Physiological functions of hemocytes newly emerged from the cultured hematopoietic organs in the silkworm, Bombyx moil

Cellular immunity is a very important part of insect innate immunity. It is not clear if hemocytes entering the hemolymph require a maturation process to become competent. The establishment of a tissue culture system for the insect hematopoietic organs would enable physiological function assays with hemocytes newly emerged from hematopoietic organs. To this end, we established a hematopoietic organ culture system for the purebred silkworm pnd pS and then studied the physiological functions of the newly emerged hemocytes. We found that Grace＇s medium supplemented with 10% heated silkworm larval plasma was better for culturing the hematopoietic organs ofpndpS. Newly emerged hemocytes phagocytosed propidium iodide-labeled bacteria and encapsulated the Iml-2 coated nickel beads as well as pupal tissue debris. This culture system is therefore capable of generating physiologically functional hemocytes. These hemocytes can be used to study the mechanisms of the hemocyte immune response among others.

Characterization of hematopoiesis in Dabry's sturgeon(Acipenser dabryanus)

The hematological indices of Dabry's sturgeon(Acipenser dabryanus)were investigated in this *** results showed that the count of Red blood cell(RBC)was(0.60±0.02)1012/L,and the blood glucose(GLU)was 9.32±1.00 mmol/***-pyruvic transaminase(ALT)and glutamic-oxalacetic transaminase(AST)were 13.75±6.65 U/L and 359.50±40.07 U/L,*** of peripheral blood and imprints of four tissues were studied in the trial with Wright's staining *** results indicated that Dabry's sturgeon's blood cells could be divided into five cell types,and included erythroid,granuloid,monocytoid,lymphoid and other *** addition,the developmental stage of each cell lineage could be subdivided into:primitive,young and *** the three developmental stages,the size of each cell type was similar and gradually decreased from primitive,young to mature blood *** the blood cells,the monoblast and macrophage were the biggest,while the thrombocyte was the *** stages of the primitive cells and young cells were observed in head kidney and all cell types,except granuloblasts,were identified in the head *** contrast,primitive cells and young cells were not observed in the spleen or liver,and few orthochromatic erythroblasts were found in peripheral *** can be concluded that head kidney and trunk kidney are the primary organs of hematopoiesis in the Dabry's sturgeon.
