Early life gut microbiota construction is affected by diverse factors, including delivery mode and feeding pattern. However, the effect of the two factors on infants’ gut microbiota construction and the variation of fecal microbial metabolites remain unclear and the conclusion from present studies are controversial. Therefore, this study aimed to further study how the two factors influence the infant’s gut microbiota construction from a birth-cohort. Methods A total of 218 fecal samples from 61 full-term born infants collected at four time points, 0, 7 days and 1, 3 months of age. Based on delivery mode and feeding pattern, infants were divided into 4 groups, namely BN(vaginal delivery and breast feeding, n=16), BC(C-section delivery and breast feeding, n=17), MN(vaginal delivery and mixed feeding, n=10) and MC(C-section delivery and mixed feeding, n=18). The gut microbiota composition and bacterial diversity were assessed using 16 S rRNA sequencing. Short chain fatty acids(SCFAs) concentrations were determined by GC-MS. Results No statistical significance were observed in baseline characteristics(including sex, birth weight, height, etc.) among the four groups. No significant changes were found in alpha and beta diversity among four groups at four time points. At 0 day, 1 month and 3 month, the relative abundance of Firmicutes, Blautia, Enterococcaceae and Enterococcus in MC group were significantly higher than that in BN and BC groups. No statistical significance found in SCFAs among the four groups at four time points. Longitudinally, BN and BC groups exhibited a stable variation and lower concentration of propionate and butyrate. However, MC group showed a significant higher concentration of SCFAs over time. At day 0, the enterotypes of infants in BC and MC groups were Streptococcaceae, while a number of infants in BN group clustered to Bifidobacteriaceae. All the groups showed an intense transition of enterotypes within 1 month and became stable at 3 month. The
