Objective:The aims of this study were to investigate the immunomodulatory function of human amniotic fluid stem cells(hAFSCs) on B lymphocytes and to provide a theoretical basis for the treatment of autoimmune diseases with these ***:hAFSCs were isolated and cultured by the adherent method,and then were identified by characteristic phenotypic *** coculture of B lymphocytes with hAFSCs,the proliferation,apoptosis,differentiation,chemotaxis,and expression of the inhibitory costimulatory molecules B7 H1,B7 H3,and B7 H4 of B lymphocytes as well as the subtypes of B lymphocytes were examined in ***:B lymphocytes were activated by the costimulatory molecules,and coculture with hAFSCs significantly decreased the expression of CD80/CD86 on activated B *** with hAFSCs resulted in a significant decrease in Ki-67 expression as well as inhibition of B lymphocyte *** could inhibit the expression of B7 H4 and PD-L1 on the activated B *** expression of the chemokine CCR7 on both activated and nonactivated B lymphocytes was downregulated by coculture with *** activated B lymphocytes,coculture with hAFSCs resulted in a reduced proportion of memory B and plasma cells,resulting in reduced amounts of immunoglobulins IgM,IgA,and *** could balance the B1 to B2 cell subpopulation ratio by downregulating the proportion of B1 ***:hAFSCs could inhibit the proliferation,apoptosis,chemotaxis,and differentiation as well as expression of costimulatory molecules of human B lymphocytes in vitro,indicating hAFSCs as ideal seed cells with therapeutic potential for treating autoimmune diseases.
