【Background】fatty acyl reductases(FARs) catalyze the reduction of fatty acyl-coenzyme A or-acyl carrier protein substrates to primary fatty alcohols, which plays essential roles in plant lipid metabolism. However, molecular mechanism of FARs involved in male development is poorly defined. Here, we found that two maize allelic mutants, ms25-6065 and ms25-6057, displayed defective anther development, impaired pollen formation and complete male sterility. 【 Method 】 Cytological observation and microscopy, genetic analysis and map-based cloning, protein alignment and phylogenetic analysis, subcellular localization, microsynteny analysis, prokaryotic expression and activity assay, RNA extraction and expression analysis.【Results】Ms25 mutation affects anther cuticle and Ubisch body formation as well as microspore development. ZmMs25 and its orthologs may arise from an ancient FAR appearing before the differential evolution of monocot and dicot species. Some of maize paralogous FARs may participate in silk and anther development, and their roles in anther development may depend on the normal function of ZmMs25. ZmMs25 encoded a plastid-localized FAR with catalytic activities to multiple acyl-coenzyme A substrates in vitro, and all four conserved residues(G101, G104, Y327 and K331) in two motifs of ZmMs25 protein were critical for its enzyme activity. Moreover, loss of ZmMs25 function had significant effects on the expression patterns of its two paralogous genes and seven cloned lipid metabolic male-sterility genes in maize during anther development. Therefore, ZmMs25 plays key roles in male fertility possibly through participating in multiple lipid metabolic pathways essential for anther and pollen development. 【Conclusion】First, ZmMs25 functions as an active FAR with multiple substrates and all four conserved amino acid residues are indispensable for its catalytic activity.
