CD93 and GIPC expression and localization during central nervous system inflammation

CD93 and GAIP-interacting protein, C termius （GIPC） have been shown to interactively alter phagocytic processes of immune cells. CD93 and GIPC expression and localization during cen-tral nervous system inflammation have not yet been reported. In this study, we established a rat model of brain inlfammation by lipopolysaccharide injection to the lateral ventricle. In the brain of rats with inlfammation, western blots showed increased CD93 expression that decreased over time. GIPC expression was unaltered. Immunohistochemistry demonstrated extensive distribution of CD93 expression mainly in cell membranes in the cerebral cortex. After lipopoly-saccharide stimulation, CD93 expression increased and then reduced, with distinct staining in the cytoplasm and nucleus. Double immunolfuorescence staining in cerebral cortex of normal rats showed that CD93 and GIPC widely expressed in resting microglia and neurons. CD93 was mainly expressed in microglial and neuronal cell membranes, while GIPC was expressed in both cell membrane and cytoplasm. In the cerebral cortex at 9 hours after model establishment, CD93-immunoreactive signal diminished in microglial membrane, with cytoplasmic transloca-tion and aggregation detected. GIPC localization was unaltered in neurons and microglia. These results are the ifrst to demonstrate CD93 participation in pathophysiological processes of central nervous system inlfammation.

Few studies have focused on the expression and localization of heat shock proteins70/90(HSP70/90) with normal physiological conditions in yaks(Bos grunniens).The objective of this study was to investigate the expression and localization of HSP70/90 in different tissues and organs of female yaks under the chronic low temperature and ***90 mRNA in this study was cloned by RT-PCR from heart samples,and 16 healthy female yaks were selected for detecting the expression of HSP70/90 in different tissues by qRT-PCR,Western blotting(WB),immunohistochemistry and immunofluorescence from mRNA and protein *** characteristics analysis showed that HSP90 cDNA open reading frame(ORF) code 722 amino *** molecular phylogenetic tree was constructed showed that the HSP90 gene of yak is highly ***70 gene and protein expression level had significant differences in kidney,heart and ***,HSP90 gene and protein expression level had significant differences in heart,liver and ***70/90 was mainly detected in kidney tubules,cardiac muscle cells,hepatocyte,purkinje cell and medulla of ***,HSP70 gene and protein expression level had significant differences in female yaks’ reproductive system(Pexpression level of HSP90 had significant difference(Pexpression in the non-reproductive is higher than reproductive,and expression of HSP70 is higher than HSP90 in all detected ***,our study information would be important as a theory evidence to research some disease and adaptability to the harsh environment in human.
