Objective To study the properties and possible mechanisms of the long-term synaptic plasticity induced by the facial tactile stimulation at mossy fiber-granule cell（MF-GC） synapses in urethane-anesthetized mice, by field potential recording technique and pharmacological methods. Methods Adult ICR mice（6–8-week-old） were anesthetized with urethane, and tracheotomized to avoid respiratory obstruction. The craniotomy was drilled to expose the cerebellar surface corresponding to Crus II. Results Our results showed that 4 Hz facial stimulation evoked long-term depression（LTD） of MF-GC synaptic transmission. However, 20 Hz facial stimulation evoked long-term potentiation（LTP） at MF-GC synapses. The MF-GC LTP was prevented by N-methyl-Daspartic acid receptor（NMDA） receptor antagonist, and could be pharmacological induced by application of NMDA. Notably, blockade of protein kinase C（PKC） pathway, 20 Hz facial stimulation failed to induce the MF-GC LTP. In addition, the MF-GC LTP could be enhanced by blocking γ-aminobutyrate A（GABAA） and cannabinoids I（CB1） receptors activities, but was decreased by inhibiting of nitric oxide synthesis. These results indicated that high frequency facial stimulation induced an NMDA receptor dependent, GABAA receptor and NO involved, and PKC signaling required LTP of MFGC synaptic transmission in vivo in mice. Conclusion Our results suggest that the sensory stimulation evoked MF-GC synaptic plasticity might be involved in motor learning in living animals.

Long-term administration of scopolamine interferes with nerve cell proliferation, differentiation and migration in adult mouse hippocampal dentate gyrus, but it does not induce cell death

Long-term administration of scopolamine, a muscarinic receptor antagonist, can inhibit the survival of newly generated cells, but its effect on the proliferation, differentiation and migration of nerve cells in the adult mouse hippocampal dentate gyrus remain poorly understood. In this study, we used immunohistochemistry and western blot methods to weekly detect the biological behaviors of nerve cells in the hippocampal dentate gyrus of adult mice that received intraperito- neal administration of scopolamine for 4 weeks. Expression of neuronal nuclear antigen （NeuN; a neuronal marker） and Fluoro-]ade B （a marker for the localization of neuronal degeneration） was also detected. After scopolamine treatment, mouse hippocampal neurons did not die, and Ki-67 （a marker for proliferating cells）-immunoreactive cells were reduced in number and reac hed the lowest level at 4 weeks. Doublecortin （DCX; a marker for newly generated neurons）-im- munoreactive cells were gradually shortened in length and reduced in number with time. After scopolamine treatment for 4 weeks, nearly all of the 5-bromo-2＇-deoxyuridine （BrdU）-labeled newly generated cells were located in the subgranular zone of the dentate gyrus, but they did not migrate into the granule cell layer. Few mature BrdU/NeuN double-labeled cells were seen in the subgranular zone of the dentate gyrus. These findings suggest that long-term administration of scopolamine interferes with the proliferation, differentiation and migration of nerve cells in the adult mouse hippocampal dentate gyrus, but it does not induce cell death.
