Prevalence of Antibodies to Toxoplasma gondiiand Neospora caninumin Pigs in Grenada, West Indies

The aim of the study was to estimate the prevalence of antibodies to Toxoplasma gondii (T. gondii) and Neospora caninum (N. caninum) in pigs in Grenada, West Indies. T. gondii is a serious zoonosis affecting the unborn fetus and immunocompromized individuals. N. caninum is a similar coccidian parasite, which is not zoonotic, but is the cause of abortion and neonatal mortality in livestock similar to T. gondii. An earlier study conducted in Grenada and using a modified agglutination test (MAT) revealed seropositivity to T. gondii in pigs. No information is available on N. caninum infection of pigs in the Caribbean islands including Grenada. Serum samples from 185 pigs in Grenada, West Indies were tested for antibodies to T. gondii and N. caninum using an indirect enzyme linked immunosorbent assay (ELISA). Antibodies to T. gondii were found in 24.3% of pigs (95% confidence interval (CI): 18.12% to 30.48%) as all the tested pigs were negative for antibodies to N. caninum. Although, seroprevalence for T. gondii was higher in females (25.75%) than in males (20.70%), this result was statistically insignificant (p = 0.57). The results were similar to a previous study in Grenada confirming the continuity of infection in pigs. Human Toxoplasmosis is transmitted mainly through ingestion of tissue cysts in contaminated raw or undercooked meat or sporulated oocysts in soil, water or vegetables. Education of farmers and the Grenadian community on epidemiology of these parasites is warranted to prevent infection in pigs and in humans. This is the first report on the seroprevalence of N. caninum in pigs in the Caribbean region.

Expression of p30, the major surface antigen of Toxoplasma gondii, in baculovirus-insect cell system and the evaluation of immune response induced by p30

Objective: To produce the major surface antigen (p30) of Toxoplasma gondii from the Baculovirus Expression System. Methods: The p30 coding sequence was cloned into a transfer vector, then the recombinant baculovirus containing p30 gene was cloned and purified by the co-transfection and plaque assay. The expression and immunoactivity of the recombinant p30 were analyzed by SDS-PAGE and Western blot. The immune responses in mice for being immunized with recombinant p30 were tested. Results: About 750μg of purified (95% purity) p30 was obtained from a culture of 108 in- sect Sf21 cells. Mice in injected with the recombinant protein produced specific humoral and cellular immune responses. Immunization with p30 also prolonged the period of mice survival infected by Toxoplasma gondii. Conclusion: It is indicated that the recombinant p30 from baculovirus expression system can stimulate mice to produce effective protection from Toxo- plasma gondii infection.

It has been shown that some parasites interfere with malignant tumor cells growth in vivo or in vitro. In this work anticancer activity of three live protozoan parasites;Toxoplasma gondii, Trichomonas vaginalis and Leishmania major on fibrosarcoma cells growth has been investigated in cell culture medium. In this experimental study, WEHI-164 fibrosarcoma cells treated with alive Toxoplasma gondii tachyzoite, Trichomonas vaginalis trophozoite or Leishmania major promastigote as case groups or left intact as control groups. Following 24 hours incubation the number of cells, lactate dehydrogenase (LDH) and apoptosis were determined in case and control groups. Toxoplasma gondii tachyzoite decreased cell proliferation and increased cell lyses’ but it did not induce apoptosis. Trichomonas vaginalis or Leishmania major didn’t show any effects on cell proliferation, cell lyses or apoptosis. Therefore Toxoplasma gondii may have anticancer activity and further works is recommended to understand the mechanisms of action.

细胞渗透肽TAT与3种弓形虫抗原的融合重组表达

为了探索新型弓形虫疫苗的传递系统,本试验分别构建了细胞渗透肽反式转录激活因子(TAT)与3种弓形虫抗原和增强型绿色荧光蛋白(EGFP)融合表达的重组质粒,即pET28a-TAT-EGFP,pET28a-TAT-SAG1,pET28a-TAT-GRA4和pET28a-TAT-AMA1质粒。重组质粒被转化到大肠杆菌中并通过IPTG诱导,成功进行了融合表达,表达产物采用Ni-NTA树脂进行纯化,然后进行SDS-PAGE分析。结果得到31 ku的TAT-EGFP融合蛋白、34 ku的TAT-SAG1融合蛋白、38 ku的TAT-GRA4融合蛋白和62 ku的TAT-AMA1融合蛋白,经过Western blotting分析,感染弓形虫的小鼠血清可特异性地识别融合TAT的SAG1、GRA4蛋白和微弱地识别TAT-AMA1蛋白。

Objective:To analyze and review the overall seroprevalence rate of Toxoplasma gondii(***) infection in cattle from ***:In the current study,data collection(published and unpublished papers,abstracts of national scientific congresses and dissertations) using particular terms was carried out systematically on the following electronic databases like PubMed,Google Scholar,Ebsco,Science Direct,Scopus,Magiran,Irandoc,IranMedex and SID(Scientific Information Database).Results:A total of 22 studies since 1983 to 2012 reporting the seroprevalence of toxoplasmosis in cattle from different regions of Iran met our eligibility *** pooled proportion of toxoplasmosis,using random effect model,among cattle in Iran from over the 30-year period was estimated 18.1%(95%CI:9.9%to 28.2%).Conclusions:This study firstly establishes a crude seroprevalence rate of Toxoplasma infection in cattle which can lead us to understand the condition of eattle toxoplasmosis,which have to take into accounted fur an appropriate and effective prevention and ***,it compares and discusses elaborately the role of risk factors including sex,age and breed in the epidemiology of the ***,it determines gaps and drawbacks in the prior studies which are greatly useful to design more accurate investigations in the future.

Is toxoplasmosis a potential risk factor for liver cirrhosis?

Objective:To document Toxoplasma gondii(***) antibody status in patients with liver disease,blood samples were taken from 180 hepatic patients and 180 healthy ***:Toxoplasma IgG antibody was detected using enzyme-linked immunosorbent assay and histopathological assessment of liver biopsy METAVIR score was ***:*** IgG antibodies were found in 32.8%of patients and in 22.2%of controls(P=0.02).Toxoplasma seropositivity was significantly associated with lymphadenopathy,history of blood transfusion and reflex impairment in *** hepatitis C virus(HCV)and chronic HCV-related cirrhosis groups compared to chronic HBV and chronic HBV-related cirrhosis groups expressed significantly higher prevalence of *** seropositivity(odds ratio(OR) =4;95%confidence interval(CI):1.3-12.6;P=0.013,OR=4.8;95%CI:1.5-14.9;P=0.006,respectively).Within the chronic HCV group,*** seropositivity significantly associated disease evolution as regards to METAVIR histopathological system for fibrosis and inflammation(OR=19.4;95%CI:2.3-165.2;P=0.0008,OR=0.29;95%CI:0.1-0.8;P=0.01,respectively).Albumin,international normalized ratio(INR) and platelets count were the laboratory parameters significantly altered in Toxoplasma-positrvc chronic HCV patients(P=0.00 l,0.03,0.04,respectively).Child-Pugh scoring for cirrhosis in chronic HCV group placed the majority of seropositive patient in class C with significant statistical difference compared to Child A reference group(OR= 0.08;95%67:0.01-0.5;P=0.003).Conclusions:Toxoplasma seropositivity was high in patients with cirrhosis and associated higher grades of inflammation and necrosis signifying disease evolution,suggesting that cirrhotic patients may thus form a risk group for toxoplasmosis.

Serological survey on some pathogens in wild brown hares(Lepus europaeus) in Central Italy

Objective: To determine the exposure of wild brown hares [Lepus europaeus(L. europaeus), pallas] to Anaplasma phagocytophilum(A. phagocytophilum), Borrelia burgdorferi(B. burgdorferi) sensu lato, Encephalitozoon cuniculi(E. cuniculi), Leishmania sp., Neospora caninum(N. caninum) and Toxoplasma gondii(T. gondii). Methods: Two hundred twentytwo blood serum samples of wild brown hares captured in protected areas of the province of Pisa(Central Italy) were tested to detect antibodies against the reported pathogens. Results: Thirty one(14.0%) animals resulted positive for at least one tested agent, with antibody titres ranging from 1:20 to 1:320. In particular, 13(5.8%) samples were positive to B. burgdorferis.l., 11(4.9%) to N. caninum, 3(1.3%) to T. gondii, 2(0.9%) to A. phagocytophilum and 2(0.9%) to Leishmania sp. No samples scored positive to E. cuniculi. Four animals(14.8%) resulted coinfected with 2 different pathogens. Conclusion: The obtained results showed that B. burgdorferi s.l. N. caninum, T. gondii, A. phagocytophilum and Leishmania sp circulate in wild brown hares in Central Italy, suggesting a possible role of L. europaeus as reservoir of these pathogens. The obtained results showed that autochthonous wild brown hares living in Central Italy have been exposed to several pathogens circulating in this area, suggesting a possible role of L. europaeus as reservoir.

Objective:To evaluate the prevalence of toxoplasmosis and rubella among pregnant women at Ouagadougou in Burkina ***:All patient sera were tested for rubella and toxoplasmosis anti-IgG using commercial ELISA kits(PlateliaTMRubella IgG and Platelia? Toxo IgG).The presence of anti-rubella and anti-toxoplasmosis tgM in serum samples was tested using commercial ELISA kits Platelia Rubella TgM and Platelia Toxo ***:Among all the pregnant women tested for toxoplasmosis and rubella,their prevalence were 20.3% and 77.0%,*** women in the age group of 18-25 years showed the highest frequency of anti-toxoplasmosis（34.5%） and anti-rubella IgG（84.6%）.The prevalence of anti-toxoplasma and anti-rubella IgG decreased between 2006 and 2008 from 32.7%to 12.1%and 84.6% to 65.0%,*** was no significant association between age and the mean titer of anti-toxoplasmosis IgG among pregnant ***:The diagnosis of toxoplasmosis and rubella is necessary in pregnant women in Burkina Faso because of the low immunization coverage rate of rubella and the high level of exposure to these two infections which can be harmful to the newborn if contracted by women before the third trimester of pregnancy.
