pWM240,a plasmid with the size of 4,277 base-pair,was isolated from Ochrobactrum anthropi KT-C024,and its complete nucleotide sequence was determined.Sequence analysis indicated a putative oriT site and 5 ORFs,including one replication protein (ORF1) and one mobilizing protein (ORF3) in pWM240.Presence of ssDNA intermediate in O.anthropi cells suggested pWM240 replicates via rolling-circle- replication (RCR).pWM241 was constructed by inserting a kanamycin resistance gene into pWM240 and pWM242 was constructed with an addition of green fluorescent protein gene into pWM241 in order to confer pWM240 to screening markers. The mutation of hypothesis mobilizing protein gene (ORF3) disrupted the conjugative function of pWM241,which indicated ORF3 endowed the plasmid with transferable characters,pWM241 was employed successfully to express cyanide-degrading gene cya-1 from Pseudomonas aeruginosa KT-C001,which showed the vector pWM241 has the potential in applying the gene enhanced technology (GET) into wastewater treatment,pWM242 had a broad host range and was stably maintained with antibiotic selection when transformed into Escherichia coli, Pseudomonas aeruginosa,Pseudomonas stutzeri,Paracoccus denitrificans and Ochrobactrum sp.The green fluorescent protein was expressed in these species too.
