To investigate the effect of basal hormone levels including follicle stimulating hormone(FSH) and estradiol(E2) and age on the pregnancy outcomes in women undergoing in vitro fertilization and embryo transfer(IVF-ET), we designed a retrospective cohort analysis and collected information of patients undergoing IVF between January 2014 and April 2017. We found that young women had much better pregnancy outcomes than older women regardless of the hormone levels. The pregnancy outcomes declined in young women with high basal E2, even though they had normal FSH. Older women with higher FSH had a worse outcome of pregnancy, especially with increased E2 level. The results suggest that a combination of FSH, E2 and age could effectively predict the pregnancy outcome for women undergoing IVF-ET, and we should encourage infertile women with diminished ovarian reserve, especially young infertile women, to try the assisted reproduction as early as possible. After 40-yearold, the cost-benefit ratio should be considered according to the ovarian function.

The hair follicle is a biological oscillator that alternates growth,regression,and rest phases driven by the sequential activation of the proliferation differentiation programs of resident stem cell *** activation of hair follicle stem cell niches and subsequent entry into the growing phase is mainly regulated by Wnt/β-catenin signalling,while regression and resting phases are mainly regulated by Tgf-β/Bmp/Smad activity.A major question still unresolved is the nature of the molecular switch that dictates the coordinated transition between both signalling *** we have focused on the role of Endoglin(Eng),a key co-receptor for members of the Tgf-β/Bmp family of growth *** an Eng haploinsufficient mouse model,we report that Eng is required to maintain a correct follicle cycling pattern and for an adequate stimulation of hair follicle stem cell *** further report thatβ-catenin binds to the Eng promoter depending on Bmp ***,we show thatβ-catenin interacts with Smad4 in a Bmp/Eng-dependent context and both proteins act synergistically to activate Eng promoter *** observations point to the existence of a growth/rest switching mechanism in the hair follicle that is based on an Eng-dependent feedback cross-talk between Wnt/β-catenin and Bmp/Smad signals.

Objective:To analyze the dynamic expression of Wnt family member 5A(Wingless-type MMTV integration Wnt site family,member 5a)in murine hair cycle and its inhibitory effects on follicle in ***:Situ hybridization in full-thickness skin was used to observe the change of mouse protein expression in different growth stages,and Ad-Wnt5a was injected after defeathering to observe the hair follicle growth in ***:The Wnt5a mRNA was expressed at birth,and was firstly increased then decreased along with the progress of the hair *** reached the peak in advanced stage of growth cycle(Pfollicle cycle,and has obvious inhibitory effects on the growth of hair follicle in vivo,indicating that it is antagonistic to Wnts pathway and interferes the growth of follicle together.

The ovary contains four morphological components: (1) the ovarian wall, (2) the reproductive epithelium, (3) the cellular layer containing oocytes, oogonia (especially for early-developing ovary) and follicle cells, and (4) the extensions of the ovarian wall. The ovarian wall and its extensions consist of blood vessels, sinuses, muscle cells and others. The extensions of the ovarian wall project into among the follicles and insert on the thick basal membrane of each follicle. From inside to outside, the follicles are composed of four parts: (1) the oocyte, (2) the perivitelline space, (3) the follicle cells, and (4) the basal membrane. The surface of the oocyte during vitellogenesis is folded into numerous long microvilli that project into the perivitelline space between the oocyte surface and the bace of the follicle cell layer. In addition, the plasma membrane of the vitellogenic oocyte contains many pinocytotic pits. The perivitelline space is engorged with with more electron- denser material as the development of the follicle. The inclusion of perivitelline space in the mature follicle is named specially as the chorine. The chorion is composed of two region, a thinner exochorion and a thicker endochorion containing electron-dense granular material. The follicle cell layer is composed of a single layer of polygonal follicle cells which exhibit higher synthetic activity. The synthetic product of the follicle cell layer is one scarce for the inclusion of the perivitelline space. The structures of the ovary and ovarian follicle in T. orientalis show that the exogenously biosyn- thetic yolk plays important roles in the vitellogenesis.

Vacuolar protein sorting 36(VPS36),a protein primarily known for its role in the Endosomal Sorting Complex Required for Transport pathway,has recently been shown to be linked to chicken *** research showed that Vps36 is significantly downregulated in sexually mature chicken ovaries compared to immature *** this study,using real-time quantitative RT-PCR,we investigated the expression pattern of Vps36 and its head-to-head gene Ckap2 mRNA in chicken *** white follicles were found to have significantly higher expression of Vps36 and Ckap2 mRNA than any other sized follicles(Pfollicles,the expression of Ckap2 in theca layers was slightly higher than in granulosa *** of small yellow follicles with folliclestimulating hormone and estradiol resulted in a marked decrease of both Vps36 and Ckap2 mRNA(Pfollicles and are involved in chicken follicle development.

Objective:To evaluate the effects of pellucid zone 3 monoclonal antibodies against B-cell lymphoma 2 (BCL-2) expression and mice follicle diameter at various time ***:The animal model of this study was 36 Balb/c mice (Mus musculus). A true experimental design was used with a post-test only control group approach. BCL-2 expression was observed using immunohistochemistry, while the follicle diameter was observed by haematoxylin-eosin staining. The data was analyzed using nested ANOVA to compare the results of the mean expression of BCL-2 on the 5th and 20th day of observation in the pre-antral and antral follicle between the control and treatment ***:No significant differences were found in BCL-2 gene expression. There were also no significant differences in BCL-2 expression on the 10th day of pre-antral follicle analysis. Moreover, there were no significant differences between the mean follicle diameter on the 5th, 10th, and 20th day of pre-antral and antral follicle development between the control and treatment groups. The addition of bovine pellucid zone 3 (bZP3) monoclonal antibodies on the 5th and 20th day of observation did not decrease the expression of BCL-2 gene in the pre-antral and antral follicle of mice. Administering bZP3 monoclonal antibodies on the 10th day of observation did not affect BCL-2 expression in the pre-antral follicle but did decrease BCL-2 expression in the antral follicle. Supplying bZP3 monoclonal antibodies on the 5th, 10th and 20th day did not affect the diameter of pre-antral and antral follicles of the ***: The monoclonal antibodies bovine zona pelusida 3 has the potential to be developed as a safe immunocontraception preparation.

The ultrastructure of follicle cells in the ovary at different developmental stages of Branchiostoma has been observed in detail with a transmission electron microscope. The results indicate that only one kind of follicle cell exists with structural features related to steroid hormone biosynthesis: (i) oval or round mitochondria with tubules; (ii) smooth surfaced endoplasmic reticulum; (iii) several large lipid droplets in the cytoplasm; (iv) a well de-veloped Golgi complex and tubular rough surfaced endoplasmic reticulum, as can be found in mammalian theca interna cells. In addition, as steroid hormone synthesizing cells, they obviously play an important role in the phagocytosis of relict gametes and cellular debris and may have a nutritive function for the oocytes. They can produce abundant secre-tory granules in stages III-IV ovaries. In mature ovaries they transform into flat epithelial cells with numerous micro-filaments which may play a role in ovulation.
