.This study aimed at evaluating its cytotoxicity and in vivo *** screening was evaluated on Hep G2 hepatoma and U20 S osteosarcoma epithelial cell lines[3,4].Systemic acute oral toxicity was assessed in Swiss albino mice,while repeated dose（28 and 90 days） toxicity were studied in wistar rats at doses of 250,500 and 1000 mgkg-1 body weight[5].Physiological,biochemical and hematological parameters were analysed to determine its effects on the liver,kidney,lipid profile,nitrogen balance,glycemic levels and the hematological *** analysed using the one way ANOVA followed by Neuman-Keuls multiple comparison *** showed no significant cytotoxic *** and repeated dose oral toxicity produced no observed adverse effects at a dose ＞5 g/kg as well as on the liver,kidney and lipid profiles ***-acute administration of Nefang significantly（p＜0.05） lowered the GLU levels more than *** also observed a dose-dependent significant（p＜0.05） increase in white and red blood cells count,hemoglobin,hematocrit and platelet *** findings suggest that Nefang is relatively safe when administered orally after short or prolonged periods,at sub-therapeutic and therapeutic *** these doses,it exhibited potentials at maintaining the integral functioning of the liver,kidney and boosting the immune *** also showed good potentials at maintaining a good nitrogen balance and hypoglycemic *** study demonstrated the safety of Nefang,thus upholding knowledge on its folk use and suggest

Two new cyclometalated Ru(Ⅱ) complexes containing isoquinoline ligand,[Ru(bpy)(1-Ph-IQ)](PF),(bpy=2,2’-bipyridine;1-Ph-IQ=1-phenylisoquinoline;Ru1) and [Ru(phen)(1-Ph-IQ)](PF)(phen=1,10-phenanthroline;Ru2) have been synthesized and ***,both of them exhibited IC values that were much lower than those of cisplatin and other structurally similar non-cyclometalated Ru(Ⅱ) complexes [1],which is consistent with those of Chao et al [2].Further studies revealed that the two complexes can cause the loss of mitochondrial membrane potential(MMP),increase the generation of reactive oxygen species(ROS),target nucleus,trigger DNA damage,and induce ***,Ru2 shows low toxicity to zebrafish *** current work suggests that the synthetic complexes may be developed as low-toxicity agents for lung cancer therapy.
