In order to determine how nociceptive input conveyed by the C-fibers terminating in superficial lam-inae of the spinal cord reaches the wide dynamic range (WDR) cells in deeper dorsal horn, which functions as ascend-ing projection pathway, the morphological features of some WDR cells in the deeper dorsal horn of the cat lumbar spinal cord were studied by intracellular injection of horseradish peroxidase and physiological characterization. One of the fully stained neurons with somata in lamina V and dendrites that entered lamina Ⅱ were examined by electron mi-croscopy. Immunogold staining of ultrathin sections through the labeled proximal dendrites in lamina Ⅱ revealed that these dendrites received numerous synapses from substance P and glutamate immunoreactive (IR) axons, which were considered originating from C-fibers. In addition, many GABA-IR terminals were found presynaptic to the labeled dendrites. The results, therefore, suggest that the information carried by primary afferent can be sent from

glutamate synthase (EC 1 4 1 14) was purified to homogeneity from a cell\|free extract of Streptomyces lincolnensis by precipitation with streptomycin sulfate and ammonium sulfate, and column chromatography on DEAE\| cellulose, Sepharose 6B, DEAE\|sephadex A\|50, hydroxyapatite and Sephadex G\|150. The enzyme activity is stabilized by addition of α ketoglutarate, PMSF,EDTA, β mercaptoethanol and glycerol. The native enzyme has a molecular weight of 138 000 and is composed of two nonidentical subunits with molecular weights of 81 000 and 57 000. Spectroscopic examination of the enzyme gave absorption maximum at 280 and none at 380 and 440 nm, indicating the absence of iron and flavin. The enzyme shows optimum activity at pH 7.2 and 30℃. Km values for α ketoglutarate, L\|glutamine and NADH were 417, 435, and 52.1 μmol/L, respectively. When NADPH was substituted for NADH as reductant, there was approximately 13% of the control activity. The activity of this glutamate synthase is inhibited by its products (i.e. glutamate and NAD), several metal ions, amino acids and tricarboxylic acid cycle intermediates.

In this study, in situ hybridization histochemistry technique was used to observe the effect of electro-acupuncture (EA) on glutamate transporter mRNA expression following focal cerebral ischemia. We observed that glutamate transporter mRNA expression decreased significantly in the area penumbra of cerebral cortex at 12h following focal cerebral ischemia. EA did not change glutamate transporter mRNA expression in normal rat brain but prevented the decrease of glutamate transporter mRNA expression in the area penumbra of cerebral cortex following focal cerebral ischemia. The results suggest that the protective effect of EA on ischemic neuronal death may bhe related to the increase of uptake of glutamate by its transporter.

Objective:Except for the well-recognized neuronal abnormalities,accumulating studies revealed the involvement of oligodendroglia(OL) and myelin deficits in major psychiatric disorders,including impaired myelin integrity,loss of OL and dysregulation of myelin related genes. Besides,some neurological disorders characterized by white matter abnormalities(e.g. leukodystrophies and multiple sclerosis) show some psychotic symptoms. However,whether and how OL and myelin deficits lead to psychiatric symptoms remains undefined. Our study aimed to test the hypothesis that impaired OL differentiation and myelination could contribute to the etiology of psychiatric disorders rather than simply accompanied pathological abnormalities. Methods:In this animal model study,we employed Olig2 conditional knockout mice(Olig2 cKO) by crossing Cnp-cre with floxed Olig2 mice. OL and myelin development were observed by immunostaining of brain slices. Ultra-structure of myelin in anterior cingulated cortex of young adult mice(P56) was analyzed by transmission electron microscope(TEM). Localized proton magnetic resonance spectroscopy was used to detect neurochemicals in cerebral cortex at juvenile(postnatal day21,P21). Mice behaviors and cognitive functions were further analyzed by several well-defined behavioral tests,including open field,forced swim test,cliff avoidance test,prepulse inhibition(PPI) and novel object recognition test. Results:We found(1) Olig2 ablation caused a loss of mature oligodendrocytes(OLs),but not oligodendroglia progenitor cells(OPCs),in both anterior cingulated cortex(ACC) and corpus callosum(CC) at juvenile and young adult age.(2) Robust developmental myelin deficits were found in Olig2 cKO mice during adolescence,leading to severe hypomyelination in ACC of young adult mice.(3) Localized proton magnetic resonance spectroscopy revealed that Olig2 cKO mice displayed abnormally elevated cortical glutamate level.(4) Both the juvenile and young adult Olig2 cKO mice showed anxious phenotype and young adult Olig2 cKO m

The neurons with coexistence of serotonin-and giutamate-immunoreactivity(5-HT/Glu cells)in medullary raphe nuclei were investigated in the present study by using immunofluorescence technique.On the basis or the investigation projections of the 5-HT/Glu cells to the cerebellar cortex were studied by means or a combined microsphere retrograde transport.The results showed that most of the 5-HT cells in medullary raphe nuclei exhibited Glu-immunoreactivity.The cell count indicated that 81.77% of the 5-HT cells contained Gin-immunoreactivity;40.35%of the Gin-cells contained 5-HT.The retrograde labeled cells by microspkere(MS cells)were found in all medullary rapke nuclei. 5-HT/MS cells were rare.The Gin/MS cells had a rate in total MS cells(40.31 %).Three trible-labeled cells (5-HT/Glu/MS cells)were found. The finding or the 5-HT/Glu cells in medullary raphe nuclei projecting to the cerebellar cortex may give an iportant significance to understanding the functional connection between medullary raphe nuclei and cerebellum.

To investigate the intracellular mechanism that interleukin 1β (IL 1β) facilitates epileptic seizure and neuronal damage, the effect of IL 1β alone or IL 1β plus glutamate (Glu) on the intracellular free calcium ([Ca 2+ ] i) of single cultured hippocampal neuron was examined by using EPC 9 light electricity measurement system. The results showed that IL 1β of different concentrations (5×10 3 U/L, 10×10 3 U/L, 20×10 3 U/L, 30×10 3 U/L, 50×10 3 U/L, 100×10 3 U/L) failed to affect the neuronal [Ca 2+ ] i, but IL 1β could facilitate the augmentation of neuronal [Ca 2+ ] i induced by Glu in a dose dependent pattern. MK 801 inhibited the effect of Glu on [Ca 2+ ] i, and also inhibited the effect of IL 1β on [Ca 2+ ] i induced by Glu, while verapamil did not influence the effect of Glu or IL 1β. It is concluded that IL 1β, as a neuromodulator, can facilitate the activation of NMDA receptor by Glu, induce the increase of intracellular calcium, which enhances the excitement of neuron.
