Electrochemical Studies of Glutathione Monolayer Assembled on A Polycrystalline Gold Electrode

The glutathione (GSH) monolayer and complex monolayer of GSH-metallic ion on polycrystalline gold electrode were studied by using K3Fe(CN)6 as the redox probe. As for the GSH monolayer, it was found that the metallic ions could open the ion-gate in the monolayer dramatically in the order La3+> Pb2+>>Ba2+>Ca2+ whereas Zn2+ ion closed the ion-gate. The complexes of GSH-metallic ions were capable of self-assembling the different kind of monolayer. All the differences were related to the structural configuration of the anchored GSH molecule, which changed with different metallic ions or pH.

Pulmonary fibrosis(PF) is currently regarded as a disorder characterized by uncontrolled proliferation of fibroblasts and excessive deposition of extracellular *** ROS production or oxidative stress induced by bleomycin(BLM) results in epithelial cells injury,its effect on lung fibrogenesis and the underlying mechanism are *** present study will investigate if BLM stimulate fibroblast proliferation and collagen secretion through its effect on cellular redox system. Through successful establishment of animal model of BLM induced PF in mice,the proliferation activity of primary fibroblasts was found greater,and collagenⅠ,Ⅲand MMP/TIMP-1 related to collagen metabolism changed *** addition,an unbalanced redox status was observed in these BLM stimulated *** the same time,the proliferation and collagen secretion of L929 cells were stimulated when BLM was added,and the Nrf2 protein level and activity were found down-regulated with increasing *** expressions of important antioxidant enzymes involved in glutathione metabolism, includingγ-GCS,GR and GPx were also altered and their changing trends were the same as that of Nrf2. Thus,glutathione in fibroblast was decreased,and cellular ROS,especially HO production was *** a result,the proliferation and collagen secretion of fibroblasts were controlled by cellular HO concentration via Nrf2/Keap1 ***,PI3K/Akt was found to be responsible for the inhibition of Nrf2 by *** results will provide important experimental evidence for exploring the pathogenesis of PF as well as useful clue for antioxidant therapy in PF.
