Dear Editor,Previous studies have proved that regulatory T cell(Treg)insufficiency contributed to the development of autoimmune conditions including systemic lupus erythematosus(SLE).Conventional immunosuppressive treatment was reported to downregulate beneficial Tregs together with pathogenic effector immune cells,which may impede a rapid achievement of optimal therapeutic effects.

BACKGROUND There are many adverse reactions in the treatment of allergic rhinitis(AR)mainly with conventional *** receptor antagonists,glucocorticoids and nasal antihistamines can all be used as first-line drugs for AR,but the clinical effects of the three drugs are not *** To examine the impact of glucocorticoids,antihistamines,and leukotriene receptor antagonists on individuals diagnosed with AR,specifically focusing on their influence on serum inflammatory *** The present retrospective study focused on the clinical data of 80 patients diagnosed and treated for AR at our hospital between May 2019 and May *** participants were categorized into the control group and the observation *** control group received leukotriene receptor antagonists,while the observation group was administered glucocorticoids and *** an observation and comparison of the symptoms,physical sign scores,adverse reactions,and effects on serum inflammatory indexes in two distinct groups of patients,both before and after *** Subsequent to treatment,the nasal itching score,sneeze score,runny nose score,nasal congestion score,and physical signs score exhibited notable discrepancies(P<0.05),with the observation group demonstrating superior outcomes compared to the control group(P<0.05).The interleukin(IL)-6,IL-10,tumor necrosis factor-alpha,Soluble Intercellular Adhesion Molecule-1,Leukotriene D4 after treatment were significantly different and the observation group It is better than the control group,which is statistically significant(P<0.05).Following the intervention,the incidence of adverse reactions in the observation group,including symptoms such as nasal dryness,discomfort in the throat,bitter taste in the mouth,and minor erosion of the nasal mucosa,was found to be 7.5%.This rate was significantly lower compared to the control group,which reported an incidence of 27.5%.The difference between the two groups was statist

The circadian rhythm of glucocorticoid receptor (GR)in human peripheral leukocytes has been reported by us previously. It remains to be shown if the circadian variation of GR has any physiological significance. In order to answer the above question, the present work studies the circadian rhythm of the reactivity of the polymorphonuclear leukocytes (PML)to glucocorticoid (GC)and analyzes the correlation between the circadian rhythm of GR and the reactivity of PML to GC.

Aim Ginseng is the dried root of Panax ginseng C. A. Mayer. Since ancient times, ginseng has been used as one kind of treatment drug or tonic in China and even other eastern countries like Korea and Japan. Phar- macological active chemical ingredients and its extract of ginseng are a mixture of triterpenoid saponins, collectively called ginsenosides. Among them, ginsenoside Rgl is the most pharmacological active one. Based on prior experi- mental results and the understanding of alcoholic hepatitis, the major aim of this study is to investigate whether Rgl is beneficial in a rodent model mimic alcoholic hepatic injury associated with binge drinking and explore the under- lying possible mechanisms. Methods C57BL/6 mice were given oral consumption of 6 g · kg^-1 alcohol 1 h after treated with Rgl （ 10, 20 and 40 mg · kg^-1） or dexamethasone （ 1 mg · kg^-1） for 9 consecutive days. Biochemi- cal analyses were performed and liver fragments were processed for microscopy, immunohistochemistry and western blot analysis. Results According to our data, Rgl treatment significantly reversed the high mortality rate induced by alcohol consumption and also alleviated liver impairment as evidenced by the decrease of serum parameters. Meanwhile, histological and ultrastructural analysis of alcoholic groups showed hepatocellular impairment but re- stored in Rgl-treated groups. Overproductive inflammatory cytokines were also suppressed by Rgl in alcohol-intoxi- cated mouse livers. In addition, changes of GR related NF-KB pathway, including phospho-IKB-ot, were also mod- ulated to normal levels. Conclusion This study demonstrates that Rgl might promote GR mediating the repression of NF-KB and inhibit the inflammatory reactions in alcoholic hepatitis.

The effect of dexamethasone with different concentrations and different stimulating periods on the expression of glucocorticoid receptors （GRα, GRβ） protein was investigated in human monocyte cell line THP-1. The cultured human monocyte line THP-1 cells were stimulated by dexamethasone with different concentrations and different periods. The expression of GRα and GRβ protein was detected by Western hlotting. The results showed that the expression of GRα and GRβ was detected in the THP-1 cells, The quantity of GRα expression was reduced by dexamethasone under the same concentration with the prolongation of expression was increased by dexamethasone treatment the stimulating periods. The quantity of GRβ in a time- and dose-dependent manner. It was concluded that dexamethasone stimulation time-dependently reduced the GRα expression in THP-1 cells. Dexamethasone stimulation time- and dose-dependently increased the GRβ expression in THP- 1 cells. The expression of GRα and GRβas regulated by glucocorticoid.

BACKGROUND: The most common complication after allogenic islet transplantation is rejection. This study was to evaluate the effect of anti-rejection of glucocorticoid-free immunosuppressive regimen on allogenic islet transplantation. METHODS: Tacrolimus(FK506)+mycophenolate mofetil (MMF) and FK506+MMF+prednisone (Pred) were administered respectively for 2 weeks to inhibit rejection after allogenic islet transplantation in rats, which were compared with the control group. The concentrations of blood glucose, insulin and C-peptide were determined dynamically in recipients and the sites of transplantation were observed morphologically. RESULTS: As compared with the control group without immunosuppressive agents, FK506+MMF and FK506+MMF+Pred could prolong the survival time of grafts significantly. There were many morphologically intact islets in the liver of recipients 2 months after transplantation. Group FK506+MMF kept normal levels of blood glucose, insulin and C-peptide beyond 60 days after transplantation. In contrast, group FK506+MMF+Pred secreted less C-peptide(Pglucocorticoid-free immunosuppressive strategy of low-dose FK506 and MMF could protect islet grafts in islet transplantation without diabetogenic side-effects.

In cases of severe sepsis and septic shock, a series of pathophysiological changes lead to multiple organ dysfunction *** study aimed to investigate the expression of glucocorticoid receptor mRNA in the rat lung following endotoxin （LPS） induced shock. Totally 56 SD rats were randomly divided into 4 groups： LPS shock group （n=16）, LPS＋vasoactive intestinal peptide group（VIP） group, （n=16）, LPS＋VIP＋ glucocorticoid （GC） group, （n=16）,and control group （n=8）. LPS shock was induced by intravenous injection of LPS （10 mg/kg） in rats. Within 15 minutes after LPS injection, rats in the treatment groups received VIP （5 nmol/kg） or VIP and methylprednisolone （3 mg/kg）. The control group was given normal saline instead of LPS. The rats of the four groups were sacrificed at 6 hours,24 hours after injection respectively, and the lung tissues were collected. Pathological changes of the lungs were examined by light microscopy and electron microscopy. GRmRNA expression in the lung tissues was evaluated by RT-PCR. In the LPS shock group, lung histopathology demonstrated destruction of the alveolar space,widening of the inter-alveolar space, inflammatory cell infiltration and interstitial edema. However,pathological changes in the LPS＋ VIP group and LPS＋ VIP＋GC group were milder than those in the LPS shock group. Six hours after LPS injection, GR mRNA expression was down-regulated in the LPS group （0.72± 0.24） and LPS＋ VIP group （0.88±0.27）（P〈0.05） as compared with the control group （1.17±0.22）. The LPS shock group showed a more significant down-regualtion than the LPS＋VIP group, but the difference was not statistically significant （P〉0.05）. In contrast, GRmRNA expression in the LPS＋ VIP＋GC group was significantly up-regulated at 6 hours and further at 24 hours （1.45±0.32 and 1.91±0.46 respectively）（P〈0.05）.
