Histochemistry of microinfarcts in the mouse brain after injection of fluorescent microspheres into the common carotid artery

The mouse model of multiple cerebral infarctions,established by injecting fluorescent microspheres into the common carotid artery,is a recent development in animal models of cerebral ischemia.To investigate its effectiveness,mouse models of cerebral infarction were created by injecting fluorescent microspheres,45–53μm in diameter,into the common carotid artery.Six hours after modeling,fluorescent microspheres were observed directly through a fluorescence stereomicroscope,both on the brain surface and in brain sections.Changes in blood vessels,neurons and glial cells associated with microinfarcts were examined using fluorescence histochemistry and immunohistochemistry.The microspheres were distributed mainly in the cerebral cortex,striatum and hippocampus ipsilateral to the side of injection.Microinfarcts were found in the brain regions where the fluorescent microspheres were present.Here the lodged microspheres induced vascular and neuronal injury and the activation of astroglia and microglia.These histopathological changes indicate that this animal model of multiple cerebral infarctions effectively simulates the changes of various cell types observed in multifocal microinfarcts.This model is an effective,additional tool to study the pathogenesis of ischemic stroke and could be used to evaluate therapeutic interventions.This study was approved by the Animal Ethics Committee of the Institute of Acupuncture and Moxibustion,China Academy of Chinese Medical Sciences(approval No.D2021-03-16-1)on March 16,2021.

Establishment of multiplexed,microsphere-based flow cytometric assay for multiple human tumor markers

Aim:The multiplexed,microsphere-based flow cytometric assay (MFCA) for mul-tiple human tumor markers was established for the early screening and detectionof suspected cancer patients.Methods:Covalent coupling of capture antibodiesdirected against their respective tumor markers to fluorescent microspheres wasperformed by following the protocols recommended by a commercial corporationwith some modifications.The coupling efficiency and cross-reactivity were iden-tified by the Luminex 100 system and associated software.The standard curvewas constructed by using serial dilution of recombinant tumor marker standardsand was validated by comparison with ELISA for quantifying the tumor markers inserum samples.Results:The identifications revealed that the coupling proce-dures were successful without non-specific cross-reactivity and the standardcurve was highly efficient.However,it was necessary to ensure the quality con-trol of the coupling process since slight variations in the coupling procedurescould profoundly affect the density of capture reagents coupled to themicrospheres and consequently adversely affect the assay precision.In additionto its multi-analyte capability,the MFCA system had definite advantages,such ashigher reproducibility,greater dynamic range of measurement,and considerablyless preparation time and labor over the conventional"gold standard",which wasthe ELISA.Conclusion:The successful establishment of the MFCA system forthe simultaneous detection of multiple tumor markers will provide the foundationfor the further study of clinical applications.

Campylobacter jejuni is a worldwide foodborne pathogen recognized as a leading cause of human gastrointestinal enteritis.A rapid, sensitive, and specific method is required to monitor food and water in cases of contamination by this pathogen.This report presents a novel immunochromatographic test(ICT) using fluorescent microspheres labeled with specific polyclonal antibodies of C.jejuni as the capture reagent dispensed onto the conjugate pad.Polyclonal antibodies against the outer membrane protein PEB1 of C.jejuni were used as the detective reagent at the test line, whereas the goat anti-rabbit Ig G was used on the control line.PEB1 was obtained by gene cloning and expression to prepare its antibody.In this study, for the first time we report a simple and rapid ICT for detecting C.jejuni with a detection limit of 106 CFU/ml.
