The reducing terminal of marine sulfated polysaccharide (911) was used to selectively insert primary and secondary amines by reductive amination. 911 was bound to 4-(2-aminoethylphenol), followed by labeling the 911-Tyr with fluorescein-5-isothiocyanate (FITC) at the secondary amino group. High-performance size-exclusion chromatography (HPSEC), ultraviolet/visible (UV/Vis) spectroscopy and nuclear magnetic resonance spectroscopy (NMR) demonstrated the binding of tyramine to 911 and the labeling of FITC to 911-Tyr respectively. Furthermore, H-NMR spectra and UV/Vis spectra revealed about 60％ binding of tyramine to 911 and about 80％ labeling of FITC to 911-Tyr. The anticoagulant activity of labeled fluorescent compound exhibited 15u/mg equivalent to that of unlabled 911. In addition, labeled compound demonstrated to fluorescently stain lymphocytes without any cytotoxity. Taken together, the selectively "end point attached" method might be widely used for fluorecent labeling to those polysaccharides with reducing terminal.

Highly Sensitive fluorescent-labeled Probes and Glass Slide Hybridization for the Detection of Plant RNA Viruses and a Viroid

In this study, a modified method of the conventional RNA dot-blot hybridization was established, by replacing32p labels with CY5 labels and replacing nylon membranes with positive-charged glass slides, for detecting plant RNA viruses and a viroid. The modified RNA dot-blot hybridization method was named glass slide hybridization. The optimum efficiency of RNA binding onto the surfaces of activated glass slide was achieved using aminosilane-coated glass slide as a solid matrix and 5×saline sodium citrate （SSC） as a spotting solution. Using a CY5-labeled DNA probe prepared through PCR amplification, the optimized glass slide hybridization could detect as little as 1.71 pg of tobacco mosaic virus （TMV） RNA. The sensitivity of the modified method was four times that of dot-blot hybridization on nylon membrane with a32P-labeled probe. The absence of false positive within the genus Potyvirus [potato virus A, potato virus Y （PVY） and zucchini yellow mosaic virus] showed that this method was highly specific. Furthermore, potato spindle tuber viroid （PSTVd） was also detected specifically. A test of 40 field potato samples showed that this method was equivalent to the conventional dot-blot hybridization for detecting PVY and PSTVd. To our knowledge, this is the first report of using dot-blot hybridization on glass slides with fluorescent-labeled probes for detecting plant RNA viruses and a viroid.

Synthesis of photolabile dUTP analogues and their enzymatic incorporation for DNA labeling

Two new photolabile nucleotide analogues with furan-fused deoxyuridine were synthesized through Sonogashira *** enzymatic incorporation into DNA was evaluated with two DNA polymerases(Taq and Deep vent exo-)by polymerase chain reaction(PCR).Deep vent exo-recognized both nucleotides as substrates for primer extension,while Taq was much less *** irradiation of PCR products released the amino and carboxyl moieties of *** labeling with fluorescein isothiocyanate for a long DNA construct with F-dUnTP incorporation was successfully achieved.

Shaping Small Bioactive Molecules to Untangle Their Biological Function： A Focus on fluorescent Plant Hormones

Modern biology overlaps with chemistry in explaining the structure and function of all cellular processes at the molecular level. Plant hormone research is perfectly located at the interface between these two disciplines, taking advantage of synthetic and computational chemistry as a tool to decipher the complex biological mechanisms regulating the action of plant hormones. These small signaling molecules regulate a wide range of developmental processes, adapting plant growth to ever changing environmental conditions. The synthesis of small bioactive molecules mimicking the activity of endogenous hormones allows us to unveil many molec- ular features of their functioning, giving rise to a new field, plant chemical biology. In this framework, fluores- cence labeling of plant hormones is emerging as a successful strategy to track the fate of these challenging molecules inside living organisms. Thanks to the increasing availability of new fluorescent probes as well as advanced and innovative imaging technologies, we are now in a position to investigate many of the dynamic mechanisms through which plant hormones exert their action. Such a deep and detailed comprehension is mandatory for the development of new green technologies for practical applications. In this review, we sum- marize the results obtained so far concerning the fluorescent labeling of plant hormones, highlighting the basic steps leading to the design and synthesis of these compelling molecular tools and their applications.

Mesencephalic Progenitors Can Differentiate into TH-Positive Neurons and Improve Dysfunction in PD Rats

This study investigated the possibility of using mesencephalic progenitors (MPs) in the treatment of Parkinson's disease (PD). MPs were prepared from E 11 13 rats and proliferated in the serum free medium with basic fibroblast growth factor (bFGF) for 10 days. Cells were then collected and implanted into the striatum only-single grafts, or simultaneously into the substantia nigra (SN) and the striatum-double grafts. Twelve weeks after transplantation, 1,1' dioctadecyl 3,3,3',3' tetramethylindocarbocyanine perchlorate (DiI), a fluorescent dye, was microinjected into the ipsilateral striatum. The results show that double grafted MPs have more potent effects on rotational behavior than single grafted MPs. Injection of the retrograde tracer DiI into the striatum results in fluorescent labeled cells within the intranigral grafts only in double graft subjects. These data suggest that MP transplants not only can improve rotational behavior, but also help to reestablish nigrostriatal connections, so that such transplants may become an efficient way of treating PD.
