Effects of fish protein hydrolysate on growth performance and humoral immune response in large yellow croaker (Pseudosciaena crocea R.)

We investigated the effects of fish protein hydrolysate (FPH) on growth performance and humoral immune response of the large yellow croaker (Pseudosciaena crocea R.). One thousand and two hundred large yellow croakers [initial average weight: (162.75±23.85) g] were divided into four groups and reared in floating sea cages (3 m×3 m×3 m). The animals were fed with 4 diets: basal diet only (control) or diets supplemented with 5%, 10% and 15% (w/w) FPH. The results show that dietary FPH levels significantly influenced the growth and immunity of the large yellow croaker. Compared with the control group, total weight gain (TWG) in all treatment groups, relative weight gain (RWG) and specific growth rate (SGR) in fish fed with diets supplemented with 10% and 15% FPH were significantly increased (Pimmune parameters [lysozyme activity, serum complements, immunoglobulin M (IgM)]. Lysozyme activity, complement C4 and IgM were also significantly increased (P<0.05) in fish fed with diets supplemented with 10% and 15% FPH, while complement C3 level was significantly increased (P<0.05) in all treatment groups. In general, with the supplementation of FPH, particularly at dose of 10%, the growth performance and immunity of the large yellow croaker can be improved effectively.

With the increasing immunological studies on camels due to the advantage of their single-chain antibodies for humanizations,it is demanding to develop an easy-to-handle evaluation method of their humoral immune response before proceeding with immunization of foreign antigens that may be toxic to *** this study,we quantitatively determined the expression levels of T-helper 2(Th2) cytokines in peripheral blood lymphocytes obtained from Bactrian camels by real-time *** recorded kinetic profiles resulting from the immunization of ovalbumin(OVA) indicated that after immunization,Th2 cytokines including interleukin(IL) families such as IL-4,IL-10,and IL-13 in the camels were up-regulated by a factor of 1.78,3.15,and 1.22,respectively,which was validated by traditional enzyme-linked immunosorbent assay(ELISA) *** ELISA which requires specific enzyme-labeled antibodies,this established method based on the minimal amount of blood samples holds an advantage in the preliminary evaluation of camel humoral immune response with desirable precision,which is meaningful for biomedical explorations of camel-derived antibodies.

A general nonlinear mathematical model for the viral infection with humoral immunity and two distributed delays is proposed and analyzed. Two bifurcation parameters, the basic reproduction number, R 0 and the humoral immunity number, R 1 are derived. We established a set of conditions on the general functions which are sufficient to determine the global dynamics of the model. Utilizing Lyapunov functions and LaSalle’s invariance principle, the global asymptotic stability of all equilibria of the model is obtained. An example is presented and some numerical simulations are conducted in order to illustrate the dynamical behavior.

The H9N2 subtype avian influenza virus(AIV)inactivated vaccine has been used extensively in poultry farms,but it often fails to stimulate a sufficiently high immune response in poultry in the field,although it works well in laboratory experiments;hence,the virus still causes economic damage every year and poses a potential threat to public *** on surveillance data collected in the field,we found that broilers with high levels of maternal-derived antibodies(MDAs)against H9N2 virus did not produce high levels of antibodies after vaccination with a commercial H9N2 inactivated *** contrast,specific pathogen-free(SPF)chickens without MDAs responded efficiently to that *** MDAs were mimicked by administering passively transferred antibodies(PTAs)into SPF chickens in the laboratory,similar results were observed:H9N2-specific PTAs inhibited humoral immunity against the H9N2 inactivated vaccine,suggesting that H9N2-specific MDAs might hinder the generation of antibodies when H9N2 inactivated vaccine was *** challenge with homologous H9N2 virus,the virus was detected in oropharyngeal swabs of the vaccinated and unvaccinated chickens with PTAs but not in the vaccinated chickens without PTAs,indicating that H9N2-specific MDAs were indeed one of the reasons for H9N2 inactivated vaccine failure in the *** different titers of PTAs were used to mimic MDAs in SPF chickens,high(HI=12 log2)and medium(HI=log 9 log2)titers of PTAs reduced the generation of H9N2-specific antibodies after the first vaccination,but a booster dose would induce a high and faster humoral immune response even of PTA *** study strongly suggested that high or medium titers of MDAs might explain H9N2 inactivated vaccine failure in the field.

Colorectal cancer(CRC)is the second leading cause of cancer-related death *** five-year survival rate of CRC patients depends on the stage at diagnosis,being higher than 80%when CRC is diagnosed in the early stages but lower than 10%when CRC is diagnosed in advanced *** against specific CRC autoantigens(tumor-associated antigens(TAAs))in the sera of patients have been widely demonstrated to aid in early ***,we herein aim to identify autoantigens target of autoantibodies specific to CRC that possess a significant ability to discriminate between CRC patients and healthy individuals by means of liquid *** that end,we examined the protein content of the exosomes released by five CRC cell lines and tissue samples from CRC patients by means of immunoprecipitation coupled with mass spectrometry analysis.A total of 103 proteins were identified as potential autoantigens specific to *** bioinformatics and meta-analysis,we selected 15 proteins that are more likely to be actual CRC autoantigens in order to evaluate their role in CRC prognosis by Western blot(WB)and immunohistochemistry(IHC).We found dysregulation at the protein level for 11 of these proteins in both tissue and plasma exosome samples from patients,along with an association of nine of these proteins with CRC *** validation,all but one showed a statistically significant high diagnostic ability to distinguish CRC patients and individuals with premalignant lesions from healthy individuals,either by luminescence Halotag-based beads,or by a multiplexed biosensing platform involving the use of magnetic microcarriers as solid support modified with covalently immobilized Halotag fusion proteins constructed for CRC *** together,our results highlight the usefulness of the approach defined here to identify the TAAs specific to chronic diseases;they also demonstrate that the measurement of autoantibody levels in plasma against the TAAs identified here

Introduction:Hepatitis C virus(HCV)vaccine candidates based on viral envelope glycoprotein E2 have shown notable effects on triggering protective immune ***:In this study,we investigated the roles of N-glycans in the immunogenicity of E2 in BALB/c mice using a DNA prime-protein boost immunization ***:We found that a specific N-glycosylation deletion could enhance the immunogenicity of E2 without affecting the expression and secretion of the ***,the mutant with the deletion of N-glycosylation named N2,induced the highest levels of IFN-γ,Gr B,Perforin,and cytotoxic T lymphocyte lysis(CTL)response compared to the wild type and other mutants of ***,the N2 mutant enhanced IL-12 production from CD11c+dendritic cells(DCs)and increased CD8+T activation and the specific antigen-presenting activity of DCs to CD4+Th1 *** from N2-immunized mice could efficiently inhibit the infection of cell cultured HCV to human Huh7.5.1 hepatocytes,suggesting that N2 can elicit effective HCV neutralizing *** on N2-immunized mice,we selected a monoclonal antibody C1 which can efficiently neutralize the infection of HCVcc with the IC50 of 10 n M,and this antibody markedly inhibited viral infection in vitro and in HCV-infected immune-competent humanized ***:Deletion of N-glycosylation of E2 can significantly enhance the capacity of E2 to induce both cellular and neutralizing antibody humoral immune *** current study may offer a promising strategy for the development of an effective B-and T-cell-based HCV therapeutic vaccine,and meanwhile highlights the role of envelope protein glycosylation in virus immune evasion.

Background:Splenic marginal zone(MZ) B cells are distinguished from conventional B cells in that they are noncirculating mature B cell located at the interface between the circulation and lymphoid *** respond to blood-borne antigens and rapidly differentiate into antibody-secreting plasma *** are also thought to react with self-antigens which can be released from the dead *** have found that MZB express high levels of the IgM Fc receptor(FcμR)and that FcμR mice have reduced MZB population,suggesting a role for FcμR in the development or maintenance of MZB ***:We compared the proportion and absolute numbers of cells at different stages during MZB development in WT and FcμR *** also analyzed the apoptosis and proliferation of these *** addition,we investigated LPS response of MZB and antibody production against a T-independent antigen NP-LPS in WT and FcμR ***:We found that both marginal zone precursor(MZP) and MZB were decreased in FcμR *** reduction of MZP and MZB was not due to impaired proliferation of these cells but rather due to their increased *** analysis revealed that FcμR MZB had reduced tonic signal,as shown by their reduced levels of phosphorylated SYK and AKT relative to WT ***,MZB in FcμR mice responded poorly to LPS in vivo as compared to MZB in WT *** with the reduced proportion of MZB and their impaired response to LPS,antibody production against the type 1 T-independent Ag,NP-LPS,was significantly reduced in FcμR ***:FcμR contributes to the survival of MZP and MZB in vivo possibly by enhancing the tonic BCR *** addition,FcμR plays a critical role in LPS response by MZB.
