The excellent adsorption and rapid separation capabilities of magnetic MOFs make them desirable pretreatment materials for solid *** this study,magnetic ZIF-8(mZIF-8)was synthesized in situ by a one-step method in the aqueous solution of magnetic *** adsorption verified that the maximum adsorption capacity of(011)crystal-exposed rhombic dodecahedral ZIF-8 for chloramphenicol was up to 128.31 mg/g(mZIF-8=67.18 mg/g).Kinetic adsorption revealed that the type of ZIF-8/mZIF-8 adsorption on chloramphenicol belongs to the pseudo-secondary adsorption kinetics of chemical monomolecular *** by FTIR,XPS,and XRD revealed that mZIF-8 interacts with chloramphenicol mainly byπ-πelectron stacking,electrostatic attraction,and hydrogen bonding *** confirmed that chloramphenicol remained antigenically active after adsorption by *** adsorption and separation of chloramphenicol residues in chicken and egg were completed by mZIF-8 within 20 ***-8 can be used directly for elution-free ELISA after the adsorption of *** limits of detection(IC10)of the mZIF-8+ELISA in chicken and eggs were 1.18 ng/mL and 0.64 ng/mL,***-8 is expected to be used as a magnetic solid-phase extraction material for the rapid pretreatment of antibiotic residues in other complex solid matrices.

Canine herpesvirus (CHV-1) causes disease associated with high mortality in infect-ed puppies, which represents large financial losses for dog breeders. Since CHV-1 at the time of the study he had not been reported in Mexico, the main objective of this study was to determine the prevalence of antibodies against CHV-1 in canine kennels in the metropolitan area of Mexico City. A commercial enzyme-linked immuno-sorbent assay (ELISA) was used, and the results were compared to those of a viral neutralization test. The ELISA kit uses the complete viral particle as the antigen. The plaque reduction neutralization test was combined with the immunoperoxidase technique because of the low cytopathic effect of CHV-1. Neutralizing antibodies were also detected in 20 randomly selected samples. The prevalence of CHV-1 with ELISA was 87%. The concordance between ELISA and serum neutralization (SN) was 0.1129, the sensitivity of the ELISA against SN was 1.0 (100%), the positive predic-tive value was 0.39 (39%), and the negative predictive value was 1 (100%). These results show that ELISA is useful for monitoring the dog population for CHV-1;a positive test result requires confirmation with an SN test, and a negative ELISA result indicates a high probability of being SN-negative. The only variables that were sta-tistically associated with CHV-1 prevalence were breed and kennel. A statistically significant relationship between the degree of ELISA and SN titer was obtained, with a confidence level of 95%. None of the clinical presentation factors was statistically significant. These results suggest that most of the canine population studied in Mex-ico is in a herpesvirus latency state.

Seroprevalence of Hepatitis E Virus in Human and Pigs in Pig Farms from Guangdong Province,Southern China

Hepatitis E is caused by hepatitis E virus （HEV）, which has been classified into four genotypes. Genotypes 3 and 4 are regarded as zoonotic pathogens. Accumulating researches indicate that genotype 4 is the main HEV strain circulating in China, and there are high levels of seropositive pigs and human in some provinces of China. In this study, serum samples from pigs and from human occupationally exposed to pigs were obtained from pig farms in Guangdong Province, in subtropical southern China, in order to investigate for the first time the prevalence of anti-HEV immunoglobulin G （IgG） in the region. Antibodies against HEV were detected by enzyme-linked immunosorbent assay （ELISA） using a commercially marketed kit. The results showed that high numbers of pigs （74/94; 78.7%） and human （50/94; 53.2%） from three pig farms in Guangdong Province were positive for anti-HEV IgG. The correlation coefficient relating the prevalence in pigs and human on different farms was 0.920. The seropositive rate in males （human） was 48.8% （20/41） and that in females was 47.7% （9/19）, which showed no statistically significant difference. These data indicated that there was a high prevalence of anti-HEV antibodies in pigs and in human with occupational exposure to pigs. The risk of infection with HEV in both human and pigs in Guangdong Province appeared to be age-dependent, to a certain extent. This study provided basic data for further researches on HEV and was a reminder that more attention should be paid to HEV infection both in pigs and workers on pig farms in the study region.

Investigation on Avian Leukosis Virus Subgroup A in Layers in Guizhou Province of China

A total of 273 serum samples were collected from 10 layer breeding farms in Guizhou Province and detected by enzyme-linked immunosorbent assay. The positive rate of antibodies against avian leukosis virus reached 14.64%. Then the samples from cases of hemangioma in one of the 10 breeding farms were detected by polymerase chain reaction, and the diagnosis indicated that the hemangioma was caused by avian leukosis virus subgroup A.

Diagnostic role of soluble triggering receptor expressed on myeloid cell-1 in patients with sepsis

BACKGROUND：Biomarkers may be helpful in risk stratification and prediction of mortality in septic patients. This study aimed to investigate the diagnostic role of soluble triggering receptor expressed on myeloid celI-I（sTREM-1）, procalcitonin （PCT）, C-reactive protein （CRP） and other inflammatory markers in patients with sepsis. METHODS：A total of 56 patients with systemic inflammation response syndrome （SIRS） who had been admitted to the ICU department of the Second Hospital of Tianjin Medical University between May 2009 and July 2010 were enrolled. They were divided into a sepsis group （n=32） and a SIRS group （n=24）. Twenty-five non-SIRS patients served as controls. The sepsis group was sub-divided into a survival group and a death group according to 28-day prognosis. The values of sTREM-1, PCT, CRP, white blood cell （WBC）, and neutrophil count percentage （N） were measured. Acute physiology and chronic health evaluation II （APACHE II） score were determined within 24 hours. The correlation between sTREM-1 and APACHE II score was analyzed. Quantitative data were analyzed by the F test or the KruskaI-Wallis test. RESULTS： The plasma level of sTREM-1 in the sepsis group was significantly higher than that in the SIRS group and control group. The plasma level of sTREM-1 in the non-survival group was significantly higher than that in the survival group. In the sepsis group, the plasma sTREM-1 level was positively correlated with APACHE II score （rs=0.426, P= 0.032）. The area under the ROC curve of sTREM-1 was 0.935, larger than that of PCT and CRP. CONCLUSION：Plasma sTREM-1 is useful in the diagnosis of sepsis at early stage. The increased level of sTREM-1 during the first 24 hours may be correlated with poor outcome of patients with sepsis.

Anti-enrofloxacin Antibody Production by Using Enrofloxacin-screened HSA as an Immunogen

A two-step zero-length cross-linking procedure using active esters was successfully adopted for conjugating en-rofloxacin (EF) to human serum albumin (HSA). The derived conjugate was characterized by UV spectrum and then usedfor immunization of BALB/C mice. In enzyme-linked immunosorbent assay (ELISA) and competitive inhibition ELISA ex-periments, the derived antiserum exhibited high antibody titer (greater than 1: 250 000) as well as varied cross-reactivity(from 97.8% to 161.7%) to three analogs of EF belonging to fluoroquinolones family. But over the concentration rangestudied, no significant cross-reactivity was observed to other group of antibiotics (chloramphenicol, oxytetracycline, sul-phamethoxazole and nysfungin). It was confirmed that the synthesized immunogen was highly antigenic and elicited specificantibody responses in BALB/C mice against EF.

Variations of Reproductive Hormones of Female Swamp Buffalo from Guangxi Region in Off Season for Estrus

[Objective] The aim of this paper is to find the change law of reproductive hormones of female swamp buffalo in off season for estrus and to investigate the relationship between various hormones. [ Method] The concentrations of follicle stimulating hormone （FSH）, luteinizing hor- mone （ LH）, inhibin （ INH）, estradiol （ E2 ） and progesterone （ P4 ） were determined by the enzyme-linked immunosorbent assay （ELISA） in off season for estrue. The changes of follicles in the estrous cycle were examined using B-mode ultrasonography. [ Result] In off season for estrus of female swamp buffalo from Guangxi region, the blood concentration of E2 and P4 varied fluctuantly. The concentration of E2 had two peaks, respec- tively 3 d before ovulation and after 13 d post ovulation. The concentration of P4 increased slowly from the first day after ovulation, increased rapidly from 5 to 13 d, peaked on Day 16 and decreased to the baseline 3 d before ovulation. The concentrations of FSH, LH and INH roached a peak re- spectively after 10 d post ovulation, 2 d before ovulation, and after 4 d post ovulation. The changes of the hormones had correlations. The concen- trations of FSH, LH and INH had highly significant correlations with each other, while the concentration of INH had highly significant correlations with that of E2 or P4- The concentration of E2 also had a highly significant correlation with that of P4- [ Conclusionl The study can guide the breeding of swamp buffalo from Guanqxi reqion and also help to improve their roproduction performance.

Dynamic change of serum protein S100b and its clinical significance in patients with traumatic brain injury

Objective: To analyze the dynamic change of serum protein S100b in patients with traumatic brain injury and its clinical value in assessing brain damage. Methods: According to Glasgow coma scale (GCS), 102 cases of traumatic brain injury were divided into mild brain injury group (GCS≥13, n=31, Group A), moderate brain injury group (8enzyme-linked immunosorbent assay (ELISA) in blood samples taken on admission, 12 h, 24 h, 48 h, 72 h and 7 days after traumatic brain injury. Results: The severe brain injury group showed significantly higher concentration of serum S100b, with earlier increase and longer duration, than the mild and moderate brain injury groups. The patients with higher S100b exhibited lower GCS scores and poor clinical prognosis. The increase in S100b could emerge before clinical image evidence indicated so. Conclusions: Serum S100b can be used as a sensitive index for assessment and prediction of traumatic brain injury severity and prognosis.

Characterization of Monoclonal Antibody to L-Homocysteic Acid and Its Immunohistochemistry in Rat Hippocampus

L-homocysteic acid (HCA) and other amino acids were conjugated to rat brain material (extracted rat brain protein) with glutaraldehyde to form HCA- and amino acids-brain material conjugates. The specificity of monoclonal antibody (McAb) was tested on serial dilution test and absorption test on enzyme-linked immunosorbent assay (ELISA) using these conjugates as antigens instead of amino acids-BSA (bovine serum albumin) conjugates used previously. The characterized McAb was applied for immunohistochemical staining using PAP (peroxidase antiperoxidase) technique in combination with silver enhancement of diamino-benzene (DAB) products. The results indicated that McAb to L-HCA reacted with L-HCA-brain material conjugates, but not with other amino acids-brain material conjugates so far tested. McAb absorbed with L-HCA-brain material abolished or decreased immunoreactivity of L-HCA-brain material with McAb. The antibody selectively stained subpopulation of cells and processes in the hippocampus fixed with glutaradehyde. Absorption of McAb with L-HCA-brain material abolished immunohistochemical staining. These results suggested that McAb was specific for L-HCA-brain materials and could be used for imunohistocytochemistry. This would provide a new tool for immunohistochemical visualization and localization of L-HCA in the nervous system.

Serum Vascular Endothelial Growth Factor-C and Vascular Endothelial Growth Factor Level in Patients with Colorectal Carcinoma and Clinical Significance

Circulating vascular endothelial growth factor-C （VEGF-C） and vascular endothelial growth factor （VEGF） levels in patients with colorectal carcinoma were determined in order to assess their clinical significance as a diagnostic tool for monitoring lymph node metastasis. In 66 patients with colorectal carcinoma and 30 healthy controls, circulating VEGF-C and VEGF levels were assessed by using enzyme-linked immunosorbent assay （ELISA）. Serum VEGF-C and VEGF levels were higher in patients with colorectal carcinoma than in healthy controls. Patients with lymph node metastasis had higher serum VEGF-C and VEGF levels than those without lymph node metastasis. The levels of VEGF-C and VEGF were higher in the invasion group than in the non-invasion group. Serum VEGF-C levels reached a sensitivity of 81% and a specificity of 76 % with a cutoff value of 1438.0 pg/mL, whereas VEGF levels reached 72 % sensitivity and 74 % specificity at 240.2 pg/ mL. If 66 patients were divided into 4 groups according to the combined determination of VEGF-C and VEGF levels, the positive predictive value was 85.3 %, the negative predictive value was 94.6 %, and accuracy was 93.7 %. It was suggested that circulating VEGF-C levels might provide additional information for distinguishing the absence from presence of lymph node metastasis in patients with colorectal carcinoma. The combined determination of VEGF-C and VEGF levels could be used as an important index for preoperatively clinical stage of colorectal carcinoma.
