Creating large EMS populations for functional genomics and breeding in wheat

Wheat germplasm is a fundamental resource for basic research,applied studies,and wheat breeding,which can be enriched normally by several paths,such as collecting natural lines,accumulating breeding lines,and introducing mutagenesis *** methane sulfonate(EMS)is an alkylating agent that can effectively introduce genetic variations in a wide variety of plant *** this study,we created a million-scale EMS population(MEP)that started with the Chinese wheat cultivars‘Luyan 128’,‘Jimai 38’,‘Jimai 44’,and‘Shannong 30’.In the M1 generation,the MEP had numerous phenotypical variations,such as>3,000 chlorophyll-deficient mutants,2,519 compact spikes,and 1,692 male sterile *** were also rare mutations,including 30 independent tillers each with double *** M1 variations of chlorophyll-deficiency and compact spikes were inheritable,appearing in the M2 or M3 *** advance the entire MEP to higher generations,we adopted a single-seed descendent(SSD)*** other seed composites of M2 were used to screen other agronomically important traits,such as the tolerance to herbicide *** MEP is available for collaborative projects,and provides a valuable toolbox for wheat genetics and breeding for sustainable agriculture.

A New Gene Conferring the Glabrous Trait in Cucumber Identified Using MutMap

Tubercles and spines on fruit peel are important commercial traits in cucumber(Cucumis sativus L.). From an ethyl methane sulfonate cucumber mutant library,we discovered a new glabrous mutant that bears no tubercle or spine on fruit peel and fewer and smaller trichomes on the stem and leaves. The new locus is here designated as glabrous2(gl2). Genome sequencing of the mutant and linkage analysis revealed that a non-synonymous mutation in the Csa1G056960 gene rendered the gl2 phenotype. The mutated gene encodes a C-type lectin receptor-like tyrosine protein kinase. This study provides a novel allele for elucidating the genetic basis of wart and trichome development and a new tool for breeding glabrous cucumber varieties.

A parent strain Aspergillus niger LW-1 producingβ-mannanase,preserved in our laboratory,was isolated naturally.A strain N-9 was screend out and further treated with vacuum microwave irradiation and ethylmethanesulfonate(EMS) in this *** results revealed that a mutant strain E-30 producing high- and stable-yieldβ-mannanase was obtained through screened by solid-state cultivation on the basic fermentation medium and several generations of bevel *** enzyme activity was increased by 2.15 times from *** LW-1’ s 17048 U g to E-30’s 36675 U *** propert ies of producing high-yieldβ-mannanase remained stable by mutant strain E-30 after maintained at 4℃for two months.
