focal ischemia due to reduction of cerebral blood flow(CBF),creates 2 zones of damage:the core area,which suffers severe damage,and penumbra area,which surrounds the core and suffers intermediate levels of ***:A novel method is introduced,which evaluates mitochondrial function in the core and in the penumbra,during focal cerebral ***:Wistar rats underwent focal cerebral ischemia by middle cerebral artery occlusion(MCAO)for 60 minutes,followed by 60 minutes of *** function was assessed by a unique Multi-Site—Multi-Parametric(MSMP)monitoring system,which measures mitochondrial NADH using fluorometric technique,and CBF using Laser Doppler Flowmetry(LDF).Results:At the onset of occlusion,CBF dropped and NADH increased significantly only in the right *** levels were significantly lower and NADH significantly higher in the core than in the *** reperfusion,CBF and NADH recovered correspondingly to the intensity of ***:Application of the MSMP system can add significant information for the understanding of the cerebral metabolic state under ischemic conditions,with an emphasis on mitochondrial function.

Backgrounds: We showed previously that 2-(2-benzofu-ranyl)-2-imidazoline(2-BFI), a ligand to type 2 imidazoline receptor(I2 R) exerts neuroprotective effects in ischemia stroke via an unknown mechanism. The present study was to investigate whether 2-BFI can protect the neurovascular unit(NVU) using a rat model of 90 min focal cerebral ***: Rats were randomly divided into three groups: the sham-operated group;the vehicle control group and the 2-BFI group which received 2-BFI(3 mg/kg) immediately after the start of middle cerebral artery occlusion(MCAO). Neurological deficit score, infarct size, apoptosis level, brain water content and Evans Blue extravasation were assessed at 24 h after stroke. Expressions of occludin and zonula occludens 1(ZO-1), collagen Ⅳ, aquaporin-4(AQP-4), matrix metalloproteinase-9(MMP-9) and MMP-2 were assessed by Western ***: 2-BFI treatment was associated with significant improvement of neurological performance and decreased infarct volume at 24 h after stroke. Apoptosis level reduced significantly by 2-BFI compared to the vehicle group(34.3±5.4% vs 56.1±7.9%, p<0.05). Significant decreased of brain water content(79.5±2.6% vs 84.62±2%, p<0.05) and Evans Blue extravasation(1.2±0.5 vs 2.5±0.41μg/g, p<0.05) of ipsilateral hemisphere was observed in 2-BFI group compared to vehicle group. Expressions of occludin, ZO-1 and collagen Ⅳ were significantly higher while MMP-9 level significantly lower in 2-BFI group. AQP-4 and MMP-2 showed no difference between 2-BFI and the vehicle ***: These results suggest that the neuroprotective effects of 2-BFI in acute ischemic brain damage are at least partly due to the drug’s ability to improve the functions of NVU.

Optimal electroacupuncture frequency for maintaining astrocyte structural integrity in cerebral ischemia

The astrocyte is a critical regulator of neuronal survival after ischemic brain injury. Electroacupuncture may be an effective therapy for cerebral ischemia, as electroacupuncture frequency can affect the structural integdty of astrocytes. In this study, a rat model of middle cerebral artery occlusion established using the modified thread embolism method was treated with electroacupuncture of the bilateral Quchi （Llll） and Zusanfi （ST36） at 15, 30, and 100 Hz frequencies. Behavioral testing, immunohistochemistry and electron microscopy were used to explore the effect of these electroacupuncture frequencies used on maintaining the structural integrity of ischemic brain tissue. Compared with the model and 100 Hz electroacupuncture groups the 15 and 30 Hz electroacupuncture groups displayed decreased neurological deficit scores, as evaluated by the ＂Longa＂ method, significantly increased glial fibrillary acidic protein expression, and alleviated ultrastructural damage of astrocytes at the edge of the infarct. Our experimental findings indicate that 15 and 30 Hz electroacupuncture intervention can favorably maintain the structural integrity of astrocytes and play a protective role in cerebral ischemic injury. Astrocyte structural integrity may be the mechanism underlying acupuncture production of ischemic tolerance

Adenovirus-mediated hypoxia-inducible factor-1 alpha gene transfer induces angiogenesis and neurogenesis following cerebral ischemia in rats

BACKGROUND： Hypoxia-inducible factor-1 （HIF-1） accumulates under conditions of hypoxia. HIF-1α target genes have pleiotropic effects on neurogenesis, neuroprotection and angiogenesis in the brain. OBJECTIVE： To investigate whether a recombinant adenovirus carrying HIF-1α can increase the expression of HIF-I a in vivo and thus promote angiogenesis and neurogenesis in a rat model of focal cerebral ischemia. DESIGN, TIME AND SETTING： The randomized, controlled experiment was performed at the Department of Neurobiology, Third Military Medical University of Chinese PLA from September 2006 to October 2007. MATERIALS： 68 healthy adult male Sprague-Dawley （SD） rats, weighing 230-250 g, were used. HIF-I a antibody was purchased from Wuhan Boster Company. Vascular endothelial growth factor （VEGF） antibody was purchased from Santa Cruz Biotech Company. METHODS： All 68 rats were induced with a transient middle cerebral artery occlusion （MCAO）, according to the method of intra-luminal vascular occlusion. 54 rats, in which MCAO was successfully induced, were randomly divided into adenovirus （Ad） group and recombinant adenovirus with HIF-1α gene （Ad-HIF-1α ） group （27 rats for each group）. Rats were injected with 10 μL Ad （Ad group） or Ad-HIF-1α （Ad-HIF-1α group） into the lateral ventricle, 1 day after MCAO induction. MAIN OUTCOME MEASURES： Reverse transcription polymerase chain reaction was used to measure the expression of HIF-1α and of VEGF. Immunohistochemistry was used to detect the localization of HIF-1α, VEGF and factor Ⅷ in ischemic penumbra. Rat newborn nerve cells were labeled with 5-bromodeoxyuridine （BrdU） after ischemia. BrdU/neurofilament 200 （NF200） and BrdU/glial fibrillary acidic protein （GFAP） double labeled immunofluorescent histochemistry was used to identify the differentiation of newborn cells. Acupuncture for stroke has been used in China for over 2,000 years and nowadays is increasingly practiced elsewhere in the ***,previous studies had conflicting findings on the results of ***,we conducted a systematic review and meta-analysis to assess the current evidence for the effect of Baihui(GV20)-based scalp acupuncture in animal models of focal cerebral *** databases from the inception of each database up to June 2013 were electronically *** outcomes were infarct size and neurobehavioral ***,54 studies involving 1816 animals were identified describing ***-analysis results showed that twelve studies reported significant effects of Baihui(GV20)-based scalp acupuncture for improving infarct volume compared with middle cerebral artery occlusion group(P<0.01),and thirty-two studies reported significant effects of Baihui(GV20)-based scalp acupuncture for improving the neurological function score when compared with the control group(P<0.01).In conclusion,Baihui(GV20)-based scalp acupuncture could improve infarct volume and neurological function score and exert potential neuroprotective role in experimental ischemic stroke.

To investigate the effects of 4-methyl-2-{[4-(toluene-4-sulfonyl)-thiomorpholine-3-carbonyl]-amino}-pentanoic acid isopropyl ester(HD5-6)against cerebral ischemia in rodents,the models with global and focal ischemia were induced by bilateral common carotid artery occlusion plus hypotension(BCAOH)and permanent cerebral artery occlusion(p-MCAO)in mice(n=10–12 per group in BCAOH;n=8 per group in p-MCAO)and rats(n=10-11 in each group).HD5-6 prolonged lifetime and improved neurological *** deficits score in HD5-6(30 mg/kg)decreased ***(MDA)in HD5-6-treated mice with ischemia considerably *** infarction volume of the HD5-6-treated rats with MCAO-induced ischemia decreased significantly in the high dose group(Pischemia in rodents and the action mechanism may involve anti-oxidation and neurogenesis.
