ganoderic acid hinders renal fibrosis via suppressing TGF-β/Smad and MAPK signaling pathways

OBJECTIVE To investigatethe anti-renal fibrotic effect of ganoderic acid（GA） and the related mechanisms in vivo and in vitro. METHODS We investigated whether GA could hinder renal fibrosis and the related mechanisms in a unilateral ureteral obstruction（UUO） mouse model and TGF-β1-stimulated HK-2 cells model. The C57 mice were randomly devided into sham group, sham with GA administration group, UUO group and UUO with GA treatment group. After UUO surgery,the mice were treated with GA(3.125, 12.5 and 50 mg·kg;per day, ip) for 14 d. Then the mice were sacrificed for collecting blood and kidneys. The kidney functions were revealed by blood urea nitrogen and serum *** fibrosis and tubules injury levels were detected by Masson staining and HE staining. The epithelial-mesenchymal transition（EMT） level was detected by Western blotting and immunohistochemical staining. The changes of related signaling pathways and underlying mechanisms were demonstrated by Western blotting. The cell model was adopted to identify the potent inhibitor of renal fibrosis. RESULTS After 14 d treatment, GA dosedependently attenuated UUO-induced tubular injury and renal fibrosis; GA(50 mg·kg;) significantly ameliorated renal disfunction during fibrosis progression. We further revealed that GA treatment significantly decreased the expression of mesenchymal cell markers alpha-smooth muscle actin（α-SMA） and vimentin, and up-regulated E-cadherin expression in the kidney, suggesting the suppression of tubular EMT. Further study showed that GA downregulated both TGF-β/Smad and MAPK（ERK,JNK, P38） signaling pathways both in vivo and in ***-A, a GA monomer, was identified as a potent inhibitor on renal fibrosis in TGF-β1-stimulated HK-2 cell *** GA inhibits renal interstitial fibrosis induced by UUO, mostly by suppressing EMT progression and inhibiting TGF-β/Smad and MAPK signaling pathways. Hydrodynamic Shear Stress Affects Cell Growth and Metabolite Production by Medicinal Mushroom Ganoderma lucidum

By investigating the shear effect on submerged cultivation of a traditional Chinese medicinal herb Ganoderma lucidum, a relatively high cell concentration of 13.8 g·L-1 by dry mass was obtained in bioreactor at an impeller tip speed (ITS) of 0.51m·s-1. At an ITS of 0.51,1.02 and 1.53m·s-1, a maximal production titer of intracellular polysaccharide was 2.64, 2.20 and 2.28g·L-1 and that of ganoderic acid was 306, 299 and 273g·L-1, respectively. Under these ITSs, the maximal mean projected area of dispersed hyphae was 3.70, 2.54 and 2.13×104μm2, and that of pellets was 0.91, 0.67 and 0.55mm2, respectively. The information obtained is useful for efficient submerged cultivation of mushrooms on a large scale.

Ganoderma lucidum（G. lucidum） is one of the best-known medicinal basidiomycetes in the world,because it containing a wide range of immuno-modulatory and bioactive compounds. ganoderic acid（GA） is one of the major secondary metabolites in G. lucidum, a basidiomycete with high commercial value. Previous studies have shown that heat stress（HS） increases GA biosynthesis via induced reactive oxygen species（ROS）accumulation in G. lucidum[1] and NO relieves HS-induced GA accumulation in G. lucidum[2]. To determine the mechanism that NO relieves HS-induced GA accumulation in G. lucidum, the effect of NO on the ROS content was examined under heat treatment. The results showed that NO decreased the production of MitROS under heat treatment. Further research found that NO reduced the MitROS content through inhibit the aconitase activity. In the aconitase silenced strains（Aconi）, treatment with NO donor（SNP） showed no obviously effect on the mitROS and GA content relative to the untreated sample. Combined with the prior results, these findings indicate that NO inhibits aconitase activity to reduced mitROS content and thereby relieved HS-induced GA overproduction in Ganoderma *** S-nitrosylation, the incorporation of a nitroso group to a Cys thiol, has been established as a significant route through which NO transmits its global cellular influence. To study the mechanism of the inhibition of the aconitase activity in G. lucidum, the aconitase proteins S-Nitrosylation was ***-switch technology（BST） and mass spectrometric analysis revealed that Acon was S-nitrosylated at amino acid residues, Cys 594. The substitution of Cys 594 with non-nitrosylatable Ser with site-directed mutagenesis causes the abolishes its responsiveness to the NO-reducted enzymatic activity. These findings demonstrate that NO inhibited the activity of aconitase through S-nitrosylation at Cys 594 thereby relieved HS-induced GA biosynthesis in Ganoderma lucidum..
