AIM To investigate the signal pathway of histamine h receptors. histamine h3 receptor（h3 R） is a potential target in the treatment of various central nervous system（CNS） diseases, metabolic disorders and pain. In the absence of crystal structure of h3 receptor, a ligand-based approach was took in this work. A 3 D-QSAR pharmacophore model was generated based on a set of known structurally diverse h3 R antagonists/inverse agonists, and its qualities were evaluated by cost function. The predictive power was validated by test set prediction and Fischer`s randomization test. On the basis of the pharmacophore model, a series of benzofuran derivatives were synthesized. The most promising compound, ethyl 2-methyl-5-（3-（3-methylpiperidin-1-yl）propoxy）benzofuran-3-carboxylate（1） showed high affinity for h3 receptor and exhibited dose-dependent anti-nociceptive effects in mice formalin model. In summary, compound 1 may facilitate the development of a novel class of drugs for the treatment of pain.

The neurological recovery following traumatic brain injury（TBI） is limited, probably due to the deficiency of neurogenesis. histamine h3 receptor（h3R） negatively regulates the synthesis and release of neuronal histamine, which is suggested to be implicated in brain development and neurogenesis. Therefore, it is intriguing to explore the effects of h3R antagonism on TBI and the neurogenesis related mechanism. The h3R antagonism reduced lesion volume and alleviated the motor dysfunction at the late phase after TBI, which can be reversed by h3R agonist. Moreover, the h3R knockout showed a protection, but no further augment of this protection was observed after h3R antagonism in these h3R-/-mice. Inhibition of h3R also increased the number of neuroblasts in the subventricular zone and subgranular zone after TBI. This action was reversed after the application of a-FMh, an inhibitor of histidine decarboxylase to block histamine synthesis or in histidine decarboxylase knockout mice. Furthermore, h1 receptor antagonist but not h2 antagonist reversed the action of h3R antagonism. By using hrh1 fl/fl;NestinCre ERT2 and hrh2 fl/fl;NestinCre ERT2 mice, it is found that the h1 receptor, but not h2 receptor, in neural stem cells was involved in the increase of neuroblast number conferred by h3R antagonism. It suggests that the h3R antagonism activates histaminergic neurons and subsequent h1 receptor in neural stem cells to promote neurogenesis. Additionally, we found that h3R antagonism had no effect on the proliferation but promoted the differentiation of neural stem cells into neuroblasts to increase their number. Those new generated neuroblasts can migrate and differentiate into mature neurons at the penumbra area to contribute to functional recovery following h3R antagonism. Our study demonstrates that h3R antagonism displays neuroprotection against TBI through promoting neurogenesis;the activation of histaminergic neuron and h1 receptor in neural stem cells are required for

The entorhinal cortex（EC）mediates the majority of the connections between the hippocampus and other cortical areas and plays a crucial role in learning and *** the EC receives a dense histaminergic innervation from the tuberomamillary nucleus of the hypothalamus,the functions of histamine in the EC remain ***,we showed that histamine acted via histamine h1 receptor to directly depolarize the principal neurons in the superficial layers of the medial EC and enhanced their sensitivity to a depolarizing ramp-like current ***-induced depolarization was mediated via inhibiting Kir channels and required the activation of protein kinase *** addition,histamine reduced the frequency but not the amplitude of miniature IPSCs by acting at histamine h3 receptor,while it did not affect the frequency and the amplitude of miniature EPSCs,suggesting that histamine specifically decreases spontaneous GABA release on to principal *** Inhibition of spontaneous GABA release by histamine h3 receptor activation was mediated by voltage-gated Ca2+channels and extracellular Ca2+.however,histamine did not influence the excitability of the principal neurons in the deep layers of the medial EC,thus indicating that the histamine-elicited excitation is ***,microinjection of the histamine h1,h3,but not h2,receptor antagonist into the superficial,but not deep,layers of medial EC impaired rat spatial learning as assessed by water maze tasks,while they did not affect the motor function and exploratory activity in an ***,our study indicates that histamine plays an important role in spatial learning by regulating neuronal excitability and synaptic transmission in the superficial layers of the medial EC.

1 Carcinine(beta-alanyl histamine)is an imidazole *** present study was designedto characterize the pharmacological effects of carcinine on histaminergic activity in the brain andon certain neurobehavior.2 Carcinine was highly selective for the histamine 1-I3 receptor over h1 or h2 receptor [Ki (laM)= 0.2939 0.2188 vs. 3621.2 583.9 or 365.3 232.8 M, respectively].3 Carcinine, at a dose of 20 mg kg-slightly increased histidine decarboxylase (hDC) activity inthe cortex (from 0.186 0.069 pmol mg protein"l min1 to 0.227 0.009 pmol mg protein"1 rein’l).In addition, carcinine (10, 20 and 50 mg kg"1) significantly decreased histamine levels in micebrain.4 Like thioperamide, a histamine ha receptor antagonist, carcinine (20, 50 gM) significantlyincreased 5-hT release from mice cortex slices, but had no apparent effect on dopamine release.5 Carcinine (20 mg kg1) significantly inhibited pentylenetetrazole- induced kindling. Thisinhibition was completedly reversed by (R)-methylhistamine, a representative ha receptoragonist, and ot-fluromethylhistidine, a selective histidine decarboxylase inhibitor.6 The results of this study provide first and direct evidence that carcinine, as a novel histamineha receptor antagonist, plays an important role in histaminergic neurons activation and might beuseful in the treatment of certain diseases, such as epilepsy.
