Transcutaneous electrical acupoint stimulation benefits postoperative pain relief of oocyte retrieval:A randomized controlled trial

Background Transvaginal oocyte retrieval is frequently followed by adverse events related to anesthesia and the procedure.Some research showed that transcutaneous electrical acupoint stimulation(TEAS)can relieve intraoperative pain and postoperative nausea.Objective This study examined whether TEAS can alleviate pain and relieve adverse symptoms after oocyte retrieval.Design,setting,participants and interventions Altogether 128 patients were randomly divided into the TEAS group and the mock TEAS group.The two groups received a 30-minute-long TEAS or mock TEAS treatment that began 30 min after oocyte retrieval.Main outcome measures The primary outcome was the visual analog scale(VAS)pain score.Secondary outcomes were pressure pain threshold,McGill score,pain rating index(PRI),present pain intensity(PPI),VAS stress score,VAS anxiety score,and postoperative adverse symptoms.Results The baseline characteristics of the two groups were comparable(P>0.05).The VAS pain scores of the TEAS group were lower than those of the mock TEAS group at 60 and 90 min after oocyte retrieval(P0.05).The TEAS group was superior to the mock TEAS group for relieving postoperative adverse symptoms(P<0.05).Conclusion TEAS treatment can relieve postoperative pain and postoperative adverse symptoms for patients undergoing oocyte retrieval.

Manure substitution improves maize yield by promoting soil fertility and mediating the microbial community in lime concretion black soil

Synthetic nitrogen(N)fertilizer has made a great contribution to the improvement of soil fertility and productivity,but excessive application of synthetic N fertilizer may cause agroecosystem risks,such as soil acidification,groundwater contamination and biodiversity reduction.Meanwhile,organic substitution has received increasing attention for its ecologically and environmentally friendly and productivity benefits.However,the linkages between manure substitution,crop yield and the underlying microbial mechanisms remain uncertain.To bridge this gap,a three-year field experiment was conducted with five fertilization regimes:i)Control,non-fertilization;CF,conventional synthetic fertilizer application;CF_(1/2)M_(1/2),1/2 N input via synthetic fertilizer and 1/2 N input via manure;CF_(1/4)M_(3/4),1/4 N input synthetic fertilizer and 3/4 N input via manure;M,manure application.All fertilization treatments were designed to have equal N input.Our results showed that all manure substituted treatments achieved high soil fertility indexes(SFI)and productivities by increasing the soil organic carbon(SOC),total N(TN)and available phosphorus(AP)concentrations,and by altering the bacterial community diversity and composition compared with CF.SOC,AP,and the soil C:N ratio were mainly responsible for microbial community variations.The co-occurrence network revealed that SOC and AP had strong positive associations with Rhodospirillales and Burkholderiales,while TN and C:N ratio had positive and negative associations with Micromonosporaceae,respectively.These specific taxa are implicated in soil macroelement turnover.Random Forest analysis predicted that both biotic(bacterial composition and Micromonosporaceae)and abiotic(AP,SOC,SFI,and TN)factors had significant effects on crop yield.The present work strengthens our understanding of the effects of manure substitution on crop yield and provides theoretical support for optimizing fertilization strategies.

Osmoregulation determines sperm cell geometry and integrity for double fertilization in flowering plants

Distinct from the motile flagellated sperm of animals and early land plants,the non-motile sperm cells of flowering plants are carried in the pollen grain to the female pistil.After pollination,a pair of sperm cells are delivered into the embryo sac by pollen tube growth and rupture.Unlike other walled plant cells with an equilibrium between internal turgor pressure and mechanical constraints of the cell walls,sperm cells wrapped inside the cytoplasm of a pollen vegetative cell have only thin and discontinuous cell walls.The sperm cells are uniquely ellipsoid in shape,although it is unclear how they maintain this shape within the pollen tubes and after release.In this study,we found that genetic disruption of three endomembrane-associated cation/H+exchangers specifically causes sperm cells to become spheroidal in hydrated pollens of Arabidopsis.Moreover,the released mutant sperm cells are vulnerable and rupture before double fertilization,leading to failed seed set,which can be partially rescued by depletion of the sperm-expressed vacuolar water channel.These results suggest a critical role of cell-autonomous osmoregulation in adjusting the sperm cell shape for successful double fertilization in flowering plants.

Glycosylphosphatidylinositol-anchored sperm hyaluronidases have long been believed to assist in sperm penetration through the cumulus-oocyte complex(COC);however,their role in mammalian fertilization remains unclear.Previously,we have shown that hyaluronidase 5(Hyal5)/Hyal7 double-knockout(dKO)mice produce significantly fewer offspring than their wild-type(WT)counterparts because of defective COC dispersal.Male infertility is mainly caused by a low sperm count.It can be further exacerbated by the deficiency of sperm hyaluronidase,which disperses the cumulus cells of the outer layer of the COC.In the current study,we evaluated the effects of a low count of Hyal-deficient sperm and conditions of ovulated oocytes on the fertilization rate using a mouse model.Our results demonstrated that a low sperm count further decreases the in vitro fertilization(IVF)rate of Hyal-deficient dKO spermatozoa.In addition,the dKO spermatozoa resulted in a fertilization rate of 12.5%upon fertilizing COCs with a thick cumulus layer,whereas the IVF rate was comparable to that of WT spermatozoa when oocytes with a thin or no cumulus layer were fertilized.Finally,we proved that the IVF rate of dKO spermatozoa could be recovered by adding rat spermatozoa as a source of sperm hyal.Our results suggest that a deficiency of proteins involved in fertilization,such as sperm hyal,has a vital role in fertilization.

Effect of pre-IVF incubation in maturation medium on oocyte maturity,fertilization,embryonic development,and clinical outcomes following embryo transfer

Objective:Collected human cumulus-oocyte complexes(COCs)are usually inseminated after 4 to 6 hours in in vitro fertilization(IVF)laboratories.The purpose of this study was to determine the effect of short-term pre-IVF incubation in culture medium on subsequent oocyte maturation,fertilization,and embryonic development,as well as clinical outcomes.Methods:Sixty patients were divided randomly into 2 groups,pre-IVF incubation for 5 hours:1)with(+)the designed oocyte maturation medium;2)without(-)the designed oocyte maturation medium(transferred directly to fertilization medium for 5 hours before insemination).Oocyte maturation and fertilization were assessed,and the rate of cleavage and good quality embryos were evaluated between the 2 groups on days 2 and 3,respectively.Blastocyst development was based on the remaining number of embryos on day 3,continuously cultured to day 5 after embryo transfer or frozen on day 3,and was compared between the 2 groups.Clinical pregnancy,implantation,and miscarriage rates were also compared.Results:Oocyte maturation rates did not differ between groups(85.8±14.1%vs.90.7±9.1%).However,the range of oocyte maturation rates(58.3%-100.0%)for each patient was significantly higher in the(-)group than in the(+)pre-incubation group(71.4%-100.0%).There were no differences in fertilization rates(89.9±10.0%vs.86.5±12.2%)and good quality embryos(70.8±19.1%vs.62.1±23.7%)between groups;however,the blastocyst development rates were significantly different between groups(73.1±20.1%vs.58.8±18.2%,P<0.05).Nevertheless,clinical pregnancy(62.5%vs.61.1%)and implantation(46.9%vs.47.2%)rates did not differ between groups.Conclusions:These results indicate that a short pre-IVF incubation time in the designed culture medium promotes oocyte maturation and embryonic development,suggesting that short pre-IVF incubation of COCs in the designed culture medium may be important for subsequent final oocyte maturation and early embryonic development.

Age-related declines in fertilization success have been reported for a wide range of species.The fertilization of parasitic wasps is closely related to egg production and sperm storage.Aenasius bambawalei(Hymenoptera:Encyridae)is a key parasitic wasp of the important invasive mealybug Phenacoccus solenopsis(Hemiptera:Pseudococcidae).The female offspring ratio of this parasitic wasp was declined with parental age in mass rearing under laboratory conditions.To investigate the molecular mechanisms involved in the reproduction of A.bambawalei,an extensive analysis of the impact of age on transcriptome profile of mated ovaries of this wasp was performed by comparing the gene expression profiles of various maternal ages:the early stage(ES),the intermediate stage(IS),and the advanced stage(AS).In total,358 differentially expressed genes were identified,with 17.60%(63 genes)of the changes associated with greater expression in fertilization.Moreover,the expression of serine protease 47 precursor,serine protease inhibitor 3/4,glucose dehydrogenase,fatty acyl-CoA reductase 1-like,major royal jelly,and acyl-CoA delta(11)desaturase-like was detected by real-time fluorescence quantitative(RT-qPCR).The results showed that fertilization related genes exhibited a stage-specific pattern.Egg production and sperm storage genes in A.bambawalei were significantly modified in the transcriptome,providing a starting point for the genetic dissection of fertilization.

Effects of fertilization on Soil CO_(2) Efflux in Chinese Hickory(Carya cathayensis)Stands

Chinese hickory(Carya cathayensis Sarg.)is a popular nut tree in China,but there is little information about the influences of fertilization on soil CO_(2) efflux and soil microbial biomass.This study evaluated the short-term effects of different fertilizer applications on soil CO_(2) efflux and soil microbial biomass in Chinese hickory stands.Four fertilizer treatments were established:control(CK,no fertilizer),inorganic fertilizer(IF),organic fertilizer(OF),and equal parts organic and inorganic N fertilizers(OIF).A field experiment was conducted to measure soil CO_(2) effluxes using closed chamber and gas chromatography techniques.Regardless of the fertilization practices,soil CO_(2) effluxes of all the treatments showed a similar temporal pattern,with the highest value in summer and the lowest in winter.The mean annual soil CO_(2) efflux in the IF treatment was significantly higher than that in the CK,OIF,and OF treatments.There was no significant difference in soil CO_(2) efflux between the OIF,OF,and CK treatments.Soil CO_(2) effluxes were significantly affected by soil temperature.Soil dissolved organic carbon(DOC)was positively correlated with soil CO_(2) efflux only in the CK treatment.Regression analysis,including soil temperature,moisture,and DOC,showed that soil temperature was the primary factor influencing soil CO_(2) effluxes.Both OF and OIF treatments increased concentrations of soil microbial biomass carbon(MBC)and microbial biomass nitrogen(MBN),but decreased the ratio of MBC:MBN.These results reveal that applying organic fertilizer,either alone or combined with inorganic fertilizer,may be the optimal strategy for mitigating soil CO_(2) emission and improving soil quality in Chinese hickory stands.

Impact of gamete health on fertilization and embryo development: An overview

A genetically and functionally proficient gamete is essential for normal fertilization and embryonic development.Any change in gamete health affects fertilization and subsequent events,including embryonic development,implantation,and successful pregnancy.This present review focuses on the role of gamete health on fertilization and embryo development.Several conventional and advanced methods are used to evaluate the morphology and functions of gametes.The abnormal spermatozoa adversely affect fertilization events,which results in reduced cleavage/blastocyst/implantation and pregnancy rate during assisted reproductive techniques.Poor oocyte quality is also one of the reasons for infertility,although the oocyte has an innate capacity to repair a certain amount of abnormality of both oocyte and spermatozoa.Therefore,oocyte health carries more responsibilities during fertilization events.The gamete,either spermatozoa or oocyte,should have optimum morphological and functional health to fertilize and develop a competent embryo successfully.Thus,it is of prime importance to consider the gamete health parameters while dealing with infertility.

Impact of fertilization on chestnut growth, N and P concentrations in runoff water on degraded slope land in South China

Growing fruit trees on the slopes of rolling hills in South China was causing serious environmental problems because of heavy application of chemical fertilizers and soil erosion. Suitable sources of fertilizers and proper rates of applications were of key importance to both crop yields and environmental protection. In this article, the impact of four fertilizers, i.e., inorganic compound fertilizer, organic compound fertilizer, pig manure compost, and peanut cake （peanut oil pressing residue）, on chestnut （Castanea mollissima Blume） growth on a slope in South China, and on the total N and total P concentrations in runoff waters have been investigated during two years of study, with an orthogonal experimental design. Results show that the organic compound fertilizer and peanut cake promote the heights of young chestnut trees compared to the control. In addition, peanut cake increases single-fruit weights and organic compound fertilizer raises single-seed weights. All the fertilizers increased the concentrations of total N and total P in runoff waters, except for organic compound fertilizer, in the first year experiment. The observed mean concentrations of total N varied from 1.6 mg/L to 3.2 mg/L and P from 0.12 mg/L to 0.22 mg/L, which were increased with the amount of fertilizer applications, with no pattern of direct proportion. On the basis of these experiment results, organic compound fertilizer at 2 kg/tree and peanut cake at 1 kg/tree are recommended to maximize chestnut growth and minimize water pollution.
