Effectiveness of Fuling(Poria) and its extracts against spleen deficiency in rats via tonifying spleen

OBJECTIVE:To observe and explore the effect of Fuling(Poria) in alleviating the spleen deficiency symptom pattern(SDSP).METHODS:We established an animal model of SDS in Sprague-Dawley(SD) rats by treating them with deficiency-inducing factors,including irregular feeding and tail clamping.Mice were administered Fuling(Poria) and its extracts(raw/cooked powder,aqueous/alcohol extract) by gavage once a day for 21 d.The body weight,rectal temperature,and spleen and thymus organ coefficients were calculated.The levels of motilin(MTL),gastrin(GAS),aquaporin 2(AQP2),interleukin 2(IL-2),IL-4,and 5-hydroxytryptamine(5-HT) in the serum and the level of AQP2 in the kidneys were evaluated by enzyme-linked immunosorbent assay.RESULTS:Fuling(Poria) and its extracts did not change the body weight,rectal temperature,and organ coefficients of the spleen and thymus.However,it reduced the levels of MTL and GAS and increased the levels of IL-2 and AQP2.In addition,the levels of IL-4 and 5-HT showed no significant alteration.CONCLUSIONS:These results suggested the crucial function of Fuling(Poria) in SDSP,especially promoting digestive function and water metabolism.

The potential of medicinal plant extracts to control rice blast fungus Magnaporthe oryzae was investigated. We screened 48 extracts prepared from 8 medicinal plant species and identified 20 extracts with ≥ 10% inhibitory activity against M. oryzae. Among them, ethanol extract of Paris polyphylla, ethyl acetate extract of Phellodendron chinense, and n-butanol extract of Menispermum dauricum plants exhibited 100%, 70% and 50% inhibition of M.

Schisandra chinensis,a widely used Chinese herbal medicine,was considered as central nervous system(CNS)drug for years.Both ethanol extracts(EES)and water extracts(WES)of it were applied clinically.Unfortunately,the difference of their efficacy and even effective material foundation of S.chinensis remains obscure.In this study,to explore the active constituents of S.chinensis,we compared pharmacodynamics and chemical profiles in vitrolin vivo of EES/WES for the first time using multiple chemical analysis,pharmacological and data processing approaches.It was proved that there was no significant difference in the anti-depressive effects between WES and EES.However,the contents of most components in vitro and in plasma were higher in EES than those in WES,which was unconvincing for their similar efficacy.Therefore,we further explored components of S.chinensis targeted onto brain and the results showed that 5 lignans were identified with definite absorptivity respectively both in EES and WES caused by the limitation of blood-brain barrier.Moreover,bioinformatic analysis predicted their anti-depressive action.Above all,the systematic strategy screened 5 brain-targeted effective substances of S.chinensis and it was suggested that exploring the components into nidi would promote the studies on herbs effective material basis.

The study was to explore the effect of the extracts of Angelica sinensis(EAs) on lipid oxidation in fish feeds compared with ethoxyquin(EQ) and the effect of dietary EAs on growth performance of carp(Cyprinus carpio var. Jian). Firstly, fish feeds were respectively added with EQ, and ethyl ether extract ethyl acetate extract(EAE), acetone extract, ethanol extract(EE) and aqueous extract(AQE) of Angelica sinensis, except for the control. The results showed that EAs and EQ inhibited lipid oxidation in fish feed(P < 0.05). Of all of the examined EAs, EAE showed the strongest protective effects against lipid oxidation(P < 0.05). Moreover, EAE at high concentrations showed a stronger effect on lipid oxidation compared with EQ(P < 0.05). Then, 7 experimental diets respectively supplemented with 0.0, 1.0, 2.0, 3.0, 4.0, 5.0 and 6.0 g/kg of EAE were fed to the respective treatment groups for 30 d. Four replicates were performed for each treatment group; 20 carp(mean weight: 12.10 ± 0.13 g) were in each replicate. The result indicated that dietary EAE improved the growth performance in carp(P < 0.05). The appropriate con centration of EAE for carp growth was estimated to be 3.643 g/kg diet. Thus, EAE could be used as a natural antioxidant in feeds for Jian carp.

Astragalus membranaceus(Radix Astragali, RA) and Atractylodes macrocephala(Rhizoma Atractylodis Macrocephalae, RAM) are often used to treat gastrointestinal diseases. In the present study, we determined the effects of polysaccharides extracts from these two herbs on IEC-6 cell migration and explored the potential underlying mechanisms. A migration model with IEC-6 cells was induced using a single-edged razor blade along the diameter of cell layers in six-well polystyrene plates. The cells were grown in control--media or media containing spermidine(5 μmol·L^(-1), SPD), alpha-difluoromethylornithine(2.5 mmol·L^(-1), DFMO), 4-Aminopyridine-(40 μ-mol·L^(-1), 4-AP), the polysaccharide extracts of RA or RAM(50, 100, or 200 mg·L^(-1)), DFMO plus SPD, or DFMO plus polysaccharide extracts of RA or RAM for 12 or 24 h. Next, cytosolic free Ca^(2+)([Ca^(2+)]cyt) was measured using laser confocal microscopy, and cellular polyamine content was quantified with HPLC. Kv1.1 mRNA expression was assessed using RT-qPCR and Kv1.1 and RhoA protein expressions were measured with Western blotting analysis. A cell migration assay was carried out using Image-Pro Plus software. In addition, GC-MS was introduced to analyze the monosaccharide composition of both polysaccharide extracts. The resutls showed that treatment with polysaccharide extracts of RA or RAM significantly increased cellular polyamine content, elevated [Ca^(2+)]cyt and accelerated migration of IEC-6 cells, compared with the controls(P extracts not only reversed the inhibitory effects of DFMO on cellular polyamine content and [Ca^(2+)]cyt, but also restored IEC-6 cell migration to control level(P < 0.01 or < 0.05). Kv1.1 mRNA and protein expressions were increased(P < 0.05) after polysaccharide extract treatment in polyamine-deficient IEC-6 cells and RhoA protein expression was increased. Molar ratios of D-ribose, D-arabinose, L-rhamnose, D-mannose, D-glucose, and D-galactose was 1.0:14.1:0.3:19.9:181.3:6.3 in RA and 1.0:4.3:0.1:5.7:2.8:2.2 in RAM. I

Chemical composition of the bark extracts of Coula edulis was investigated to find potential antioxidant,anti-termite and antifungal compounds which can find useful applications in the fields of food,nutraceuticals,cosmetics or agrochemical.Phytochemical screening revealed the presence of several groups of active molecules such as alkaloids,polyphenols,flavonoids,saponins and sterols and/or terpenes in the different extracts.Total phenols,condensed tannins and flavonoids contents corroborated phytochemical screening.Gas chromatographymass spectrometry(GC-MS)analysis revealed compounds in dichloromethane extract different from those obtained with all the other solvents.Hexadecanoic and trans-9-octadecenoic acids,as well as stigmasterol andβ-sitosterol have been identified as the major compounds in the dichloromethane extract.extracts obtained with acetone and toluene/ethanol mixture(2/1,v/v)indicated the presence of few amounts of fatty acids and sugars,catechin in small amount and huge amounts of phenolic acids like gallic and ellagic acids.The radical 2,2-diphenyl-1-picrylhydrazyle(DPPH)and the cationic radical 2,2’-azinobis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS+)were used for evaluation of antioxidant properties of the different extracts.The dichloromethane extracts had a very low antioxidant activity,while acetone and toluene/ethanol extracts presented EC50 values similar to those of catechin and BHT used as reference antioxidant compounds.Effect of the different extracts of the bark of C.edulis on fungal growth inhibition indicated better inhibition of the mycelium growth of brown rot fungi compared to white rot fungi.Low anti-termite activities were recorded with the aqueous extracts,while stronger activities were recorded with dichloromethane,acetone and toluene/ethanol extracts.

The present study investigated the chemical composition of ethylacetate extracts from an endophytic actinomycete Streptomyces sp. A0916 and its host Polygonum cuspidatum. A comparative analysis of the antimicrobial and antioxidant properties of the extracts was also conducted. 32 compounds of P. cuspidatum and 23 compounds of Streptomyces sp. A0916 were isolated and identified by GC/MS. Antimicrobial activities of the extracts were evaluated using eight microbial strains(3 Gram-positive bacteria, 3 Gram-negative bacteria, and 2 fungi). The Streptomyces sp. A0916 extracts showed a wide range of antimicrobial activities and presented greater antimicrobial effectiveness than the P. cuspidatum extracts. The minimum inhibitory concentration(MIC) of Streptomyces sp. A0916 extracts against the ampicillin-resistant strain Enterococcus faecium SIIA843 was 32 μg·m L-1. Furthermore, the extracts had greater antimicrobial effect against Gram-positive bacteria than Gram-negative bacteria. Finally, the antioxidant activity of the Streptomyces sp. A0916 extracts was equal to that of the P. cuspidatum extracts. In conclusion, our results suggest that the endophytic actinomycetes of the medicinal plants are an important source of bioactive substances.

The efficacy of seven plant extracts(neem,fennel,lavender,thyme,pennyroyal,salvia and asafetida) in controlling postharvest of apple(caused by Botrytis cinerea) was evaluated in vitro and in vivo.In vitro,all plant extracts treatments inhibited spore germination.Inhibitory rates of pore germination was 17.41 and 20.83% for neem extract treatment(methanolic and aqueous extracts,respectively) with significant difference compared to control(73.6 and 85.33%) for aqueous and methanol extracts.In the storage conditions,the application of aqueous extract of neem(at concentration of 25%) resulted in 89.11% reduction of disease severity compared with the untreated control.Results of enzymes activity showed the plant extracts can increase the activity of peroxidase,phenylalanine ammonia-lyase,β-1,3-glucanase and polyphenol oxidase in the presence of pathogens,in apple fruits.However,the results of this research revealed that application of neem extracts was more effective than the application of other plant extracts.According to this study,it could be concluded that plant extracts may be useful to control postharvest disease as a safe alternative option to chemical fungicides.

OBJEVTIVE: To investigate the effects of extracts from Chuanwu(Aconitum Carmichaelii) and Banxia(Rhizoma Pinelliae) on the excisional wound healing in a rat's model.METHODS: Rats were performed a surgical lesion with a 2.0 cm resected tissue in the dorsal fascia.Following, animals were randomly divided into model group, YNB group and APE group those were respectively treated with saline, 1 mg/mL of Yunnan Baiyao and 1 mg/mL of Chuanwu(Aconitum Carmichaelii) and Banxia(Rhizoma Pinelliae) extracts. Wound contractions in days 0, 3, 7, 14 and 21 were calculated by an image analyzer. Histological analysis was analyzed using hematoxilin and eosin.Levels of tumor necrosis factor-2(IL-2), IL-4, IL-10, transformαin(TNF-g growα), interleukinth factor-1(TGF-nd basic fibroblast growth factor(bFββ1) a-GF) were determined by real-time quantitative PCR.RESULTS: Compared with that of YNB group and APE gtoup, the skin of rats showed poor re-modeling and re-epithelization characterized by a significant decrease of neovascularization, epithelialization and fibroblast in the model group. In the APE group, levels of TNF-α-and IL-2 were significantly down-regulated and IL4 and IL-10 significantly upregulated in contrasted with that of model group.In addition, levels of TGF-roup were significantly iβ1 and bFGF in the APE gnduced compared with that of model group.CONCLUSION: The results suggest that the extracts from Chuanwu(Aconitum Carmichaelii) and Banxia(Rhizoma Pinelliae) promote wound healing in the rats, which is associated with enhancing antiinflammatory ability and inducing fibroblast formation.
