The volume of hippocampal subfields is closely related with early diagnosis of Alzheimer's *** to the anatomical complexity of hippocampal subfields,automatic segmentation merely on the content of MR images is extremely *** presented a method which combines multi-atlas image segmentation with extreme learning machine based bias detection and correction technique to achieve a fully automatic segmentation of hippocampal *** diffeomorphic registration driven by symmetric mutual information energy was implemented in atlas registration,which allows multi-modal image registration and accelerates execution *** exponential function based label fusion strategy was proposed for the normalized similarity measure case in segmentation combination,which yields better combination *** test results show that this method is effective,especially for the larger subfields with an overlap of more than 80%,which is competitive with the current methods and is of potential clinical significance.

Associations of Cognitive Deficits and Clinical Symptoms with hippocampal subfields in First-Episode and Never-Treated Patients with Schizophrenia

Objective:Cognitive impairment is a core debilitating symptom of schizophrenia(SCZ),with memory dysfunction and associated hippocampal disturbances playing a crucial role. The hippocampus is composed of inter-connected subfields,including the cornu ammonis(CA),which is abnormal in chronic SCZ. However,the relationships between cognitive function and the hippocampal subfields have not been examined in first-episode never treated(FENT) schizophrenia patients. Methods:Using a state-of the-art automated algorithm,the hippocampal subfields were segmented in 39 FENT SCZ patients and 30 healthy controls. We utilized the Chinese-language version of the MATRICS consensus cognitive battery(MCCB) for cognition and the PANSS for psychopathology. Resutls:We did not find significant differences in any hippocampal subfields(all P>0.05) between the patients and controls. However,multivariate regression analysis identified the left CA1,left hippocampal tail,left presubiculum and right molecular layer as contributing 40% to the variance of the PANSS negative symptom score. Compared to healthy controls,patients had significantly lower cognitive performance on the MCCB total and all(all P0.05). After adjusting for sex,age,education and intracranial volume,the partial correlation analysis showed that the volumes of left CA1,CA3,CA4,molecular layer,granule cell layer and both left and right subiculum were negatively correlated with the MCCB Hopkins Verbal Learning Test(HVLT). Additional multiple regression analysis showed that the left CA1 and CA3 hippocampal abnormalities contributed 66% to the variance of the HVLT score. Conclusion:Our results suggest no detectable hippocampal subfield deficits in FENT SCZ patients. However,the hippocampal subfields may be involved in the symptoms and cognitive deficits of SCZ patients in the early phase of their illness.

Background: Many studies have found that the hippocampus plays a very important role in major depressive disorder(MDD). The hippocampus can be divided into three subfields: the cornu ammonis(CA), dentate gyrus(DG) and subiculum. Each subfield of the hippocampus has a unique function and are differentially associated with the pathological mechanisms of MDD. However, no research exists to describe the resting state functional connectivity of each hippocampal subfield in MDD. Methods: 55 patients with MDD and 25 healthy controls(HCs) matched for gender, age and years of education were obtained. A seed-based method that imposed a template on the whole brain was used to assess the resting-state functional connectivity(rsFC) of each hippocampal subfield. Results: Patients with MDD demonstrated increased connectivity in the left premotor cortex(PMC) and reduced connectivity in the right insula with the CA seed region. Increased connectivity was reported in the left orbitofrontal cortex(OFC) and left ventrolateral prefrontal cortex(vlPFC) with the DG seed region. The subiculum seed region revealed increased connectivity with the left premotor cortex(PMC), the right middle frontal gyrus(MFG), the left ventrolateral prefrontal cortex(vlPFC) and reduced connectivity with the right insula. ROC curves confirmed that the differences between groups were statistically significant. Conclusion: The results suggest that the CA, DG and subiculum have significant involvement with MDD. Specifically, the abnormal functional connectivity of the CA may be related to bias of coding and integration of information in patients with MDD. The abnormal functional connectivity of the DG may be related to the impairment of working memory in patients with MDD, and the abnormal functional connectivity of the subiculum may be related to cognitive impairment and negative emotions in patients with MDD.

Research for Early Recognition of Alzheimer's Disease Based on hippocampal Subfield Volume and Regional Cerebral Blood Flow

Objectives:We intended to describe the reduction of hippocampal subfields volume as well as the changes of regional CBF in individuals at the risk of AD. We hypothesized that combination of hippocampal subfields volumes and regional CBF can acquire higher accuracy than either method alone in discriminating mild cognitive impairment( MCI) patients from cognitively normal elderly adults. Methods:44 early mild cognitive impairment( EMCI) patients,50 late mild cognitive impairment( LMCI) patients and 40 healthy controls( HC) underwent sMRI and ASL-MRI scan along with a battery of neuropsychological tests. hippocampal subfields volume was acquired by institutional specially-design software,with total intracranial volume normalization. Normalized regional and whole-brain voxel-based CBF differences were calculated using voxel-based analysis software. We compared the differences in hippocampal subfields volumes and regional CBF among the three groups. Combined with the results of multidimensional cognitive psychology evaluation,we analyzed the significant correlation between hippocampal subfield volumes and regional CBF with behavioral performance. Finally,Various combinations of these parameters were used to create the Receiver Operating Characteristic curves( ROC) in distinguishing MCI patients from the controls. Results:1. There were statistically significant differences in episodic memory score and executive function score among the three groups. 2. The normalized volume of multiple subfields in bilateral hippocampus was significantly lower in LMCI patients than in the controls( P<0.05). 3. The normalized volumes of each hippocampus,left DG,right CA2,right CA3 and right DG were significantly positive correlated with performance in episodic memory( P<0.001) and Logical Memory Test-Delay recall(LMT-DR)(P<0.001) in all subjects. 4. Left medial frontal gyrus CBF was significantly higher in the EMCI patients than the controls( P<0.01). Right anterior/meddle cingulate gyrus
