Objective:To evaluate whether the methanol extract of Codium fragile(MECF) regulates tumor necrosis factor-α(TNF-α)-induced invasion of human breast cancer MDA-MB-231 cells by suppressing matrix metalloproteinase-9(MMP-9).Methods:Reverse transcriptionpolymerase chain reaction(RT-PCR) and western blot analysis were performed to analyze the expression of MMP-9 and nuclear factor-κB(NF-κB) subunits,p65 and p50,and IκB in MDA-MB-231 cells.3-(4,5-Dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide(MTT) assay was used for cell viability.MMP-9 activity and invasion were measured by gelatin zymography and a matrigel invasion assay,respectively.NF- κB activity was measured by an electrophoretic mobility shift assay and luciferase activity.Results:MECF had no effects on cell viability up to a concentration of 100 μg/mL in human breast cancer MDA-MB-231 cells regardless of the presence of TNF-α.MDA-MB-231 cells that were stimulated with TNF-α showed a marked increase of invasion compared to the untreated control,whereas pretreatment with MECF downregulated the TNF-α-induced invasion of MDA-MB-231 cells.Additionally,zymography,western blot analysis,and reverse transcriptase-polymerase chain reaction(RT-PCR) confirmed that MECF decreased TNF-α-induced MMP-9 expression and activity which is a key regulator for cancer invasion.According to an electrophoretic morbidity shift assay,pretreatment with MECF in MDA-MB-231 cells significantly decreased the TNF-α-induced DNA-binding activity of nuclear factor- κB(NF- κB),which is an important transcription factor for regulating cancer invasion-related genes such as MMP-9.Furthermore,treatment with MECF sustained the expression of p65 and p50 in response to TNF-α in the cytosolic compartment.The luciferase assay demonstrated that MECF attenuated TNF-α-induced NF- κB luciferase activity.Conclusion:MECF exhibited its antiinvasive capability by downregulating TNF-α-induced MMP-9 expression,resulting from the suppression of NF- κB activity in the human breast cancer cell line MDA-MB-2

Fragiie X syndrome is the most common form of inherited mental retardation disease, resulting from absent of expression of its disease gene FMR1. To study the function of the fragiie X mental retardation protein (FMRP) through protein/protein interaction, a mouse embryo cDNA library was screened by the yeast two-hybrid system. A clone was found to interact specifically with FMRP. The cDNA of this clone ( Genbank accession number af102875 ) encoded a protein highly homologous to human G/T mismatch-specific DNA thymine glycosylase ( hTDG ). Interactions between various alternatively spliced FMRP isoforms and a series of mTDG deletion proteins were further studied in the yeast two-hybrid system and their interaction amino acid regions were determined. interaction between FMRP and TDG existed inside exon 13 of FMRP ( amino acid residue 397-425 ) and around amino acid residue 122-346 of TDG. These results will be helpful to the study of the biological role of FMRP.

Isolation and genetic charac-terization of a fragile plant mutant in rice (Oryza sativa L.)

The fragile rice mutant was isolated from an M2 population of indica variety Shuang Ke Zao (SKZ) treated with γ-rays, and designated as fp1 {fragile plant 1) because of its fragile leaves and culms. To map FP1 locus, an F2 mapping population was derived from a cross between the fp1 and C-bao, a polymorphic japonic variety. The primary mapping result places the FP1 locus in an interval between two molecular markers, microsatellite marker RM16 (3.1 cM proximal to FP1) and STS marker G144a (9.1 cM distal to FP1) in the centromere region of chromosome 3. A CAPS marker C524a was further developed between RM16 and G144a, with 0.4 cM genetic distances to the FP1 locus, providing a practical starting point for constructing a BAC con-tig spanning the FP1 locus and cloning the fp1 gene. Allelism test demonstrated that fpl is allelic to be 1, a fragile rice mutant reported previously.

The fragile eco-environment is a special type of ecosystem,its response to the change of environmental condi -tions is very susceptive.So it is rather prone to be disturbed under unfav orable conditions.Human activity h as greatly changed the geo-chemical process in the ecosystem,thus caused a series o f positive and negative effects.In t he ecosys-tem,especially in the fragile eco-e nvironment,different systems and r egimes are interconnected and interdetermined.For the suntainable development of ecosystem and the protection and rational utilization of resources,it is of great impor-tance to study these internal relati onships and seek rational regulatio n and control measure.This paper tak es the fragile eco-environment in the west of the Songnen Plain as an example.Based on th e study of the topograph,physiognom y,soil,vegetation and their geographic distribution in the landscape,th e paper explains the structure of the ecologic land-scape and quantifies the ecologic ge o-chemical processes under differe nt landscape conditions.In additio n,the paper al-so tries making coupling analyses of the ecologic succession and the landscape geochemical environment.And in the pa-per,some research results are given.

fragile Watermarking Based on Robust Hidden Information

Block-wise fragile watermarks can be used to reveal maliciously tampered areas in multimedia products. However a forged content containing a cloned fragile watermark can be constructed by using a series of watermarked data. To defeat this type of counterfeit attack, a novel fragile watermarking technique is proposed in which different pseudo-random data are selected for different host products, and the generated fragile watermark is dependent upon the selected information. While inserting the fragile watermark, the pseudo-random information is also robustly embedded into the host data. Because of the difference between the selected information, different watermarked data cannot be used to forge illegal contents containing a valid fragile watermark.

Study on Rural Poverty in Ecologically fragile Areas——A Case Study of the Tibetan Areas in Sichuan Province

This article offers an overview of natural environment, economic development, and social development in the Tibetan areas in Sichuan Province. The major manifestations of the status quo of poverty in the Tibetan areas are as follows: there is a wide range of poverty and a large poor population; the poverty degree is high and it is difficult to alleviate poverty; the natural disasters are frequent and the phenomenon of poverty arising from disasters is serious. We analyze the factors responsible for poverty of farmers and herdsmen in Sichuan Province as follows: first, the natural conditions are inclement and the ecological environment is fragile; second, the social attitudes are stale and the religious influence is deep; third, the development of education lags behind, the human capital inputs are short and the self-development capacity is weak; fourth, the state poverty alleviation and development policy is weakly directed at region; fifth, the Tibetan areas are facing the dilemma of choosing resources endowments or ecological protection. Following proposals are put forward to resolve the poverty issue in the Tibetan areas of Sichuan Province: strengthen infrastructure construction, and underpin self-development capacity of the Tibetan areas in Sichuan Province; reinforce organic connection of social security and poverty alleviation and development, and promote self-development capacity of impoverished farmers and herdsmen in the Tibetan areas; strengthen policy support, and improve self-development policy and mechanism of the Tibetan areas in Sichuan Province; drive the popularization of technology and culture in the Tibetan areas, and update the development concept of farmers and herdsmen in the Tibetan areas; accelerate the development of competitive industries in Tibetan areas, and promote economic development capacity of the Tibetan areas.

Aberrant crypt focus and fragile histidine triad protein in sporadic colorectal carcinoma

AIM:To characterize aberrant crypt focus (ACF) in adjoining mucosa in sporadic colorectal carcinoma and to evaluate fragile histidine triad (Fhit) protein and Ki67. METHODS:ACF was identified grossly and classified histologically in 75 resected specimens. ACF was typed into hyperplastic ACF (HACF) and dysplastic ACF (DACF). Sections of ACF, carcinoma and normal colonic mucosa as control were studied for Fhit and Ki67 expressions by immunohistochemistry and were grouped according to staining intensity and the number of positive stained cells observed in different histological groups. Comparison was done between the different groups by Pearson's χ 2 test and γ test for the ordinal data. P value < 0.05 was considered as significant.RESULTS:Age range was 40 to 86 years in males (mean = 43.36) and 45 to 70 years in females (mean = 56). HACF was identified in all cases studied in the non-tumorous colonic mucosa; ACF was observed as non-contiguous scattered foci, which supports the hypothesis of acquisition of single focus monoclonality by colonic epithelial cells in tumor generation. Twenty-four (32%) had DACF and were observed as closure to carcinoma foci. Intensity of Fhit expression:(1) HACF 40% exhibited strong intensity, similar to normal, moderate in 36% and weak in 24%; (2) DACF strong in 25%, moderate in 37.5% and weak in 37.5%; and (3) carcinoma negative in 16%, strong in 43% and moderate and weak in 28.5% each. Significant difference was observed in intensity of the Fhit protein expressions by HACF and DACF (P < 0.05). Tumor in older patients showed a stronger Fhit intensity compared to younger patients (P = 0.036). Vegetarian diet intake and nonsmokers showed stronger Fhit intensities. Advanced stage tumor, non-vegetarian diet and younger age was associated with loss of Fhit protein. Ki67 positivity was an extended crypt pattern in HACF and DACF showed extension up to the neck region of the crypts and surface epithelium. Prevalence of fragile X syndrome in males and females in Indonesia

AIM: To investigate the prevalence of fragile X syndrome(FXS) in intellectually disabled male and female Indonesians.METHODS: This research is an extension of a previously reported study on the identification of chromosomal aberrations in a large cohort of 527 Indonesians with intellectual disability(ID). In this previous study,87 patients had a chromosomal abnormality, five of whom expressed fragile sites on Xq27.3. Since FXS cannot always be identified by cytogenetic analysis, molecular testing of the fragile X mental retardation 1 CGG repeat was performed in 440 samples. The testing was also conducted in the five previously identified samples to confirm the abnormality. In total, a molecular study was conducted in 445 samples(162 females and 283 males).RESULTS: In the cohort of Indonesian ID population, the prevalence of FXS is 9/527(1.7%). The prevalence in males and females is 1.5%(5/329) and 2%(4/198), respectively. Segregation analysis in the families and X-inactivation studies were performed. We performed the first comprehensive genetic survey of a representative sample of male and female ID individuals from institutions and special schools in Indonesia. Our findings show that a comprehensive study of FXS can be performed in a developing country like Indonesia where diagnostic facilities are limited.CONCLUSION: The prevalence of FXS is equal in females and males in our study, which suggests that the prevalence of FXS in females could be underestimated.
