Screening　and　identification　of　down- regulated　genes　in　colorectal　carcinoma　by　subtractive　hybridization: a　method　to　identify　putative　tumor　suppressor　genes

Objective To search for new putative tumor suppressor genes in colorectal carcinoma. Methods Subtractive hybridization technologies were applied to screen and select genes, the expression of which was down regulated in colorectal carcinoma. mRNAs uniquely expressed in normal cells but not in colorectal carcinoma were recovered as cDNA (sub cDNA) after two rounds of subtractive hybridization with mRNA prepared from colorectal carcinoma. The sub cDNAs were then used as probes to screen a normal human colon cDNA library constructed in lambda Zap II phage. The DNAs of positive clones were in vivo excised, and partial DNA sequences were analyzed and compared with DNA sequence database Genbank. Results A total of 46 different clones with an average of about 1 kilobases in transcript size was recovered. Among these 46 down regulated genes in colorectal carcinoma were genes encoding immunoglobulin (n=32), 40 kDa keratin intermediate filamentous protein or IFP (n=1), major histocompatibility complex related protein (n=1), unrelated structural proteins (n=10) and gene products yet to be identified (n=2). RNA dot blot hybridizations confirmed that all 46 clones contained genes that were down regulated and have not been reported before in colorectal carcinoma. Conclusions The results of this study suggested that the 46 clones were down regulated in colorectal carcinoma,they should be further studied as new putative tumor suppressor genes and could be used as new tumor markers of colorectal carcinoma.

The COⅠgene in six indigenous chicken breeds was amplified using PCR and *** sequences were analyzed to explore utility as DNA Barcoding for identif- ying these chicken *** study revealed that there were 24 polymophic sites and 24 haplotypes,22 of them were special to 6 chicken *** 2-parameter distance be- tween the 6 breeds are 0.00181～*** special sites were found in each chicken breed which was the bases to identify chicken *** taxonomy is consistent with morphological taxonomy among the six chicken *** phylogenetic relationships of the breeds can be well resolved by COⅠgene *** experiment provides the pri- mary information for using COⅠgene to identify indigenous chicken breeds.

OBJECTIVE To establish a method for primary cultured and iden-tified Sprague-Dawley(SD) rats cerebral cortical astrocytes. METHODS Cerebral co-rtex of SD neonatal rats within 24 h was taken with stereo microscope and was cut topieces(1 mm^3),digested by Accutase and 0.1% DNAase(37℃,15 min),anddispersed cell suspension was made by mechanical method and filtered. The fibroblast cells and microglia were removed through differential adhesion and sha-ke. Passaged cells were identified by immunofluorescent with anti-Glial fibrillaryacidic protein(GFAP) antibody. RESULTS The astrocytes of rats cerebral cortex were cultured in this method,which had a large number of cells,good activity,high purity,abundant and elongated cell processes,and were interwoven into a network,showing a typical and good growth state. The third generation of the cel s comprised >95% astrocytes. CONCLUSION This simple and reliable cultivation method of astrocyte from rat cerebral tissue is established with high purity,and in a good growth condition.

Very recently, the Belle and BESIII experiments observed a new charmonium-like state X(3823), which is a good candidate for the D-wave charmonium (13D2). Because the X(3823) is just near the DˉD threshold, the decay X(3823) ! J= +?? can be a golden channel to test the significance of coupled-channel effects. In this work[1], this decay is considered including both the hidden-charm dipion and the usual quantum chromodynamics multipole expansion (QCDME) contributions.

To explore biodegradation of 2-naphthol and its metabolites accumulated in wastewater treatment,a series of bio-degradation experiments were *** main metabolites of 2-naphthol,1,2-naphthalene-diol and 1,2-naphthoquinone,were identified by high-performance liquid chromatography with *** fungus Aspergillus niger with bacterium Bacillus subtilis in the treatment enhanced 2-naphthol degradation efficiency,lowered the accumulation of the two toxic *** were two main phases during the degradation process by the kinetic analysis：2-naphthol was first partly degraded by the fungus,producing labile and easily accumulated metabolites,and then the metabolites were mainly degraded by the bacterium,attested by the degradation processes of 1,2-naphthalene-diol and 1,2-naphthoquinone as sole source of carbon and *** succinate,as a co-metabolic substrate,was the most suitable compound for the continuous *** optimum concentration of 2-naphthol was 50 mg/*** overall 2-naphthol degradation rate was 92%,and the COD Cr removal rate was 80% on day *** results indicated that high degradation rate of 2-naphthol should not be considered as the sole desirable criterion for the bioremediation of 2-naphtholcontaminated soils/wastewater.
