Two ligand oligopeptides GV1 and GV2 were designed according to the putative binding region of VEGF to its receptors. GV1, GV2 and endosome releasing oligopeptide HA20 were conjugated with poly-L-lysine or protamine and the resulting conjugates could interact with DNA in a noncovalent bond to form a complex. Using pSV2-β-galactosidase as a reporter gene, it has been demonstrated that exogenous gene was transferred into bovine aortic arch-derived endothelial cells (ABAE) andhuman malignant melanoma cell lines (A375) in vitro. In vivo experiments, exogenous gene was transferred into tumor vascular endothelial cells and tumor cells of subcutaneously transplanted human colon cancer LOVO, human malignant melanoma A375 and human hepatoma graft in nude mice. This system could also target gene to intrahepatically transplanted human hepatoma injected via portal vein in nude mice. These results are correlated with thegene delivery system targeting VEGF receptors relevant receptors (flt-1, flk-1/KDR) expression on the targeted cells and tissues.

＜正＞Purpose To establish an efficient and stable gene delivery system based on recombinant adeno-associated viral vectors for retina research,such as dendrite development patterns of mouse retinal cells,especially retinal ganglion cells, and how the cell fate of various retinal cells is *** A bicistronic plasmid vector,pSIEWB,was acquired by molecular manufacture with elements derived from pSEWB which contains AAV genome and provides main structure of the vector and pCAGGS which provides the fragment containing IRES and EGFP *** of recombinant AAV vector was performed by a dual-vector,calcium phosphate-mediated transfection approach with pSIEWB and pD1rs which provides the AAV coat protein genes and helper virus genes necessary for packaging of recombinant AAV vector. Results The bicistronic plasmid vector,pSIEWB,has been acquired via replacing EGFP gene of pSEWB by fragment containing IRES-EGFP sequence derived from pCAGGS and it possesses the genome of recombinant AAV including two inverted terminal repeat elements（ITRs） which are necessary for viral packaging,human synapsin 1 promoter（hSyn 1） which can drive gene expression in neuron specially,IRES which can make functional gene and reporter gene express individually,EGFP which is a popular reporter gene to localize gene expression of vector,and woodchuck hepatitis post-transcriptional regulatory element（WPRE） which can optimize gene expression in *** the same time,an dual- vector, calcium phosphate-mediated transfection approach for viral packaging has been performed efficiently and stably in our lab,which needs two plasmids,pSIEWB and pD1rs which works as a helper plasmid for viral packaging because it contains AAV coat protein genes and adenovirus genes necessary for *** This study has provided a primary form of a gene delivery system for retina *** study on the process of dendrite development of retinal ganglion cells and cell fate determinatio

Circumvent PEGylation dilemma by implementing matrix metalloproteinase-responsive chemistry for promoted tumor gene therapy

Surface modification by poly(ethylene glycol)(PEGylation)has been acknowledged as a powerful strategy in minimizing non-specific reactions for biomedical *** applied into manufacture of drug/gene delivery systems,PEGylation has demonstrated to significantly improve their biocompatibility and stealthiness in physiological ***,reluctant cell membrane affinities thus cellular uptake efficiencies owing to PEGylation brought up further issues that are imperative to be *** to this PEGylation dilemma,we attempted to introduce peptide(GPLGVRG)linkage between block copolymer of PEG-poly{N’-[N-(2-aminoethyl)-2-aminoethyl]aspartamide}PAsp(DET),wherein the cationic PAsp(DET)could self-assemble with pDNA into nanoscaled complex *** was the peptide linkage whose amino acids sequence could be specifically recognized and degraded by matrix metalloproteinases(MMPs)(overexpressed in extracellular milieu of tumors).Therefore,our subsequent studies validated facile detachment of PEGylation from the aforementioned polyplex micelles upon treatment of MMPs,which elicited improved cytomembrane affinities and cellular uptake *** addition,promoted escape from endosome entrapment was also confirmed through direct endosome membrane destabilization by PAsp(DET),which was further elucidated to be attributable to dePEGylation as well as elevated charged density of PAsp(DET)in acidic *** benefits from dePEGylation eventually contributed to promoted gene expression at the affected cells and potent tumor growth suppression based on anti-angiogenic ***,our developed strategy has provided a facile approach in overcoming the dilemma of PEGylation,which could be informative in design of drug/gene delivery systems.

gene transfer vector is one of the key technologies of gene *** carriers generally can be divided into two major categories of virus and non-virus,the former is subjected to security *** latter much favored in medical science and also is currently the main object of pharmaceutical research because of its safety,vector design diversity and low *** non-virus carrier,polycation gene carrier due to the high rate of transfection and become a main research ***(PAMAM) is one of the important member in the family of cationic polymer carrier,PAMAM dentrimers are attracted highly attention recently due to their special properties in the drug and gene delivery systems,especially the high generation PAMAM as the gene vector shows strength transfection ability compared with the other non-viral carrier *** main drawback for the materials is presenting serious cell *** order to overcome this problem,one new gene delivery system has been designed by our research *** is that PAMAMs are covalently conjugated onto α-cyclodextrin(α-CD) via amide bonds to obtain the starburst-like cationic polymers(CD-PG2).The chemical structure and composition of CD-PG2 was characterized by *** physicochemical and biological properties of CD-PG2/pDNApolyplex were evaluated by agarose gel retardation,stability test against DNase I,MTT assay,DLS measurement,CLSM observation,LDH leakage test,and in vitro cell *** test results tell us that the CD-PG2 could efficiently condense pDNA into nano-scale particles with a narrow size distribution,and protest from DNase I ***-PG2 shows excellent gene transfection efficiency without serum interference as well as relatively low cytotoxicity,compared with free PET-25 *** commercial product Lipofectamine 2000(one good gene vector).The cell uptake route analysis shows cellular uptake CD-PG2/pDNApolyplex is mainly through Clathrin-mediated endocytosis,CME and Caveolae-medi
