Cloning and Characterization of an mRNA Encoding F_1-ATPase Beta-Subunit Abundant in Epithelial Cells of Mantle and gill of Pearl Oyster, Pinctada fucata

In oyster biomineralization, large amounts of calcium are absorbed from external media, transported to the mineralization site, and finally deposited via a matrix-mediated process, All these activities are very energy intensive; therefore, investigations of the energy metabolism pathways of different oyster tissues will facilitate understanding of oyster biomineralization physiology. A full-length cDNA encoding the F1- ATPase beta-subunit （the F1-β-subunit, a major calalytic subunit of F-ATPase） from the pearl oyster （Pinctada fucata） was cloned using the homology strategy with a pair of degenerated primers based on the conserved regions of other animals＇ F1-β-subunit genes. Sequencing and structural analyses showed that the obtained sequence shared high identity with other animals＇ F1-β-subunits, and had a unique phosphorylation site of PKC and CK II on the external surface of the putative protein. Results from semi-quantitative reverse transcription-polymerase chain reaction and in situ hybridization demonstrated this oyster F1-β-subunit mRNA is abundant in the gill and mantle, and distributed widely in the periostracal groove, the outer folder. and the dorsal region of the mantle and in the gill epithelial cells. These tissues were the main regions that participate in biomineralization processes such as calcium uptake, transport, and matrix secretion. The results indicate that tissues involved in biomineralization have stronger energy metabolic processes and that F1-ATPase might play an important role in oyster biomineralization by providing energy transport.

The Lateolabrax maculatus is an important marine fish in China that can adapt to a wide range of water *** gill is the main organ involved in fish osmoregulation and in this study,a cDNA library of the *** gill was constructed to understand gill function in Lateolabrax maculatus and it adaptation to salinity.A total raw data of 5.3G base pairs was obtained,followed removal of low-quality and linker DNA sequences and it yielded a total of 23,038,590 clean *** splicing and clustering,31,976 unigenes were *** average transcript length was 1327 bp and N50 was 2057 *** comparisons with other tissues analysis of the gill transcriptome revealed a high level of relative expression of apolipoprotein E,CD74,keratin,type I cytoskeleton 13,carbonic anhydrase,betaine-homocysteine S-methyltransferase,and aquaporin 3(AQP3)*** *** aqp3(Lm-aqp3)gene was specific for the gills Lm-aqp3 and this gene has a typical transmembrane structure of six alpha helices and a water channel *** analysis indicated that Lm-aqp3 clusters with the homologue genes from Labrus bergylta and Dicentrarchus *** RT-PCR confirmed that Lm-aqp3 was highly abundant in the gills and low expressed in other tissues(stomach,liver,brain,spleen and testis).Analysis of Lm-aqp3 in freshwater aquaculture fish revealed that gene expression was significantly statistical higher than in seawater(20 ppt)fish,suggesting that Lm-aqp3 may play an important role in water metabolism and osmotic pressure regulation in ***.

Serum osmolality and ions,and gill Na^(+)/K^(+)-ATPase of spottedtail goby Synechogobius ommaturus(R.)in response to acute salinity changes

This study was carried out to determine the effects of abrupt salinity change on osmoregulatory ability of the spottedtail goby Synechogobius ommaturus.720 juvenile fish(65.3±11.8 g)were transferred to 200 L tanks(with 40 juveniles in each tank),in which salinities were abruptly changed from 10 to 20,30,40,50 and *** rate,serum osmolality,electrolytes(Na^(+),Cl^(-),and K^(+))and gill Na^(+)/K^(+)-ATPase(NKA)activity were assessed successively in 528 *** showed serum osmolality,ion concentrations and gill Na^(+)/K^(+)-ATPase activity increased significantly when fish were transferred to salinity 40 and 50 and all fish in these groups died by the end of the *** osmolality,Na^(+),Cl^(-)and K^(+)in fish transferred to a salinity of 20,30 and freshwater were not affected and no mortality was *** with the control group,a significantly decrease of NKA activity happened in the freshwater group,but the activity in 20 and 30 groups was not affected *** results indicated that *** could adapt rapidly and maintain homeostasis in a wide range of salinities(from freshwater to salinity 30)and this species may be suitable for aquaculture in estuarine and coastal areas where rapid salinity fluctuations commonly occur.

The impact of mercury stress on Venerupis philippinarum gill and haemocytes was assessed through investigating the dynamic changes of several biochemistry parameters, including superoxide dismutase（SOD）, catalase（CAT）, glutathione reductase（GR）, acid phosphatase（ACP）, alkaline phosphatase（ALP）, total glutathione content, as well as malondialdehyde（MDA） content, under different Hg2+ acute exposure in the present study. Results showed that SOD activities and GSH contents in the gill displayed depressed expression. The correlation analysis indicated significant positive correlations of the two parameters（0.833, P ＜ 0.01）. GR showed elevated activities in all examined exposure doses. While the activities of CAT and ALP were only induced at higher Hg2+ concentrations with significant positive correlations（0.917, P ＜ 0.01）, and MDA showed decreased contents in middle exposure doses. In term of haemocytes, Hg exposure had an induced effect on the activities of CAT, GR, ACP and ALP, and MDA content increased with increasing concentration of Hg at 24 h. All these parameters had significant positive correlations（P ＜ 0.01）. No obvious dose-dependent or time-dependent expression profiles were detected in other indexes in both tissues. All these results showed that acute Hg exposure led to harmful effects on clams immune system, which are likely linked to Hg induced oxidative stress. CAT, ACP, and MDA in gill have great potential as biomarkers of Hg concentration determination in the aquatic environment for the studied species.

In order to understand the acclimation strategies of Red tilapia to different environments, this present study aimed to evaluate different responses of gill ionocytes of Red tilapia（O. mossambicus ♀× O. niloticus ♂） transferred from freshwater（FW） to saline water（SW, 10-30‰）, alkaline water（AW, 1-3‰） and saline ＆ alkaline water（S＆AW, 10/1-30/3‰）. Localization, type, size, and numeration of gill ionocytes were investigated on same specimens by scanning electron microscopy（SEM） and immunohistochemistry（IHC） with antibodies of Na+/K+-ATPase（NKA）, Na+/K+/2Cl-contransporter（NKCC）, cystic fibrosis transmembrane conductance regulator（CFTR） and carbonic anhydrase（CA）. Ionocytes were only located on filaments conducted by SEM. Four types of ionocytes, namely pit, convex, concave and transitory types were determined morphologically by their apical openings, of which concave and transitory type weren’t presented in FW and SW（10‰）. Both ionocytes size and number increased with elevated stress levels. In comparison to FW, density of ionotypes increased 4.75-, 3.00- and 3.44-fold in SW（30‰）, AW（3‰） and S＆AW（30/3 ‰）, respectively. The size of pit type increased dramatically in SW, but kept constant in AW and S＆AW, while the size of convex and transitory types increased greatly in AW and S＆AW. IHC results on gill filaments also confirmed branchial distribution of ionocytes. Immunoreaction of NKA, NKCC and CA appeared in FW except for CFTR, while they all appeared in SW（30‰）, AW（3‰） and S＆AW（30/3‰）.

腰椎间盘突出症(lumber disc herniation,LDH)是临床常见病,首先推荐保守治疗,但研究认为,腰椎间盘突出症的很多保守治疗方法并不被循证医学所支持,许多学者为此又根据新的生物力学研究成果进行了探索,Mcgill法和悬吊技术就是其中的代表[1—2]。Stuart Mcgill是脊柱损伤预防及康复领域的专家,他认为躯干稳定就是躯干在活动中保持不变形的能力[1],这可减少脊柱的剪切力,使脊柱在较低椎间盘内压下完成活动,降低损伤的风险。但患者习惯用腰椎的运动来代偿髋关节的运动,这种误用是腰部损伤的原因之一。

BIOMONITORING OF AMBIENT CONCENTRATIONS OF CADMIUM,COPPER,LEAD AND ZINC IN THE COASTAL WETLAND WATER BY USING gillS OF THE GREEN-LIPPED MUSSEL PERNA VIRIDIS

The distribution and concentrations of Cd, Cu, Pb and Zn were determined in the gills and remaining soft tissues of Perna viridis collected from 12 geographical sites (10 from the west and 2 from the east coastal waters) of Peninsular Malaysia. All samples showed that the levels of Cd, Pb and Zn were generally higher in the gill than those in the remaining soft tissues. These results could be due to the fact that gills are the first organ of metal accumulation and larger surface area with mucus sheets in the organ. Since the mussel gill is a better accumulator of Cd, Pb and Zn of ambient seawater than remaining soft tissue, it is a potential indicator of ambient levels of dissolved metals in the ambient seawater. However, further validations based on laboratory conditions are needed.
