Thai pigmented rice bran extracts inhibit production of superoxide, nitric oxide radicals and inducible nitric oxide synthase in cellular models

Objective:To study the inhibitory effect of rice bran extracts of Thai black Kam Muang and red Hawm Dawk Mali Deang on oxidative stress factors including superoxide(O2·-),nitric oxide(NO·),and inducible nitric oxide synthase(iNOS).Methods:Bran extracts(40%ethanol)of Kam Muang and Hawm Dawk Mali Deang were obtained and evaluated for in vitro 2-2′-azino-di-(3-ethylbenzthiazoline sulfonate)(ABTS)and NO·scavenging activity.Their inhibitory effects on cellular O2·-and NO·were measured in phorbol 12-myristate 13-acetate-stimulated neutrophil-like HL-60 cells and lipopolysaccharidestimulated RAW264.7 macrophages,respectively,and their viability was monitored using the MTT assay.The effect on iNOS expression was also assessed by the Western blotting assay.Total contents of phenolics,flavonoids,and subtypes were also determined.Results:Hawm Dawk Mali Deang exhibited about 3.5-fold greater cellular O2·-inhibitory activity than Kam Muang[EC50 values of(23.57±4.54)and(81.98±1.45)μg/mL,respectively]in phorbol 12-myristate 13-acetate-stimulated HL-60 cells.Hawm Dawk Mali Deang exhibited about 2-fold higher in vitro ABTS·+and NO·scavenging activity than Kam Muang,but it exerted cellular NO·inhibitory activity of only about 26%(undetermined EC50 value)in lipopolysaccharide-stimulated RAW264.7 cells.Conversely,Kam Muang exerted potent cellular NO·inhibitory activity[EC50 value:(281.13±59.18)μg/mL]and dose-dependently decreased iNOS levels.No cytotoxicity of both extracts was detected in both cell types.As for corresponding contents,Hawm Dawk Mali Deang contained higher contents of phenolics and flavonoids than Kam Muang.Moreover,Kam Muang and Hawm Dawk Mali Deang had a high content of total anthocyanins[(14.73±0.52)mg C3GE/g of extract]and total proanthocyanidins[(115.13±1.47)mg CE/g of extract],respectively.Conclusions:Based on these data,bran extracts of Thai black Kam Muang and red rice Hawm Dawk Mali Deang can help lower oxidative stress and inflammation attributed partly to O2·-and NO·.

Larvicidal and adulticidal potential of medicinal plant extracts from south India against vectors

Objective:To determine the larvicidal and adulticidal activities of hexane,ethyl acetate and methanol extracts of Momordica charantia（M.charantia）,Moringa oleifera（M.oleifera）,Ocimum gratissimum（O.gratissimum）,Ocimum tenuiflorum（0.tenuiflorum）,Punica granalum（P. granatum） and Tribulus terrestris（T.terrestris） against Culex gelidus（Cx.gelidus） and Culex quinquefasciatus（Cx.quinquefasciatus）.Methods:Bioassay test was carried out by WHO method for determination of larvicidal and adulticidal activity against mosquitoes.Results:All plant extracts showed moderate larvicidal and adulticidal activities,however the effective larval mortality was found in the leaf ethyl acetate and methanol extract of O.gratissimum and bark methanol extract of M.oleifera against Cx.gelidus with LC50 values of 39.***.66.28,and 21.83μg/mL respectively,and methanol extract of 0.gratissimum,0.tenuiflorum and P.granatum against Cx.quinquefasciatus with LC50 values of 38.***.24.90 and 67.20μg/mL,respectively.The adult exposed for 1 h and mortality was recorded at 24 h recovery period.Above 90%- mortality was found in the ethyl acetate and methanol extract of all experimental plants at the concentrations of 500μg/mL.Conclusions:The present results suggest that the medicinal plant extracts provided an excellent potential for controlling Cx.gelidus and Cx.quinquefasciatus.

Anti-hemolytic, antibacterial and anti-cancer activities of methanolic extracts from leaves and stems of Polygonum odoratum

Objective: To investigate anti-hemolytic, antibacterial and anti-cancer activities of leaf and stem extracts from Polygonum odoratum. Methods: Leaves and stems of Polygonum odoratum were extracted using methanol and their anti-hemolytic activity was assessed using 2, 2′-Azobis(2-methylpropionamidine) dihydrochloride which is known to generate free radical damage on cell membranes of red blood cells. This damage, represented by hemolysis, was measured using spectrophotometry. Antibacterial activity was tested by using a broth microdilution method to find minimal inhibitory concentrations against eight bacterial strains. Anti-cancer activity of the extracts was evaluated against a human promyelocytic leukemic cell line(HL-60) by using MTT assay for cell viability and flow cytometry for apoptosis induction and cell cycle analysis. Results: Both leaf and stem extracts have anti-hemolytic activity. The results showed a significantly increased percentage of inhibition in a concentration-dependent manner. Interestingly, the leaf extract showed anti-hemolytic activity to a greater extent than the stem extract. Antibacterial activity of the extracts, as indicated by their minimal inhibitory concentration, using 12.5, 50, 25, 25 μg/mL, was measured against Staphylococcus epidermidis, Enterococcus faecium, Enterococcus faecalis and Staphylococcus aureus. The leaf extracts also exhibited anti-cancer activity, demonstrated by significantly decreased cell viability of human promyelocytic cells(HL-60), with an IC_(50) of(350.00±1.85) μg/mL for 48 h and(38.00±0.92) μg/mL for 72 h. Additionally, HL-60 became apoptotic and accumulated in G_1-phase after 48 hours of treatment. Conclusions: The extracts of Polygonum odoratum exhibit potential antihemolytic activity. They also have antibacterial activity by inhibiting growth of Gram-positive bacteria. Acaricidal activity of different extracts from Syzygium cumini L.Skeels(Pomposia) against Tetranychus urticae Koch

Objective:To investigate the acaricidal activity of different extracts from Syzygium cumini(S.cumini)(Pomposia)againsst Tetranyckus urticae Koch(71 urticae)and the biochemical changes in antioxidants enzymes.Methods:Six extracts of S.cumini(Pomposia)at concentrations of 75,150 and 300μg/mL were used to control T.urticae(Koch).Results:The ethanol extract showed the most efficient acaricidal activity agent against T.urticae(98.5%)followed by hexane extract(94.0%),ether and ethyl acetate extract(90.0%).The LC_(50)values of the promising extract-were 85.0,101.0,102.0 and 98.0μ/mL,respectively.The activities of enzymes including ascorbate peroxidase(APX),peroxidase(POD),superoxide dismutase(SOD)and catalase(CAT)in susceptible mites were increased.The activities of all antioxidant enzymes reach the maximum value in mites at LC_(50)with ethanol and ethyl acetate extracts,respectively.Conclusions:The extract of S.cumini has acaricidal acivity against T.urticae,and the ethanol extract is the most efficient.

Specific degradation of keratin in Xenopus laevis egg extracts undergoing apoptosis

Cytochrome c activates apoptosis specific protease XCPP32 when being added to Xenopus laevis egg extracts, and induces apoptosis in this cell-free system. During apoptosis, cyto-skeleton proteins in egg extracts are degraded. Western blot assay indicates that 42-ku acidic keratin in egg extracts has been degraded by XCPP32. The degradation of 42-ku keratin may be crucial in apoptosis.

Nicotiana ovule extracts induce nuclear reconstitution of demembranated Xenopus sperm in cell-free system

Nicotiana tabaccum ovule extracts induced nuclear reconstitution of demembranated Xenopus leavis sperm in a cell-free system. Demembranated Xenopus sperm began to swell after 15 min of incubation with Nicotiana ovule extracts. Accompanying the process of incubation, Xenopus sperm decondensed and their shapes changed gradually from long and ellipse to round. The completely decondensed chromatin was surrounded with membrane structure, which was a mixture envelope of a double membrane and a single membrane. Nucleosome assembly was verified by means of micrococcal nuclease digestion to re-constituted nuclei and DNA electrophoresis. Nicotiana ovule extracts supplied one more experimental model and system. The new system could promote powerfully the research on mechanisms of cell division and cell cycle regulation.

Objective:To explore the effect of herbal polyphenolics on filariasis in vitro.Methods:Two herbal extracts,methanolic extracts of roots of Vitex negundo Linn.（Nirgundi） and leaves of Aegle marmelos Juss.（Beal） in different concentrations ranging from 40-80 ng/mL were tested for their antifilarial activity either alone or in combination with diethyl carbonate（DEC）（300μg/mL） and/or H2O2（0.5 mM）.Results:Combination of DEC and each extract had significant anti-filarial effect.And fractions of both extracts were not effective as crude herbal extract.Conclusions: Such unique pharmacodynamics reported in this study might provide new drug development stratagem against filariasis.

Dengue fever treatment with Carica papaya leaves extracts

The main objective of the current study is to investigate the potential of Carica papaya leaves extracts against Dengue fever in 45 year old patient bitten by carrier mosquitoes.For the treatment of Dengue fever the extract was prepared in water.25 mL of aqueous extract of C.papaya leaves was administered to patient infected with Dengue fever twice daily i.***.morning and evening for five consecutive days.Before the extract administration the blood samples from patient were analyzed.Platelets count(PLT),White Blood Cells(WBC) and Neutrophils(NEUT) decreased from 176×10~3/μ L,8.10×10~3/μ L,84.0%to 55×10~3/μ L,3.7×10~3/μL and 46.0%.Subsequently,the blood samples were rechecked after the administration of leaves extract.It was observed that the PLT count increased from 55×10~3/μ L to 168×10~3/μ L,WBC from 3.7×10~3/μ L to 7.7×10~3/μ L and NEUT from 46.0%to 78.3%.From the patient feelings and blood reports it showed that Carica papaya leaves aqueous extract exhibited potential activity against Dengue fever.Furthermore,the different parts of this valuable specie can be further used as a strong natural candidate against viral diseases.

Gas chromatography-mass spectrometry analysis of different organic crude extracts from the local medicinal plant of Thymus vulgaris L

Objective:To isolate and analyze the chemical composition in different crude extracts of from the leaves of locally grown of Thymus vulgaris L(T.vulgaris)by gas chromatography-mass spectrometry(GC-MS).Methods:The shade dried leaves powder was extracted with methanol by using Soxhlet extractor.Methanol crude extracts of T.vulgaris and the derived fractions of hexane,chloroform,ethyl acetate and butanol were obtained.Results:Qualitative analyses of various organic crude extracts of T.vulgaris by using GC-MS showed that there were different types of high and low molecular weight compounds.Most of the isolated and identified compounds by GC-MS in the crude extracts are basically biologically important.Further,the T.vulgaris leaf possessed certain characteristics that can be ascribed to cultivation on a domestic plantation.The crude extracts were prepared from the powder leaves of T.vulgaris for respective compounds can be chosen on the basis of above GC-MS analysis.Conclusions:All the major compounds were identified and characterized by spectroscopic method in different organic crude extracts of T.vulgaris are biologically active molecules.Thus the identification of a good number of compounds in various crude extracts of T.vulgaris might have some ecological role.

Antibacterial activity of eight Iranian plant extracts against methicillin and cefixime restistant Staphylococcous aureus strains

Objective:To assess the antibacterial activity of eight Iranian plant extracts against Staphylococcus aureus(S.aureus) strains which were isolated from infected patients.Methods: The studied strains were isolated from urine,stool,blood and wound of infected patients and identified by biochemical tests.In further,the antibacterial activity of 8 ethanolic local plant extracts including Querqus brantii(Q.brantii),Ziziphus spina-christi(Z.spina-christi),Peganum harmala(P.harmala),Oliveira decumbens(O.decumbens),Galium tricornutum(G.tricornutum), Vitex pseudo negundo(Vi.pseudo negundo),Salvia officinalis(S.officinalis),Vaccaria pyramidata (V.pyramidata) were then evaluated using agar disc diffusion method.Results:A total of 9 isolates were isolated and identified as S.aureus.Antibacterial profile of the strains showed that the strains were resistant to methicillin and cefixime.The highest antibacterial activity against the studied strains were belong to Q.brantii,P.harmala,Z.spina-christi and O.decumbens vent extracts with 11-40 mm,15-28 mm,8-26 mm and 10-20 mm of diameters,respectively. Intermediate antibacterial activity was exhibited by G.tricornutum and Vi.pseudo negundo against some of the studied strains with 7-20 mm and 7-13 mm of diameters,respectively. Noteworthy,out of 9 studied strains;6 and 2 of them were resistant to the G.tricornutum and Vi. pseudo negundo extracts,respectively.The S.officinalis and Va.pyramidata,however,showed no antibacterial activity against the studied strains.Conclusions:The Q.brantii,P.harmala,Z. spina-christi and O.decumbens extracts were shown to possess strong antibacterial activity against methicillin and cefixime resistant S.aureus strains and can be considered as the promising natural antibiotics for treating the studied strains.
